PCR 방법으로 병원성 유전자를 결과, flaA (87%), cadF (100%), racR (100%), dnaJ (100%), pldA (96%), ciaB (78%), virB11 (13%), cdtA (96%), cdtB (96%), cdtC (91%), wlaN (22%) 유전자 보유함을 확인할 수 있었다. 이 연구를 통해서 오리유래의 Campylobacter 사람에 대해 병원성을 가질 수 있을 것으로 예상된다. 오리유래 Campylobacter의 분자생물학적 특성을 구명하고자 multilocus sequence typing (MLST) 분석하였다. 그 결과, 오리농장에서 분리된 균은 총 28개의 sequence types (STs)으로 동정되었고, 오리고기에서는 25개의STs로 구분되었다. 오리농장과 오리고기에서 유래된 Campylobacter 중 12개의 STs를 공유하는 것으로 분석되었다. 절반이상이 사람에서 분리된 보고(51.1%)가 있었으며, 14 STs (34.0%)가 높은 빈도로 분석되었다. Pulsed-field gel electrophoresis (PFGE)로 분석한 결과, 동일한 STs에서 오리농장 유래와 오리고기 유래 Campylobacter균이 동일한 클론으로 확인되었다. 이러한 결과를 통해서, 오리농장에서 분리하는 Campylobacter가 오리고기로 오염된 것으로 사료된다. 이 연구는 최근 국내 오리농장과 오리고기에서 Campylobacter의 오염현황을 조사하였으며, 감염이 확인된 Campylobacter의 특성을 확인하였다는데 의의가 있으며 결과는 방역활동의 기초자료로 활용될 수 있을 것으로 판단된다. 또한 오리가 식중독 감염증의 매개체로 오리의 역할을 확인함으로써 예방대책 수립에 중요하게 활용될 것으로 기대된다.

It has been known for a long time that chicken is the main sources of human Campylobacter infections. Since chickens are considered the major reservoir of this food-borne pathogen, the prevalence along with the antimicrobial resistance of Campylobacter spp. in operations is a matter of concern. However, recent studies have suggested that duck is potential emerging sources for human infections. Duck may contribute to human infections through several transmission routes such as direct contact and ingestion of contaminated food. Campylobacter usually colonizes in duck without showing clear symptoms. Some studies have reported that 80% of flocks and 40–60% of individuals may shed Campylobacter. However, the direct and indirect contributions of duck in spreading of human infections are still under investigation whether in the farm level or the retail level. In the first part, the prevalence and antimicrobial resistance of Campylobacter jejuni and Campylobacter coli from domestic duck in Korea was determined. Between May and September 2012, 58 duck farms were analyzed and 96.6% were positive for Campylobacter. Except two farms in Chonbuk and Chonnam, all farms from Gyonggi, Chungbuk and Chungnam were positive for Campylobacter. Among the isolates, 80.4% were C. jejuni, 17.9% were C. coli, and one was unidentified by PCR. In C. jejuni isolates, 86.7%, 11.1%, and 22.2% were resistant to ciprofloxacin, erythromycin, and azithromycin, respectively. All 10 C. coli strains were resistant to ampicillin, 80.0% and 30.0% were resistant to ciprofloxacin and azithromycin. The results indicated that 92.7% of the strains were resistant to at least two antimicrobial agents, and the most frequent multi-drug resistance patterns were resistant to ampicillin, ciprofloxacin, nalidixic acid, and tetracycline at the same time. One C. jejuni and one C. coli showed resistant to all 8 antimicrobial agents. In the second part, we investigated and determined the prevalence and antimicrobial resistance of Campylobacter isolates from retail chicken and duck meat in Chonbuk province. From our results showed that retail duck meat with the isolation rate of 96.2% was significantly higher (p<0.05) than chicken of 58.8%. The Campylobacter isolation rates ranged from 83.8% to 100.0% among the traditional stores, where the online store was significantly (p<0.05) lower than the traditional stores with the isolation rate of 50.0%. All of the C. jejuni and C. coli isolates from poultry meat were resistant to at least an antimicrobial agent. Resistance to nalidixic acid was the most common (93.2%), followed by ciprofloxacin (87.8%) and tetracycline (85.1%) in all the duck and chicken isolates. In the third part, a total of 23 Campylobacter isolates from Korean ducks were tested for the presence of virulence-associated genes by PCR. The presence of flaA (87.0%), cadF (100%), racR (100%), dnaJ (100%), pldA (95.7%), ciaB (78.3%), virB11 (13.0%), cdtA (95.7%), cdtB (95.7%), cdtC (91.3%), and wlaN (21.7%) genes were detected in Campylobacter by PCR. Adhesion, invasion and survival of Campylobacter isolates were determined in a HeLa cell culture model. All tested duck-associated Campylobacter were capable of adhesion to the HeLa cell, could invasion and persist with variable efficiency. These results demonstrated that these putative pathogenic determinants were widespread among Campylobacter isolates from ducks. And the potential source of human Campylobacter infection by its adhesion, invasion and persist in a cell culture model. Our studies call for further investigation for the elimination of Campylobacter infection in industrial duck production and in industrial food chains. The last part determined the molecular epidemiological Campylobacter strains from ducks. Multilocus sequence typing (MLST) was used to genotype Campylobacter from farmed duck and retail duck. A total of 28 sequence types (STs) from duck farm and 25 STs from retail duck meat; 12 STs of Campylobacter strains were overlapped between duck farm and meat. Most of the STs (51%) had been found in human and 14 STs (34%) only found in human and duck. Pulsed-field gel electrophoresis (PFGE) was used to clonality among Campylobacter strains from farm and meat with the same STs. The hmogeneous groups were emerged between the duck farm and retail meat showed Campylobacter in retail meat transmitted via the animal origin. Our study fills the gap of prevalence, distribution, genotypic diversity and molecular epidemiology of Campylobacter strains from farmed duck as well as retail duck meat that exists particularly in Korea. The high isolation rates of Campylobacter species reported here may have implications for food safety, public health and environmental contamination. The results from our study would enhance pre-harvest efforts to establish the effective control and preventive measures. Further, the pathogenesis studies of duck-associated Campylobacter would provide a better understanding of the pathogenic mechanisms of these pathogens and may identify novel targets to develop control strategies.
