염증성 장질환은 특정 원인이 없이 반복적으로 발병되는 만성 염증성 질환으로 궤양성 대장염과 크론병이 이에 포함되며, 주로 대식세포 활성화에 의한 유도에 의한 것으로 보고되어 있다. 본 연구는 대식세포와 염증성 장질환 동물모델을 이용하여 항염증 효과가 있는 유산균을 선발하고자 실시하였다. 본 연구에서는 유산균의 항염증을 알아보기 위해서, in vitro에서 세포독성실험, nitric oxide(NO), inducible nitric oxide synthase(iNOS), cyclooxygenase 2(COX-2), tumor necrosis factor-alpha(TNF-α)와 interleukin 6(IL-6)를 측정하였고, in vivo 실험으로 dextran sulfate sodium(DSS)으로 유도한 염증성 장질환 모델에서 체중과 장 길이 변화, 장의 조직학적 검사, 장 조직의 iNOS, COX-2, TNF-α와 IL-6를 측정하였다. 1차적으로 23개 유산균을 처리하여 양성 대조군에 비해 NO를 20% 이상 억제한 8개 균주 45, 54, 55, 64, 106, 224, 273, 301와 균주의 음성대조군인 364를 선발하였다. Weissella viridescens인 224를 제외하고는 모두 Lactobacillus 균주이였다. 9개의 균주로부터 추출한 peptidoglycan을 가지고 세포 독성실험을 실시하여 세포에 독성이 없음을 확인하고 세포 실험을 진행하였다. 54, 55, 64, 106, 301의 PG에서 NO의 억제능이 높았고 iNOS와 COX-2 단백질에서는 54, 55, 64, 106, 301의 PG가 억제를 많이 하였다. ELISA를 통한 pro-inflammatory cytokine인 TNF-α와 IL-6을 측정한 결과, 54, 55, 64, 106, 224, 301이 항염증 효과를 가지는 것 확인할 수 있었다. 항염증 효과가 좋은 54, 55, 64, 301과 균주의 음성대조군인 364를 가지고 동물 실험을 진행하였다. 54, 55, 64, 364는 Lactobacillus plantarum 균주이고 301은 Lactobacillus salivarius 균주이다. 55를 처리한 군에서 DSS를 단독 처리한 군에 비해 유의적으로 체중이 증가하였고, 54, 55, 64와 301 군에서 장의 길이의 단축을 억제하였다. 장의 조직학적 분석에서는 양성 대조군에 비하여 54과 64군에서 점막손상이 적은 것으로 판단되었다. 장 조직의 iNOS의 발현은 55와 64군에서 유의적으로 감소하였고 COX-2는 55, 64과 301군에서 유의적으로 감소하였으며, TNF-α와 IL-6 cytokine에서는 55와 64군에서 양성 대조군에 비해 억제를 많이 하는 것을 확인하였다. 이상의 결과로부터 선발된 유산균주는 Lactobacillus는 대식세포와 염증성 장질환 동물모델에서 항염증 효과를 가지는 것으로 확인되었으며, 염증성 장질환의 예방 및 치료가 가능할 것으로 판단되므로 이에 대한 추가적인 연구가 필요할 것으로 생각된다.

The purpose of this study was to screen lactic acid bacteria (LAB) strains for the anti-inflammatory activities using RAW 264.7 cells and dextran sulfate sodium (DSS)-induced colitis. For in vitro, the anti-inflammatory activities of LAB strains was investigated by measuring the inhibitory activity against production of nitric oxide (NO), related enzyme activities such as, inducible nitric oxide synthase (iNOS) and cyclooxygenase 2 (COX-2), and key cytokine levels, such as tumor necrosis factor-alpha (TNF-α) and interleukin 6 (IL-6) in RAW 264.7 cells. For in vivo assay, DSS-induced colitis mouse model was used for the investigation of the body weight, colon length and the morphological change of colonic mucosa. In addition, important protein level in their colon tissue, including iNOS, COX-2, TNF-α and IL-6, were measured by western blot and ELISA. Among twenty three isolated LAB strains, eight strains (45, 54, 55, 64,106, 224, 273, and 301) were screened based on the inhibitory activity against NO production (more than 20% inhibition compared with the LPS control) and the strain 364 was selected as a negative control. All screened strains were belonging to the genus Lactobacillus with the exception of strain 224, which was Weissella viridescens strain. Peptidoglycan (PG) fractions extracted from nine LAB strains induced negligible cell toxicity up to the addition level of 1,000 μg/mL. PG fractions of the strains (54, 55, 64, 106, and 301) exhibited the inhibitory activity against NO production and significant inhibition of iNOS and COX-2 proteins. Pro-inflammatory cytokines, TNF-α and IL-6, were remarkably down-regulated at protein level by PG fractions from strains 54, 55, 64, 106, 224, and 301. The animal experiment was conducted with four candidate strains (54, 55, 64, and 301) and strain 364 as a negative control. Significant body weight increase was observed in strain 55 fed group and the shortening of intestinal length was suppressed in strain 54, 55, 64, and 301 fed groups. In mucosal histological analysis, strain 54 and 64 fed groups showed less mucosal damage. The iNOS level was remarkably down-regulated by strains 55 and 64 and COX-2 was significantly decreased by strains 55, 64 and 301 group. And pro-inflammatory cytokines, TNF-α and IL-6 were inhibited by strains 55 and 64 than positive control group. According to the results observed in this study, selected Lactobacillus strains have been shown to have some anti-inflammatroy activities in RAW 264.7 cells and dextran sulfate sodium (DSS)-induced colitis model. The LAB strains screened from this study could be used as candidate probiotic strains for the prevention and treatment IBD, however, further studies will be needed.
