Considering this instability, the aim of the current study was to develop a film-coated tablet formulation, which is advantageous for production, by minimizing the exposure to light and moisture with a direct compression manufacturing process. After selecting the appropriate formulation and excipients, rosuvastatin calcium was produced by a manufacturing process involving a combination of granules suited to direct compression, and the drug was evaluated. Suitability of the manufacturing process was confirmed by checking for uniformity and deviations in the pill. To ensure drug equivalency, ideal elution conditions were ascertained. First, the effect of punch pressure on elution was investigated. When the drug pill became lighter with raised punch pressure, a short crumbling time of less than 30 seconds was obtained due to the characteristics of the excipients, and there was no considerable impact on elution. When the proportion of the disintegrating agent was adjusted to check the effect on elution, smaller proportions of disintegrating agent led to lower initial elution speeds, but had little impact on the final elution rate. In experiments verifying the effect of the coating on elution, almost no difference in initial elution speed for the test drug was observed, but there was a significant difference for the control drug. With a suitable combination of disintegrating agent and coating agent, an elution profile of the test drug was obtained, which was almost identical to the control drug.
