Ethanol 추출물은 DPPH 라디칼 소거능, α-glucosidase 저해활성 및 tyrosinase 저해활성이 있는 것으로 나타났다. 또한, ethyl acetate fraction은 DPPH 라디칼 소거능 측정에서 IC50 52.3 μg/ml, butanol fraction은 α-glucosidase와 tyrosinase에 대한 저해능 측정에서 각각 IC50 42.0 μg/ml, 3.1 mg/ml로 측정되어 높은 활성이 있는 것으로 확인되었다. LC-MS/MS를 이용한 분석 결과를 바탕으로 다변량 통계분석을 진행한 결과, (±)-catechin, kaempferol hexoside, apigenin-7-O-glucoside, syringetin 3-O-glucoside, abscisic acid, procyanidin dimer, phloretin diglucoside가 검출되었다. 특히, 각 fraction에서의 abundance와 OPLS-DA를 통하여 얻은 S-plot을 토대로 syringetin 3-O-glucoside와 phloretin diglucoside를 ethyl acetate fraction과 butanol fraction에서 나타난 활성의 후보물질로 하였다. 그 중에서도 phloretin diglucoside의 aglycone 물질인 phloretin으로 α-glucosidase와 tyrosinase에 대한 저해능을 측정하였다. Torreya nucifera is commonly found in southern Korea and Jeju. Its seed is edible and has been used as a parasiticide or for improving constipation and hemorrhoids. Recently, a lot of researches about high anti-oxidative effect and physiological activities of the seed have been conducted, but there has been no reports on its shell. In this study, physiological activity of ethanolic extract from Torreya nucifera seed’s shell was determined and active compounds were screened using LC-MS/MS to investigate availability as health beneficial material. To extract functional compounds from the shell, ethanolic extraction and fractionation with ethyl acetate and butanol were conducted. The colorimetric methods using DPPH radical, α-glucosidase and tyrosinase were performed to evaluate physiological activities . Ethanolic extract had DPPH radical scavenging effect, α-glucosidase inhibitory activity and tyrosinase inhibitory activity. Ethyl acetate fraction (52.3 μg/ml) showed higher DPPH radical scavenging effect compared to butanol fraction (98.3 μg/ml), when the activities were calculated as IC50. Especially, the inhibitory activity of ethanolic extract and its solvent fractions against α-glucosidase and tyrosinase were higher than positive controls, acarbose and arbutin. In determination of α-glucosidase and tyrosinase inhibitory activity, butanol fraction (42.0 μg/ml, 3.1 mg/ml) had lower IC50 value than ethyl acetate fraction (177.1 μg/ml, 4.7 mg/ml). To screening of physiologically active metabolite in the shell extract, LC-MS/MS data was analyzed using multivariate statistical analysis such as PCA (principal component analysis), PLS-DA (partial least squared discriminant analysis) and OPLS-DA (orthogonal partial least squared discriminant analysis). As a result, (±)-catechin, kaempferol hexoside, apigenin-7-O-glucoside, syringetin 3-O-glucoside, abscisic acid, procyanidin dimer and phloretin diglucoside were detected. Especially, syringetin 3-O-glucoside and phloretin diglucoside were determined as key compounds on the basis of OPLS-DA (S-plot) and their abundances in ethyl acetate and butanol fraction, respectively. Phloretin showed strong α-glucosidase inhibition than acarbose, while similar tyrosinase inhibition with arbutin, which supported phloretin diglucoside as a putative candidate for the activities. This is the first study demonstrating the physiological activity and unearthing its major compound, phloretin diglucoside in Torreya nucifera seed’s shell.
