Western blot과 면역형광염색법을 통해 단백질의 발현양식을 살펴보았다. 금속의존성 단백질 분해효소 모티프의 결실돌연변이인 HEWAH (Hmut) 또는 LRWGVFDEYNED (Bmut)에서는 rbCLCA1의 가공이 영향을 받아 ?130kDa과 ?50kDa 부근에서 Western blot을 통해 GFP 항체로 관찰이 되었던 산물이 오직 ?130kDa 에서만 나타나는 것을 확인하였다. pcDNA3.1+/c-myc-rbCLCA1 에 모티프의 특정 부위를 다음과 같이 점돌연변이를 일으킨 후 다음 실험을 진행하였다 (H155A, E156Q, H159A, D166A, E167A, E170A, D171A). 세포막에서의 rbCLCA1 국재화를 확인하기 위해 면역세포화학적 방법을 통해 형광 이미징 장비를 이용하여 실험을 한 결과, H155A, E156Q, H159A, D166A, E167A, E170A, 그리고 D171A 점 돌연변이는 HEK293T 세포 내에서 막 국재성을 보인 반면, H159A 와 E167A 돌연변이는 permeabilized 또는 non-permeabilized 조건 둘 다에서 세포질 내에 국재하는 것을 확인할 수 있었다. 본 결과는 세포 표면 비오틴화을 이용하여 재확인 하였다. 흥미롭게도, rbCLCA1-GFP 는 돌연변이의 여부에 상관없이 세포질에 국재하는 것을 확인할 수 있었다. 금속의존성 단백질 분해효소 모티프 중 H159와 E167 부분이 rbCLCA1의 세포국재화에 가장 큰 역할을 차지하는 것을 확인할 수 있었으며, H155부터 E167까지의 부분은 rbCLCA1의 가공에도 영향을 미치는 것으로 확인할 수 있었다. 그 이후 rbCLCA1의 가공에 있어서 예상되는 촉매부위의 생화학적 연구를 위해 다음 연구를 진행하였다. 첫 번째 연구에서 사용한 점 돌연변이체들 중, H155A, E156Q, H159A를 선택하여 rbCLCA1과 함께 칼슘, 아연, 마그네슘 등의 2가 이온이 세포 내 가공 단계에서 미치는 영향을 실험하였다. 이를 위해, phosphate buffered saline (PBS)으로 각 construct들이 과발현된 HEK293T 세포를 균일하게 나눈 후, 각 이온들을 각각 또는 적당한 조합으로 첨가하였다. 또는, 산도의 변화에 따라 rbCLCA1이 세포막에서 떨어져 나올 수 있는지를 확인하기 위해 산성, 중성, 염기성 조건의 pH를 이용하여 pGFPN1-rbCLCA1에 미치는 영향에 대해 확인하였다. 마지막으로, co-transfection 방법을 실시하여 rbCLCA1의 자가분해능력에 대해 알아보았다. 그 결과, rbCLCA1, H155A 와 H156Q를 HEK293T 세포에서 과 발현 시킨 세포 lysate에 아연을 첨가하였지만 예상했던 단백질 분해산물이 c-myc 항체를 이용한 Western blot에서는 확인할 수 없었다. 흥미롭게도, pEGFPN1-rbCLCA1과 그 결실돌연변이 일부인 Hmut 의 과 발현산물을 이용하여 같은 절차를 밟았을 때는 분해산물로 추정되는 30kDa과 50kDa 사이의 단백질의 발현을 GFP 항체를 통해서 확인할 수 있었던 반면, Bmut에서는 확인할 수 없었다. 이를 통해 금속의존성 단백질 분해효소 모티프 중 HEXXH 부분이 rbCLCA1의 분해에 큰 영향을 미치지 않고, 몇 잔기 뒤로의 E 또는 D 가 영향을 미치게 되는 것으로 추정할 수 있다. 또한, 이를 통해 rbCLCA1의 분해능은 금속의존성 단백질 분해효소 모티프에 의존적이나, 자가분해능력이 있지는 않았다. 또한, pEGFPN1-rbCLCA1은 산성과 염기성 pH 조건에서 세포막으로부터 분리되는 것을 확인할 수 있었다. 이를 통해, 외부자극에 의해 다른 신호전달과정에 참여할 수 있는 가능성을 제시한다. 따라서, rbCLCA1의 분해는 아연 이온에 의존적일 수 있다는 결론을 내릴 수 있었으며, 산도에도 의존적인데, 특히 산성 조건일 때 아연 이온을 처리했을 때와 비슷한 것으로 보아, 다른 세포환경의 영향도 고려해 볼 필요가 있었다. 다른 한 편으로는 CLCA 유전자군을 다양화하기 위한 노력으로, 아프리카 발톱 개구리에서 유래한 신규 CLCA 유전자를 성공적으로 클로닝 하였다. National Center for Biotechnology Information (NCBI)의 Basic Local Alignment Search Tool (BLAST) 를 이용하여 스크리닝을 한 후, expressed sequence tags (EST) 서열을 얻었다. 이후, 전체 RNA를 이용하여 역전사중합효소 연쇄반응을 이용하여 제작한 1st stranded cDNA를 제작하여 5말단 또는 3말단 Rapid Amplification of cDNA Ends (RACE) 법을 이용하여 4회에 걸쳐 전체 cDNA 서열을 얻어내었다. 각 조직의 발현양상은 역전사 중합효소 연쇄반응 또는 실시간 중합효소 연쇄반응을 통해 확인하였다. 그 결과, 아프리카 발톱 개구리의 RNA 중 CLCA 유전자군으로 예상되는 2,958-bp 의 943 아미노산을 암호화하는 것을 확인할 수 있었고, 발현 양상이 다음과 같이 나타나는 것을 확인할 수 있었다; 뇌>대장>소장>폐>신장>비장>난자>간장>심장. 이를 통해 이미 발표된 개구리의 유전자 발현 분석을 하고, 이를 통해서 기능 연구의 토대를 마련할 수 있을 것으로 사료된다. 최종적으로, 본 연구를 통해 랫드의 CLCA1과 아프리카 발톱 개구리 유래의 CLCA 유전자의 특성을 가늠하여 여러 특정 도메인 중 metalloprotease 모티프를 확인하였다. 특히, rbCLCA1에서는 단백질 가공 중의 세포막 수송 메커니즘과 아연 의존적인 본 모티프의 역할을 확인하였으며, 그 중 159번째 히스티딘과 167번째 글루탐산이 세포막 국재화에 중요한 역할을 담당하고 있는 것으로 생각된다. 또한, rbCLCA1 그 자체는 세포막으로 향하는 막관통단백질이 아니라, 세포표면까지 이동을 하며, 외부자극에 의해 분리가 가능한 단백질이기 때문에, 기타 세포신호 전달에 작용을 한다고 볼 수 있다. 그와 동시에 본 연구에서는 아프리카 발톱 개구리에서 새로이 CLCA 클로닝을 성공하여 기초적인 분석을 통해 CLCA군에 본 유전자를 추가하였다. 따라서, CLCA 유전자군의 다양화에 기여하여, 향후 본 유전자의 기본적인 구조와 다양성에 대한 연구방향을 제시하였다.

Rat brain (rb) calcium-activated chloride channel accessory (CLCA) 1 is a member of the CLCA family cloned from rat brain tissues, which regulates calcium-activated chloride channel (CaCC) current. Researches on CLCAs have been studied for their relationship with diseases such as asthma and cystic fibrosis (CF) and their roles in cell cycle control or cancer. Most studies on CLCAs are focused on their ion channel current production in an electrophysiological method, but their basic biochemical functions have not been studied. Several studies showed that metalloprotease motif could be a candidate to characterize CLCA functions. Since there are diverse in CLCA family members, the relationship between metalloprotease motif and rbCLCA1 has not been studied. In order to elucidate rbCLCA1 processing and localization in intracellular level and to characterize HEXXH-E/D?specified metalloprotease motif, rbCLCA1 was overexpressed in human embryonic kidney 293T (HEK293T) cells. rbCLCA1 was inserted into either pEGFPN1 having C-terminal GFP or pcDNA3.1+/c-myc which has c-myc sequence adjacent to N-terminal signal sequence. Protein expression pattern was observed by both Western blot and immunocytochemistry. Western blot analysis with GFP antibody showed that rbCLCA1 processing was affected by deletion of metalloprotease motif which is referred to as ΔHEWAH (Hmut) and ΔLRWGVFDEYNED (Bmut) respectively, producing only ?130kDa instead of yielding ?130kDa and ?50kDa cleavage products. Metalloprotease-specific mutation was introduced by site-directed mutagenesis of H155A, E156Q, H159A, D166A, E167A, E170A, and D171A on pcDNA3.1+/c-myc-rbCLCA1. Membrane localization of rbCLCA1 was visualized by confocal microscopy. Mutants of H155A, E156Q, D166A, E170A, and D171A were localized on membrane while H159A and E167A mutants were localized in cytosol under both permeabilized and non-permeabilized conditions. This result was confirmed by cell surface biotinylation analysis. It was interesting that pEGFPN1-rbCLCA1 was localized regardless of mutation. The result suggests that rbCLCA1 localization may be affected by H159 and E167 of metalloprotease motif and the region from H155 to E167 may have an effect on rbCLCA1 processing. A next study was conducted to understand biochemical property of putative catalytic site of rbCLCA1 for its processing. Among aforementioned site-directed mutation constructs, H155A, E156Q, and H159A were chosen to evaluate the effect of Ca2+, Zn2+, or Mg2+ on intracellular rbCLCA1 processing. Phosphate buffered saline (PBS) was added to distribute HEK293T cells overexpressing each construct in equal volume for each ion or combination. To see if rbCLCA1 is detached from cell surface according to pH change, pHs of acid to basic conditions were chosen. Autoproteolytic activity of rbCLCA1 was identified by co-transfection method. There was no cleavage product when zinc ion was treated to rbCLCA1, H155A, or H156Q lysates which were overexpressed in HEK293T cell. The same steps were taken to test on pEGFPN1-rbCLCA1 and its mutants. Zn2+ addition to pEGFPN1-rbCLCA1 and Hmut resulted in cleavage product between 30kDa and 50kDa, while Bmut did not show any products. It suggests that 167E may have an effect on rbCLCA1 degradation. It is assumed that rbCLCA1 may take part in signal transduction with other molecules based on the membrane stripping test under various pH conditions. The result leads to a conclusion that rbCLCA1 cleavage may be dependent on Zn2+ and pH. The result prompts to consider that other cellular environments may affect rbCLCA1 processing since the result is similar between an acidic condition treatment and Zn2+ treatment. In an effort to diversify CLCA family members, cloning Xenopus laevis cDNA predicted to encode the xCLCA gene was conducted. Expressed sequence tags (EST) was obtained after screening National Center for Biotechnology Information (NCBI)?Basic Local Alignment Search Tool (BLAST). Total RNA was extracted, 1st stranded cDNA was synthesized by reverse-transcriptase (RT)?PCR, and 5?? or 3?? Rapid Amplification of cDNA Ends (RACE) was performed. Total 4 rounds of RACE were performed to obtain total cDNA sequence. Each tissue distribution pattern analysis result was achieved by semi-quantitative reverse transcription (RT) PCR or real?time PCR. As a result, a 2,958 bp xCLCA cDNA sequence with an open reading frame encoding 943 amino acids was obtained. Expression analysis showed that xCLCA was expressed in a number of tissues, with high expression in the brain, colon, small intestine, lung, kidney, and spleen, and poor expression in the oocyte, liver, and heart. Based on the results, it is assumed that gene expression pattern comparison between previously reported other CLCAs from X. laevis and xCLCA in this study was analyzed with by functional studies. In the present studies, metalloprotease motif was identified among several domains by characterizing rbCLCA1 and xCLCA. It was shown that the processing mechanism and zinc-ion dependency of the metalloprotease motif. It has been shown that there are two key sites for rbCLCA1 processing and membrane localizing (i.e., H159 and E167). These data demonstrated that rbCLCA1 may be not anchored in the plasma membrane. These studies provide possibility that rbCLCA1 may be a detachable cell surface protein, suggesting that it may participate in cell signaling pathway. Additionally, a novel CLCA from X. laevis was cloned, analyzed, and put into CLCA family members. Thus, these studies widen diversity in CLCA family members and open a new direction for studying of basic structure and diversity of CLCAs.

In order to elucidate rbCLCA1 processing and localization in intracellular level and to characterize HEXXH-E/D?specified metalloprotease motif, rbCLCA1 was overexpressed in human embryonic kidney 293T (HEK293T) cells. rbCLCA1 was inserted into either pEGFPN1 having C-terminal GFP or pcDNA3.1+/c-myc which has c-myc sequence adjacent to N-terminal signal sequence. Protein expression pattern was observed by both Western blot and immunocytochemistry. Western blot analysis with GFP antibody showed that rbCLCA1 processing was affected by deletion of metalloprotease motif which is referred to as ΔHEWAH (Hmut) and ΔLRWGVFDEYNED (Bmut) respectively, producing only ?130kDa instead of yielding ?130kDa and ?50kDa cleavage products. Metalloprotease-specific mutation was introduced by site-directed mutagenesis of H155A, E156Q, H159A, D166A, E167A, E170A, and D171A on pcDNA3.1+/c-myc-rbCLCA1. Membrane localization of rbCLCA1 was visualized by confocal microscopy. Mutants of H155A, E156Q, D166A, E170A, and D171A were localized on membrane while H159A and E167A mutants were localized in cytosol under both permeabilized and non-permeabilized conditions. The result leads to a conclusion that rbCLCA1 cleavage may be dependent on Zn2+ and pH. The result prompts to consider that other cellular environments may affect rbCLCA1 processing since the result is similar between an acidic condition treatment and Zn2+ treatment.
