And also, the protective effect of flavonoids, kaempferol on MPTP-induced cytotoxicity was analysed. In addition, the microscopic observation was performed on C6 glioma cells treated with MPTP or kaempferol. The results were as follows : 1. MPTP significantly decreased cell viability in a dose-dependent manner compared with control after treating C6 glioma cells with 5-45 μM of MPTP for 48 hours, respectively. 2. XTT90 and XTT50 values were measured at 15 μM and 35 μM of MPTP, respectively after cultured C6 glioma cells were treated with MPTP for 48 hours. 3. The cytotoxicity of MPTP was highly-toxic by the toxic criteria of Borenfreund and Puerner. 4. In the protective effect of kaempferol on the MPTP-induced cytotoxicity, kaempferol significantly increased cell viability according to the treated dosage in C6 glioma cells pretreated with kaempferol for 2 hours before the treatment of MPTP. 5. In the light microscopic observation, MPTP decreased cell number and cell processes in dose-dependently. While, kaempferol increased cell number and cell processes compared with MPTP. From these results, it is suggested that MPTP, Parkinsonism-induced agent showed highly-toxic on cultured C6 glioma cells, and also, kaempferol of flavonoids was effective in the prevention of the MPTP-induced cytotoxicity.
