마비성패류독소의 분석은 AOAC 959.08 official method (OM)에 근거한 동물시험법(AOAC MBA)를 표준법으로 하여 전 세계적으로 이루어지고 있으나 정량한계가 높고, 염이나 중금속 등의 방해물질에 의해 영향을 받으며, 독소 함량이 높을 경우 편차가 큰 단점을 가지고 있다. 또한 동물윤리 문제와 관련하여 실험동물의 사용을 배제하거나 최소화하고자 하는 여론이 확산되고 있어 전 세계적으로 동물시험법의 사용빈도를 줄이거나 완전히 배제할 수 있는 대체 시험법을 찾으려는 노력을 진행하고 있다. 따라서 본 연구에서는 동물시험법을 대체할 분석방법으로 가장 많은 연구가 이루어지고 있는 HPLC pre-column oxidation법과 HPLC post-column oxidation법, 그리고 분석시간이 짧고 한 번에 모든 독소를 분석할 수 있는 LC-MS/MS법을 대상으로 우리나라 시료특성에 부합하고 규제를 목적으로 하는 분석환경에 적용 가능한지를 알아보기 위하여 동 분석법에 대한 유효성 검증과 동물시험법과의 비교 분석을 실시하였다. 그리고 유효성 검증 및 동물시험법과의 비교 분석에서 우리나라 시료특성에 가장 부합하고 해역의 모니터링 방법으로 이용 가능한 방법을 선정하여 패류양식장에서 생산되는 굴과 진주담치를 대상으로 총 독소함량과 독소 구성성분을 분석하여 패류 품종별 및 시기별 총 독소함량 및 독소 구성성분의 변화를 알아보았다. HPLC pre-column oxidation법으로 표준물질을 이용하여 분석하고 유효성 검증을 실시한 결과, 직선성의 상관계수(r2)는 0.99 이상으로 매우 양호하였다. 굴 추출물로 제조한 표준용액의 정량한계는 0.100 μg STX eq/g으로 동물시험법의 정량한계를 약 4배 향상시켰다. 일내 정확성 및 정밀성 분석에서 회수율과 상대표준편자는 각각 77.0~103.3%, 2.0~5.7%, 일간 정확성 및 정밀성 분석에서 회수율과 상대표준편차는 각각 77.3~101.3%, 2.4~6.0%였으며, 회수율은 75.2-112.1%로 AOAC에서 요구하는 기준에 부합하였다. 또한 동물시험법과의 비교에서도 상관계수(r2)는 0.8976으로 양호한 상관관계를 가지고 있었다. 그러나 동 분석법은 우리나라 패류에서 많이 검출되고 있는 GTX1와 GRX4, GTX2와 GTX3의 독소분리가 어려웠으며, 독소 함량이 높게 검출되는 경우 분석시간이 많이 소요되는 단점이 있었다. HPLC post-column oxidation법으로 표준물질을 이용하여 분석하고 유효성 검증을 실시한 결과, 모든 개별 독소성분의 분리가 가능하였으며 직선성은 상관계수(r2)가 0.99 이상으로 매우 양호하였다. 굴과 진주담치 추출물로 제조한 표준용액의 정량한계는 각각 0.0862 및 0.0832 μg STX eq/g으로 동물시험법의 정량한계를 약 4.6배 향상시켰다. 굴과 진주담치 추출물로 제조한 표준용액을 이용한 반복성 시험에서 상대표준편차는 모두 6% 이하였으며, 시험소 내 재현성 시험에서 HorRat value는 0.27~1.20 범위였고, 회수율은 83.5∼112.2% 범위였다. 대부분의 결과가 AOAO (2002)에서 제시하고 있는 기준에 부합하였으며, 동물시험법과의 비교분석에서도 두 시험법간의 상관계수는 굴과 진주담치에서 각각 0.9534와 0.9109로 매우 양호한 상관관계를 나타내었다. LC-MS/MS법으로 표준물질을 이용하여 분석을 실시한 결과, 모든 독소성분을 동시에 분리할 수 있었으며, 상관계수(r2)가 0.999 이상으로 매우 양호한 직선성을 나타내었고, 15분 이내에 정량이 가능하였다. 표준물질의 정량한계는 0.027 μg STX eq/g이었다. 표준물질을 이용한 분석법의 일내(Intra-day) 정확성 및 정밀성 분석에서 회수율과 상대표준편자는 각각 88.6%~106.1%와 1.0~5.4%이었으며, 일간(Inter-day) 정확성 및 정밀성 분석에서 회수율과 상대표준편차는 각각 88.5%~105.9%와 1.0%~4.4%으로 AOAC (2002) 기준에 부합하였다. 그리고 동물시험법과의 비교분석에서도 두 시험법간의 상관계수는 굴과 진주담치에서 각각 0.9211과 0.8878로 매우 양호한 상관관계를 나타내었다. 그러나 굴과 진주담치 추출물을 이용하여 제조한 표준용액에서 회수율은 59.4%~91.0% 및 60.2%~90.2% 범위로 일부 독소 성분의 회수율이 낮았다. 회수율만 개선된다면 매우 유효한 분석방법이 될 것으로 판단되었다. 유효성 검증 및 동물시험법과의 비교 분석에서 HPLC post-column oxidation법이 우리나라 시료 특성에 잘 부합하였으며, 동물시험법을 대체하여 해역의 모티터링을 위한 시험법으로 이용 가능할 것으로 판단되었다. 따라서 동 분석법을 이용하여 매년 마비성패류독소가 발생하고 있는 진해만에서 굴과 진주담치를 대상으로 2010년 1월부터 12월까지 월별 총 독소 함량 변화 및 독소 구성성분에 대한 분석을 실시하였다. 마비성패류독소는 3월 하순에 처음 발생하여 점차적으로 독소함량이 증가하다가 4월 중에 최고치를 나타낸 다음 급격하게 감소하기 시작하여 6월에는 완전히 소멸하였으며, 그 외의 시기에는 마비성패류독소 발생이 없었다. 마비성패류독소는 수온 9℃ 내외에서 발생하여 12℃ 내외에서 최고치를 나타낸 다음 15℃ 이상이 되는 5월 초순에 급격히 감소하기 시작하였다. 총 독소함량은 굴에서는 39~866 μg STX eq/100g 범위였으며, 진주담치에서는 37~5,427 μg STX eq/100g 범위였다. 독소 함량이 가장 높았을 때 진주담치의 독소 함량은 굴에 비하여 약 6.3배 높았다. 그리고 진주담치가 굴에 비하여 발생 시기는 다소 빠르고 소멸 시기는 늦었다. 진해만에서 생산되는 굴의 마비성패류독소 주요 구성성분은 GTX2, GTX1 및 GTX4였으며, 이들 3개 독소 성분이 전체 구성성분의 70.8∼97.1%를 차지하였고, 진주담치에서는 GTX3, STX 및 GTX2가 주요 구성성분이었으며, 이들 3개 독소 성분이 전체 구성성분의 88.5∼96.3%를 차지하였다.

Paralytic shellfish poisoning (PSP) is caused by consumption of shellfish containing certain level of neurotoxins produced by naturally occurring phytoplankton group and PSP toxins are accumulated in filter feeding shellfish through food chain. These toxins act as potent neurotoxins through the blockage of the sodium channels of mammals and result in a range of symptoms including numbness, respiratory distress, potentially paralysis and death. Consumption of shellfish contaminated with these toxins may result in a serious risk to the health of human consumer. Hence, the accumulation of paralytic shellfish poisoning toxins in shellfish creates a serious public health problem and affect to fisheries industry. The health standards of most countries indicate that live bivalve mollusks for human consumption must not contain PSP toxins in total that exceed 80 μg/100g STX eq. For this reason, many countries have monitoring and regulatory systems including routine sampling of shellfish fresh for the presence of biotoxins and the examination of water samples for the presence of toxin producing phytoplakton. The mouse bioassay (MBA), developed in 1937 to check toxicity in the acidic extracts of mussel, is the worldwide preferred testing method used as routine monitoring programs of paralytic shellfish poisoning toxins for more than 50 years, and is the official method of Association of Official Analytical Chemists (AOAC). The time from exposure to death of mouse is used to estimate the amount of toxins present in shellfish in the MBA, and the assay has a detection limit of 40 μg STXeq/100g. MBA has a additional advantage of reporting the total toxicity of samples in a short period of time, and it does not require specific instruments. But, this method has low sensitivity and high limit of quantification and interferences from other components in the extract, and it can not determine toxic profile. And there are ethical problems against the continued use of this live mouse assay. For this reason, since the 1980s, several alternative methods for paralytic shellfish poisoning toxin detection have been researched, such as bioassays, electrophoresis, chemosensors, immunoassays and instrumental analysis methods. The LC-FLD, HPLC pre-column oxidation method and post-column oxidation method, were successful in an inter-laboratory study, and was accepted by AOAC as the alternative method to the MBA. LC-MS/MS method also have been studied more recently. However, these methods may differ depending on the type of species and toxin profile. To application of alternative instrumental analysis method of mouse bioassay for PSP toxins in Korean shellfish, I reviewed the reports related to alterative analysis method for PSP toxins such as HPLC pre- and post-column method and LC-MS/MS method. And the analysis conditions were optimized and validated. Comparative analysis of those methods and AOAC MBA method also was carried out in the same sample in order to check the validity. And then the suitable method was selected for Korean shellfish. The toxin profiles and total toxins amount of field sample were analysed by those methods for investigating the possibility of applying to Korean shellfish. In HPLC pre-column oxidation method, linearity was more than 0.99 for the working standard solutions of PSP toxins. The limit of quantification of total toxins was about 0.100 μg STX eq/g for spiked extracts of oyster and this was about 4.0-fold improvement in the detection capability of the conventional AOAC MBA method. The intra-day accuracy and precision of PSP toxins for spiked extracts of oyster were 77.0∼103.3% and 2.0∼5.7%, respectively. The inter-day accuracy and precision of PSP toxins for spiked extracts of oyster were 77.3∼101.3% and 2.4∼6.0%, respectively. The mean recovery of PSP toxins for spiked extracts of oyster was 75.2∼112.1%. Accuracy, precision and recovery were met the criteria of AOAC guidelines for single laboratory validation of chemical methods for dietary supplements and botanicals. But several toxin components(GTX1 and 4, GTX2 and 3, dcGTX2 and 3, C1 and 2) were not differentiated by HPLC pre-column oxidation method. In HPLC post-column oxidation method, the clear peak and the isolation of PSP toxins was obtained from the working standards solution of PSP toxins. The mean recovery of HPLC post-column oxidation method was 83.5∼112.2% and the relative standard deviation of repeatability test for spiked extracts of oyster and mussel was below 6%, which met the criteria of AOAC guidelines for single laboratory validation of chemical methods for dietary supplements and botanicals. The results of comparison study showed good correlations between the results of the HPLC post-column oxidation method and those of AOAC MBA. The correlation factors were 0.9534 and 0.9109 for oyster and mussel matrix, respectively. The analysis results of working standards solution of PSP toxins showed very reasonable and total analysis time was 15 minutes. The results of comparison study showed good correlations between the results of the LC-MS/MS method and those of AOAC MBA. The correlation factors were 0.9211 and 0.8878 for oyster and mussel matrix, respectively. But the recovery of PSP toxins for spiked extracts of oyster and mussel were 53.1∼91.0% and 60.2~90.2%, respectively. Some toxin components were below the criteria of AOAC guidelines for single laboratory validation of chemical methods for dietary supplements and botanicals. Considering the results of validation and characteristics of Korea shellfish, HPLC post-column oxidation method was considered the most desirable to adopt for analysis of PSP toxins in Korean shellfish. The toxin profiles and total toxin amounts of field sample collected in the coastal area of Korea where the PSP occurrence has been common were examined by HPLC post-column oxidation method. PSP toxins in Jinhae Bay were detected in late March and increased continuously and the highest values were shown in April. After the late of April PSP toxins had been decreased sharply and not detected in June. The highest total toxin level was recorded in mussel, and that was 6.3 times higher than that of the oyster. The major toxin components were GTX2, GTX1 and GTX3 (70.8∼97.1%) in oyster and were GTX3, STX and GTX2 (88.5∼96.3%) in mussel.

Linearity was more than 0.99 for the working standard solutions of PSP toxins. The limit of quantification of total toxins was about 0.0862 μg STX eq/g for spiked extracts of oyster and 0.0832 μg STX eq/g for that of mussel and this was a 4.6-fold improvement in the detection capability of the conventional AOAC MBA method. The mean recovery of HPLC post-column oxidation method was 83.5∼112.2% and the relative standard deviation of repeatability test for spiked extracts of oyster and mussel was below 6%, which met the criteria of AOAC guidelines for single laboratory validation of chemical methods for dietary supplements and botanicals. The results of comparison study showed good correlations between the results of the HPLC post-column oxidation method and those of AOAC MBA. The correlation factors were 0.9534 and 0.9109 for oyster and mussel matrix, respectively. In LC-MS/MS method, the clear peak and the isolation of PSP toxins was obtained, and linearity was more than 0.999 for the working standard solutions of PSP toxins. PSP toxins in Jinhae Bay were detected in late March and increased continuously and the highest values were shown in April. After the late of April PSP toxins had been decreased sharply and not detected in June. The highest total toxin level was recorded in mussel, and that was 6.3 times higher than that of the oyster. The major toxin components were GTX2, GTX1 and GTX3 (70.8∼97.1%) in oyster and were GTX3, STX and GTX2 (88.5∼96.3%) in mussel.
