천연유래 보존료 (안식향산, 소르빈산, 프로피온산)는 원료에 천연적으로 존재하거나 발효 중에 생성될 수 있어 인위적으로 식품의 보존을 위해 첨가하는 경우와 구분이 되지 않아 첨가 여부를 판단하기가 어렵다. 또한, 식품 중 천연유래 보존료 함유량 분석 연구 자료가 부족하여 식품의 안전성 확보를 위해 광범위한 모니터링 및 기초자료가 필요한 실정이다. 따라서 본 연구에서는 과일류 및 향신료에 대하여 천연유래 보존료의 함유량을 문헌을 통해 조사하고, 국내에 유통되고 있는 과일류 및 향신료를 수거하여 천연유래 보존료의 함유량 조사를 실시함으
천연유래 보존료 (안식향산, 소르빈산, 프로피온산)는 원료에 천연적으로 존재하거나 발효 중에 생성될 수 있어 인위적으로 식품의 보존을 위해 첨가하는 경우와 구분이 되지 않아 첨가 여부를 판단하기가 어렵다. 또한, 식품 중 천연유래 보존료 함유량 분석 연구 자료가 부족하여 식품의 안전성 확보를 위해 광범위한 모니터링 및 기초자료가 필요한 실정이다. 따라서 본 연구에서는 과일류 및 향신료에 대하여 천연유래 보존료의 함유량을 문헌을 통해 조사하고, 국내에 유통되고 있는 과일류 및 향신료를 수거하여 천연유래 보존료의 함유량 조사를 실시함으로써 천연유래 판단을 위한 근거자료를 마련하고자 하였다.
대상 시료는 과일류 (사과, 감귤, 대추, 포도 등) 874건, 향신료 (후추, 계피, 강황 등) 493건과 추가로 조미료의 역할로 많이 사용되는 버섯류(양송이, 표고버섯 등) 98건을 수거하여 총 1,465건이었다. 각각의 보존료 분석은 HPLC-UVD(과일류 중 안식향산 및 소르빈산), LC-MS/MS(향신료 중 안식향산 및 소르빈산) 및 GC-MS(프로피온산)를 사용하였다. 과일류 및 향신료 중 분석법의 유효성을 검증하기 위해서 직선성, 회수율, LOD, LOQ 및 측정불확도를 검토한 결과, 모든 항목에서 양호한 결과를 나타내었다.
함유량 조사 결과 안식향산은 대상시료 1,465건 중 502건의 시료에서 검출되었으며, 과일류에서는 ND-764.11 mg/L의 범위로 크랜베리(128.39-764.11 mg/L), 아로니아(ND-290.73 mg/L) 순으로 높게 검출되었고, 향신료에서는 ND-391.99 mg/L의 범위로 계피(54.40-391.99 mg/L), 정향(54.10-263.18 mg/L) 순으로 높게 검출되었고, 버섯류에서는 ND-25.39 mg/L의 범위로 양송이버섯(2.20-25.39 mg/L), 느타리버섯(ND-18.21 mg/L) 순으로 높게 검출되었다. 소르빈산은 대상시료 1,465건 중 371건의 시료에서 검출되었으며, 과일류에서는 ND-113.34 mg/L의 범위로 라임(ND-113.34 mg/L), 레몬(10.07-83.62 mg/L) 순으로 높게 검출되었고, 향신료에서는 ND-57.70 mg/L의 범위로 바질잎(ND-57.70 mg/L), 정향(ND-16.90 mg/L) 순으로 높게 검출되었고, 버섯류에서는 ND-3.83 mg/L의 범위로 양송이버섯에서 검출되었다. 프로피온산은 대상시료 1,465건 중 1,323건의 시료에서 검출되었으며, 과일류에서는 ND-172.02 mg/L의 범위로 두리안(8.18-172.02 mg/L), 아사이베리(0.33-108.92 mg/L) 순으로 높게 검출되었고, 향신료에서는 ND-188.21 mg/L의 범위로 생강(ND-188.21 mg/L), 자소엽(ND-58.86 mg/L) 순으로 높게 검출되었고, 버섯류에서는 ND-4.29 mg/L의 범위로 팽이버섯(0.34-4.29 mg/L), 송화버섯(1.40-2.35 mg/L) 순으로 높게 검출되었다.
본 연구의 결과를 통해 과일류 및 향신료에 대한 천연유래 보존료 함유량 DB를 마련하였고, 향후 식품 검사 시 보존료 사용기준 위반 판정으로 인한 민원제기나 국가 간 무역마찰 시 기초자료로 활용될 수 있을 것이다.

Natural preservatives (benzoic acid, sorbic acid and propionic acid) are contained already in natural raw materials or occur during food fermentation process, so that the judgment whether the addition was done artificially or not is difficult. Also, there are little reports about natural preservatives contents analysis research on foods. Therefore, the broad scale monitoring for acquirement of various data base related to natural preservatives in fruits and spices is necessary to ensure for food safety.
In this study, we investigated the levels of natural preservatives of benzoic acid, sorbic acid and propionic acid in fruits and spices. Total 1,465 samples(874 fruits, 493 spices and 98 mushrooms) were collected for determination of the contents of preservative naturally produced. The quantitative analysis was performed using HPLC-UVD analysis for benzoic acid and sorbic acid in fruits, LC-MS/MS analysis for benzoic acid and sorbic acid in spices, and GC-MS analysis for propionic acid. We carried out the validation for analytical methods of natural preservatives in fruits and spices. That methods showed good results in terms of linearity, recovery, precision, LOD, LOQ and uncertainty.
Benzoic acid, sorbic acid and propionic acid in total 1,465 samples of raw materials were detected in 502 samples, 371 samples and 1,323 samples, respectively. The amounts of benzoic acid were in the range of ND-764.11 mg/L in fruits, ND-391.99 mg/L in spices, ND-25.39 mg/L in mushrooms, and levels of cranberry(128.39-764.11 mg/L), cinnamon(54.40-391.99 mg/L) and button mushroom(2.20-25.39 mg/L) were the highest in fruits, spices and mushrooms, respectively. The amounts of sorbic acid were in the range of ND-113.34 mg/L in fruits, ND-57.70 mg/L in spices, ND-3.83 mg/L in mushrooms, and levels of lime(ND-113.34 mg/L), basil leaves(ND-57.70 mg/L) and button mushroom(ND-3.83 mg/L) were the highest in fruits, spices and mushrooms, respectively. The amounts of propionic acid were in the range of ND-172.02 mg/L in fruits, ND-188.21 mg/L in spices, ND-4.29 mg/L in mushrooms, and levels of durian(8.18-172.02 mg/L), ginger(ND-188.21 mg/L) and enoki mushroom(0.34-4.29 mg/L) were the highest in fruits, spices and mushrooms, respectively.
The results obtained in this study provide fundamental data regarding naturally occurring preservatives in fruits and spices. Moreover, utilize the results as basic data in case of civil complaint or trade friction due to breach judgment on utilization standard of preservatives when inspecting foods.
