대량 염기서열 분석 기술의 발달에 맞추어 다양한 포유류에 대한 전장유전체분석이 완료되었으며 향후 더욱 많은 게놈분석정보가 활용 가능 할 것으로 예상된다. 그러므로 직접적인 염기서열의 변화 이외에도 환경적 요인과 유전적 요인의 상호작용에 의한 후성유전체의 변화에 대한 연구가 가능하게 되었다. 후성유전 관련변화 중 DNA 메틸화는 생화학적 안정성이 높아 우발적 변화가 적은 주요 후성유전 기전의 하나이다. 특히 사이토신과 구아닌 이중핵산(CpG)의 사이토신 메틸화는 포유류의 초기 발달과정과 세포분화특이성과 관련한 유전자 발현조절 및 후성유전적 변화와 관련성이 높아 이에 대한 많은 연구가 진행되고 있다. 포유류의 유전체에서 유전자 발현 조절에 관여하는 프로모터 지역 또는 CpG 밀도가 높은 지역의 DNA메틸화의 양상은 유전자의 발현억제, 염색사의 구조조절, 중심체 부근에 위치한 반복염기서열의 발현억제, X 염색체 불활성화, 유전자 각인 및 특정 질병의 유발과도 연관성이 깊은 것으로 알려져 있다. 인간의 경우 고해상도의 전장후성유전체 분석이 2014년 ENCODE 사업을 통해 진행되었으며 이를 통하여 유전자 발현조절 및 세포 또는 조직의 발생학적 특이성과 관련한 DNA메틸화의 역할에 대한 많은 결과가 도출되었다. 의생명과학분야의 주요 동물모델인 돼지의 경우 그 중요성에도 불구하고 현재까지 단일 염기서열 수준의 고해상도 DNA메틸화 연구결과가 발표되지 않았으며, 따라서 본 연구에서는 해당 분야의 선행연구의 일환으로 발현과 연관된 CpG 밀도가 높은 지역을 포함하는 저비용 고효율의 reduced representation bisulfite sequencing 기술을 이용하여 다양한 돼지 조직의 (대뇌피질, 후각상피, 간, 근육, 비장, 폐 대식세포) 단일염기수준의 고해상도의 전장유전체적 DNA 메틸화 변화에 대한 분석을 실시하였다. 평균적으로 유전자 발현 조절과 연관된 CpG 밀집지역(CGI)의 약 40 %의 CpG와 프로모터지역에 해당하는 전사시작위치(TSS)로부터 5’쪽으로 2 kbp 지역의 CpG들의 약 62 %를 분석 할 수 있었다. 전체적인 DNA 메틸화 양식은 인간의 것과 유사하였고, 분석된 돼지의 DNA 메틸화 양식은 유전자 발현조절과의 연관성을 확인하기 위하여 유전자 발현 정보와 비교 분석하였다. 본 연구에 이용된 조직 별로 메틸화 정도의 차이가 있는 사이토신을 유전학적 위치를 기준으로 관련 유전자의 주석을 달고 연결된 관련 유전자의 발현 양식을 DNA 메틸화 정도와 상관 분석하였다. 이러한 연구 결과는 돼지 후성유전체 관련연구를 위한 기초자료로써 돼지 후성유전체 연구의 토대 구축에 크게 기여 할 것으로 사료된다.

The publications of draft whole genome assemblies for multiple mammalian species and development in technologies allowed the acceleration of studies on epigenetic features in the genomes. DNA methylation, especially cytosine methylation of cytosine and guanine dinucleotides (CpG), is well known and important phenomena which associate with developmental stage and cell type specific epigenetic memory. DNA methylation is stable and heritable component of epigenetic regulation. DNA methylation patterns of mammalian genomes in promoter or high density of CpGs regions (CpG islands) correlate with transcriptional repression, chromatin organization, silencing of repetitive and centromeric sequences, X chromosome inactivation, genetic imprinting and some diseases. In humans, high resolution genome-wide epigenetic associated data were produced from the ENCODE project and EPIGENOME ROADMAP project recently, accelerating our knowledge on the epigenetic modification of the genome which could be critical to understand gene regulation during development or determining tissue or cell types. Pigs have been recognized as an important livestock animal for the use of biomedical animal models. However, a genome wide single base pair resolution reference DNA methylation map is not available for the species. Here we reports the high resolution genome wide DNA methylation profiling in the pig genome from multiple tissues including neocortex, olfactory epithelium, liver, muscle, spleen, and pulmonary alveolar macrophage using reduced representation bisulfite sequencing (RRBS) which is cost effective method to study CpG rich regions. In our result, on average, about 40% and 62% of CpG sites which locates on CpG islands and 2kb upstream regions from the transcript start site were covered, respectively. The global patterns of DNA methylation were similar with those of humans. Observed DNA methylation patterns were analyzed with the gene expression patterns of relevant tissues in order to reveal the gene expression regulating relationships. The detected differentially methylated cytosines (DMCs) among the five different tissues were annotated by Ensembl gene annotation on the basis of their genomic locations. Our results should play an essential role as an early study to establishing the framework to understand the relationship between epigenetics and gene expression of the swine genome.
