유전자 각인(genomic imprinting)이란 특정 유전자가, 기원한 부모에 따라 그 발현 양상이 달라지는 현상으로, 두 대립 유전자 중에서 부계 또는 모계의 유전자만이 발현되는 현상이다. 1980년대 초반에 각인 유전자의 존재가 밝혀진 이래로 현재까지 인간과 생쥐에서 약 50여개의 각인 유전자가 밝혀졌으며 이러한 각인 유전자가 태아와 태반의 생장, 세포 분화, 성체의 행동 등과 밀접한 관련이 있다고 보고되었다. 최근들어 이러한 유전자 각인 현상과 암을 비롯한 인간의 다양한 유전적 질병과의 연관성이 밝혀지고 있어 유전자 각인
유전자 각인(genomic imprinting)이란 특정 유전자가, 기원한 부모에 따라 그 발현 양상이 달라지는 현상으로, 두 대립 유전자 중에서 부계 또는 모계의 유전자만이 발현되는 현상이다. 1980년대 초반에 각인 유전자의 존재가 밝혀진 이래로 현재까지 인간과 생쥐에서 약 50여개의 각인 유전자가 밝혀졌으며 이러한 각인 유전자가 태아와 태반의 생장, 세포 분화, 성체의 행동 등과 밀접한 관련이 있다고 보고되었다. 최근들어 이러한 유전자 각인 현상과 암을 비롯한 인간의 다양한 유전적 질병과의 연관성이 밝혀지고 있어 유전자 각인 현상은 더욱 주목받고 있다. 본 과제에서 연구진은 각인 유전자들이 염색체상의 특정 위치에 모여있다는 점에 착안하여 주변에 다른 각인 유전자의 존재가 알려져 있지 않은 각인 유전자들을 중심으로 새로운 각인 유전자의 탐색을 시도하였고 관련 유전자의 DNA methylation, 발현양상, antisense RNA의 발현 양상에 대해 연구하였다.
이미 알려진 인간의 각인 유전자 주위에 존재하는 유전자들을 GenBank database로부터 알아낸 후, 이 인간 유전자들과 상동성을 보이는 생쥐 유전자들을 BLAST 프로그램을 이용하여 GenBank database로부터 찾아내고 이들의 각인 여부를 조사하였다. 유전자 각인 여부를 조사하기 위해서는 개체가 지니고 있는 두 대립유전자가 각각 어느 쪽 부모에서 유전되었는지를 알아야 한다. 이를 위하여 생쥐 종간의 유전자 다형(polymorphism)을 이용한 Restriction Fragment Length Polymorphism (RFLP) 방법을 이용하였다. 1차년도에는 생쥐 Peg1/Mest 유전자 주변에 존재하는 유전자들을 database로부터 retrieve하여 PCR, sequencing을 통해 적절한 polymorphism을 동정한 후, 이를 이용하여 유전자의 각인 여부를 종간교배 F1에서 확인하였다.
2차년도에는 1차년도 연구결과에 의해 각인된 것으로 알려진 Mit1/Lb9 유전자의 genomic DNA structure, expression pattern등을 조사하였고, 이 과정 중에서 인접한 유전자인 Copg2유전자를 발견하여 Copg2 유전자의 유전자 각인여부, 발현양상, 생체 내 기능 유추 등과 관련된 연구를 수행하였다.
3차년도에는 Copg2 유전자의 promoter에 존재하는 CpG-rich region의 DNA methylation 양상을 조사하고, 이와 관련이 있을 것으로 예상되는 새로운 Copg2 antisense transcript를 동정하였다. 또 이 Copg2 antisense의 유전자 각인 여부를 조사하였고 in situ hybridization, immunocytochemistry 등을 통해 Copg2 유전자의 조직발현 특이성, Copg2 단백질의 세포 내 존재 영역을 조사하였다.
1차 년도의 실험 결과 PEG1/MEST 유전자와 연관된 Expressed Sequence Tag (EST) 중 하나인 stSG1728의 생쥐 homologue (accession locus : AA61151)가 생쥐의 뇌에서 각인되어 있음을 밝혔다. 본 연구진은 이 유전자를 Mit1 (Mest-linked imprinted transcript 1: GenBank Accession No. AF217545)이 라 명명하였으며, 이 유전자의 genomic DNA의 구조를 밝히고 조직별, 발생단계별 각인 여부를 조사한 결과 주로 뇌에서 많이 발현되고 모계 각인되어 있음을 알 수 있었다. 2차 년도에서는 Mit1 유전자의 genomic DNA 구조를 조사하는 과정에서 Mit1/Lb9을 intron에 포함하고 있는 새로운 유전자인 Copg2 유전자를 발견하고 이 유전자의 각인 여부 및 발현 양상을 조사한 결과 모든 조직에서 발현되며 부계 각인되어 있음을 알 수 있었다. Copg2 유전자는 세포내의 골지체에서 물질 수송에 관여하는 것으로 알려진 Copg와 매우 유사한 아미노산 서열을 지니고 있다. 3차 연도에는 Copg2 유전자의 promoter 부위의 CpG island의 메틸화 상태를 조사한 결과 이 부위가 각인 현상과 밀접한 관계가 있는 차등메틸화 부위(DMR)임을 알 수 있었다. 그리고 Mit1/Lb9을 포함해서 Copg2 유전자의 전체 부위에 걸쳐 antisense 전사체가 존재하며 이들이 그 위치에 따라 부계 혹은 모계 각인 되어 있음을 밝혔다. In situ hybridization 실험을 통해 조사한 결과, Mit1/Lb9은 소뇌와 대뇌의 전뇌 부위에서 주로 발현되고, Copg2은 소뇌와 대뇌의 posterial brain 중 intermediate gray layer에서 주로 발현됨을 알 수 있었다. Copg1과의 기능 유추를 위해 Copg1과 Copg2의 세포내 위치를 fluorescent microscopy로 조사한 결과 두 단백질의 세포 내 위치가 중첩되어 나타나는 것을 관찰하여, 두 단백질의 기능이 유사함을 추정할 수 있었다.

To search for novel imprinted genes, we exploited the clustering behavior of imprinted genes and investigated imprinting status of genes closely linked to the known imprinted genes. The object of this study is the identification and characterization of novel imprinted genes nearby a known imprinted gene. We also tried to investigate other aspects of newly identified imprinted genes, such as gene expression pattern, DNA methylation, and existence of antisense transcript(s).
Informations were gathered especially about imprinted genes which are present on a chromosomal region considered to harbor imprinted genes through UPD study of mice. We focused on the region neighboring mouse Peg1/Mest gene, which had already been identified to be imprinted maternally. In the first year, expressed gene sequences, nearby mouse Peg1/Mest, were identified and retrieved from GenBank In this process, we used draft human genome sequence and its synteny to mouse chromosomes. We identified RFLPs for each gene between inbred mouse strains used in this study, and recruited them to assay the expression of each parental allele in the F1 generated by breeding. In the second year, investigation for the genomic structure and expression of Mit1/Lb9, a novel imprinted transcript identified in the first year, was undertaken. We also assayed the allele-specific expression pattern of mouse Copg2 gene, which was found during the investigation of genomic structure of the Mit1/Lb9. Expression pattern of Copg2 was examined through various mouse tissues by Northern blot analysis.
In the last year, DNA methylation of a CpG-rich region embedded in the putative mouse Copg2 promoter was examined by Sodium-Bisulfite sequencing. We searched for antisense transcripts of Copg2 and tested whether the antisense transcripts were also imprinted. For the speculation of the function of Copg2, subcellular localization of Copg2 protein was compared with that of Copg1 through fluorescence microscopy. In-situ hybridization was tried to precisely identify distribution of Copg2 expression within the mouse brain.
It was verified that the EST AA61151, mouse homologue of human EST stSG1728 located around PEG1/MEST, was imprinted. EST AA61151 was named as Mit1/Lb9. The transcribed products of Mit1/Lb9 seem to be highly heterogeneous since multiple transcripts were detected in Northern blot analysis. It is of interest that no considerable open reading frame could be predicted from the sequences of Mit1/Lb9 cDNAs. Mit1/Lb9 is maternally imprinted in adult brain, heart, lung, muscle, embryos and, neonates. Sequencing analyses showed that Mit1 is located within an intron of an adjacent gene, Copg2. Copg2 consists of 24 exons within the >40 kb genomic region, being expressed ubiquitously in mouse tissues with a partial imprinting pattern in embryos, neonates, and adult brain. A CpG island was found in the promoter region of Copg2. We presumed the promoter by determination of transcription start site of mouse Copg2 through primer extension analysis. The CpG island of Copg2 promoter region was differentially methylated. Novel antisense transcripts of Copg2, including Mit1/Lb9, were identified. They showed monoallelic expression. In situ hybridization analyses showed that Mit1/Lb9 was expressed in cerebellum and frontal cortex and Copg2 was expressed in cerebellum and intermediate gray layer of posterial brain.
