VLPs를 복강 내 주입한 그룹은 항-RGNNV 캡시드 단백질에 대한 항체 및 중화항체를 효과적으로 유도 해 냈으며 RGNNV 공격시험에 대한 보호 면역을 나타냄을 확인하였다. RGNNV 공격시험 결과, 복강 내로 RGNNV VLP를 면역시킨 그룹은 100%의 생존율을 보인 반면, phosphate-buffered saline (음성대조군: PBS)을 주입한 그룹은 37%의 생존율을 나타내었다. RGNNV VLPs를 경구로 면역 시킨 그룹은 중화항체를 유도 할 수 있었고, RGNNV 공격시험 시 57%의 양호한 생존율을 나타내었다. 본 연구 결과는 항-RGNNV 캡시드 단백질을 생산하는 S. cerevisiae 로부터 생산된 RGNNV VLPs가 RGNNV 감염 예방을 위한 백신으로서 우수한 잠재성을 가지고 있음을 제시한다.

NNVs infect more than 40 marine fish species and the infection with NNV inflicts serous economic losses in marine fish cultivation. Virus-like particles (VLPs) are promising vaccine candidate for preventing infection of NNV. VLPs are multimeric protein complexes consisting of recombinant virus capsid proteins, whose shapes are similar to native virions. VLPs have been considered as a new vaccine platform because they have the ability to induce neutralizing antibodies efficiently and are not infectious since they do not possess viral genomes. However, there have been few studies of protective immunity employing virus challenge following immunization with NNV VLPs in fish. In the present study, we produced red-spotted grouper NNV (RGNNV) VLPs in Saccharomyces cerevisiae (S. cerevisiae) Y2805 and investigated prophylactic immune responses and protective immune responses in convict grouper (Epinephelus septemfasciatus) following intraperitoneal injection and oral gavage with the RGNNV VLPs. The intraperitoneal injection group elicited anti-RGNNV capsid protein antibodies and neutralizing antibodies with high efficacy, and provided the fish with full protection against RGNNV challenge: 100% of the immunized fish survived, compared to only 37% survival of the control fish receiving phosphate-buffered saline (negative control : PBS). The oral immunization group provoked neutralizing antibodies systemically and exerted satisfactory protection against virus challenge: 57% of the fish in the oral immunization group survived. My results demonstrate that immunization with RGNNV VLP produced in S. cerevisiae has great potential as vaccine in fish.
