The inhibitory effect of ethanol extracts from Hizikia fusiforme(HF) on osteoarthritis was investigated in primary cultured rat cartilage cells and a monosodium-iodoacetate (MIA)-induced arthritis rat model. To identify the effects of HF Hot water extracts(HW), HF 20% ethanol extracts(HF20) and HF 50% ethanol extracts(HF50) against H2O2 (800 μM) treatment in vivo, cell survival was measured by the MTT assay. Cell survival increased by HW 900 μg/mL and HF20 700 μg/mL close to normal H2O2 treatment. To determine the anti-inflammatory effects of HF extracts, tumor necrosis factor alpha (TNF-α) and Interleukin-6(IL-6) expression were measured after lipopolysaccharide(LPS) activation. TNF-α and IL-6 levels were decreased by HF extract treatment in a dose-dependent manner. In this study, we examined the effect of HF administration on cartilage cell metabolism regarding the changes of anabolic and catabolic factors. As a result, HF showed markedly different treatment responsed that it enhanced gene expression of anabolic factors with decrease in the catabolic factors. For the in vivo animal study, osteoarthritis was induced by intra0articular injection of MIA into knee joints of rats. Consumption of a diet containing HF extract (250 and 500 mg/kg) for 4 weeks significantly inhibited the development and severity of osteoarthritis in rats. To determine the genetic expression of arthritic factors in articular cartilage, real-time PCR was applied by measuring matrix metalloproteinases (MMP-3, MMP-7), collagen type I, collagen type II, and aggrecan, and HF extract had protective effects at a concentration of 500 mg/kg. In conclusion, HF extract was able to inhibit articular cartilage degeneration by preventing extracellular matrix degradation and chondrocyte injury. It could be considered that HF extract may be a potential therapeutic treatment for degenerative osteoarthritis.
