In this study, elements of mammalian expression vector were investigated to improve cellular productivity and transgene stability in Chinese hamster ovary (CHO) cells. Methods: A genomic DNA library was constructed in green fluorescent protein (GFP) and screened to identify novel DNA regulatory element from CHO cells. Short hairpin RNA (shRNA) targeting the 3′-untranslated region of endogenous dihydrofolate reductase (DHFR) gene was contained in a plasmid vector, which express erythropoietin (EPO) and DHFR, and introduced into wild-type CHO cells. Stable cell pools expressing GFP were generated by transfection of a dual promoter vector or a bicistronic vector. Results: A novel regulatory DNA element (E77) was isolated and identified from CHO-K1 genomic DNA. E77-containing expression vector increased GFP and antibody expression levels in CHO cells and maintained the transgene stability upon removal of selection pressure. An expression vector including shRNA enhanced the transgene amplification rate and specific EPO productivity in wild-type CHO cells. Methods of expressing the selection marker gene affected expression heterogeneity in stable polyclonal cell populations. Unlike a dual promoter vector, a bicistronic vector generated only GFP-positive cells. Conclusion: Three vector engineering methodologies were investigated to improve the productivity, transgene stability, and expression homogeneity in stable CHO cells. These approaches may be applied to develop stable mammalian cell lines for biopharmaceutical production.
