Although it has been reported that Poria cocos (P. cocos) has effects on anti-oxidation and inhibition of melanogenesis, systemical in vivo studies for it clinical effects have not been shown yet. In this thesis, effects of P. cocos extracts-containing cream on the human skin barrier function have been clinically evaluated. Ten female adults between the ages of 20 and 35 were selected as the study's subjects, and the experiment was carried out for 4 weeks. All subjects were made to apply a cream containing 4% (v/v) of P. cocos extracts on the left side of the face, and the same cream not containing P. cocos extracts was applied on the right side of the face with equal amounts twice a day, morning and evening. To evaluate the skin barrier function, the skin’s moisture, percutaneous moisture damage level, and dead skin cells were measured. Also, before and after the experiment, a self-assessment regarding their own skin condition and satisfaction evaluation regarding the use of the test material were conducted via a survey. In regards to the level of improvement in moisture of the facial skin, the control group that used the cream not containing P. cocos extracts showed a result of 116.30% 2 weeks after use, and 106.44% 4 weeks after use. In the experimental group that used the cream containing P. cocos extracts, they showed a result of 300.41% 2 weeks after use, and 326.98% 4 weeks after use. Consequently, the moisture improvement level of the experimental group showed to be higher in comparison to the control group, there was a significant statistical difference (p<.001) among the groups, and there showed to be a significant difference (p<.001) as time progressed, thereby verifying the effectiveness of skin moisture improvement. In regards to the level of improvement in percutaneous moisture damage of the facial skin, the control group that used the cream not containing P. cocos extracts showed a result of 9.45% 2 weeks after use, and 10.11% 4 weeks after use. In the experimental group that used the cream containing P. cocos extracts, they showed a result of 20.13% 2 weeks after use, and 35.39% 4 weeks after use. Consequently, the percutaneous moisture damage improvement level of the experimental group showed to be higher in comparison to the control group, there was a significant statistical difference (p<.001) among the groups, and there showed to be a significant difference (p<.001) as time progressed, thereby verifying the effectiveness of percutaneous moisture damage level improvement. In regards to the level of improvement in dead skin cells of the facial skin, the control group that used the cream not containing P. cocos extracts showed a result of 33.46% 2 weeks after use, and 49.17% 4 weeks after use. In the experimental group that used the cream containing P. cocos extracts, they showed a result of 80.83% 2 weeks after use, and 87.50% 4 weeks after use. Consequently, the dead skin cell improvement level of the experimental group showed to be higher in comparison to the control group, there was a significant statistical difference (p<.05) among the groups, and there showed to be a significant difference (p<.001) as time progressed, thereby verifying the effectiveness of dead skin cell level improvement. Therefore, a superior effect in the improvement of skin moisture, percutaneous moisture damage, and dead skin cells was revealed in the experimental group that used the moisture cream containing P. cocos extracts in comparison to the control group that used the moisture cream not containing P. cocos extracts, and no particular abnormal skin reactions were observed as a result of the test material by any of the study subjects. This study is expected to result in P. cocos extracts being utilized as raw material for moisturizing cosmetics and having a positive influence on the improvement of skin barrier’s function
