본 연구에서는 기능성을 가진 난용성 물질들의 용해도와 생체이용률을 향상시키기 위한 방법으로 다양한 가용화 기술들을 제시하였다. 본 연구에서 사용된 가용화 기술에는 1) 기능성을 가진 저분자 물질을 생산할 수 있는 고온고압 가수분해기술과 2) 지용성 물질을 에멀젼화하여 수용화 할 수 있는 캡슐화 기술이 있다. 난용성 물질로써는 고분자물질인 콜라겐 단백질과 지용성물질인 시나몬 오일과 피쉬오일을 사용하였으며, 이러한 물질들을 외부로부터 안정하게 보호하고 기능성을 증대시키기 위하여 리포좀화, 나노에멀젼화, 다중층 에멀젼화과 같은 캡슐화 기술을 개발하였다. 다양한 캡슐화 기술로부터 최적의 포물레이션과 가공처리조건을 확인하였으며, 식품모델의 최종제품에 적용하여 안정성 테스트를 실시하였다. 1. 첫 번째 연구에서는 돈피 콜라겐을 가용화하기 위한 방법으로 고온고압 가수분해기술을 사용하여, 최종적으로 저분자화된 펩타이드를 생산하고자 하였다. 돈피콜라겐은 자체 제작된 고온고압 장치의 온도 150-250oC, 압력 350-3,900 kPa 조건에서 처리되었으며, 최적 펩타이드를 생산하기 위하여 1-10 kDa 분자량 크기 별로 분획하였다. 생산된 돈피가수분해물 및 분자량 크기 별 펩타이드 산물은 다양한 물리학적•이화학적 (온도 및 압력변화에 따른 노출량 계산, pH, 색도, 단백질 및 유리 아미노산 함량, 온도 및 압력 변화에 따른 회귀 모델, 유리 아미노산 함량변화의 삼차원 모델, 분자량 분포, 아미노산 조성, 펩타이드 수율) 및 기능학적 분석 (항산화 및 항노화 활성)을 실시하였다. 1) 고온고압 가수분해를 통해 생성된 돈피가수분해물은 온도와 압력이 높을수록 단백질 함량은 감소하나 유리 아미노산 함량은 유의적으로 증가하였으며, 전기영동 및 겔크로마토그래피 분석 결과를 살펴보면 온도와 압력의 증가로 인해 고분자에서 저분자 형태로 바뀌는 것을 확인할 수 있었다. 2) 돈피가수분해물 생성을 위한 고온고압장치의 최적 조건은 온도는 190 oC, 압력은 2,100 kPa이며 (PSH-IV), 단백질 가수분해 시 온도와 압력의 시너지 효과는 가수분해 효능을 증대시켰다. 3) 돈피 펩타이드의 최적 분자량 크기는 1-3 kDa이며 (CP-II), CP-II의 프롤린, 글리신, 글루타민의 총 함량이 가장 높게 나타났다. 4) 최적의 돈피가수분해물(PSH-IV) 및 펩타이드 (CP-II)는 항산화 및 항노화 활성이 매우 높게 나타났으나, 두 산물 사이의 유의적 차이는 보이지 않았다. 2. 두 번째 연구는 위에서 생성된 펩타이드와 같은 수용성 물질을 안정화하고 외부환경으로부터 보호하고자 리포좀을 이용하여 캡슐화하였다. 리포좀 제조방법은 thin film hydration과 ultrasonic emulsification기법을 사용하였으며, 레시틴의 전하 (음/양), ultrasonication 시간 및 power, 콜라겐 펩타이드 농도를 다양하게 조절하여 최적의 포물레이션을 구축하고자 한다. 또한 바이오폴리머 (키토산 또는 펙틴) 및 레시틴을 이용하여 코팅된 리포좀을 제조하였다. 생성된 리포좀은 입자특성 (입자크기, 분산도, 전하, 포집효율, 입자형태) 및 저장안정성을 평가하였다. 1) 콜라겐 펩타이드를 함유한 리포좀은 콜라겐 펩타이드 농도, ultrasonication 시간에 영향을 받으며, 리포좀 생성을 위한 최종 조건은 펩타이드 농도 3%, ultrasonication 20% power에서 5분이며, 약 70 nm의 입자크기와 60% 이상의 펩타이드 포집효율을 가지는 리포좀을 생산할 수 있었다. 2) 코팅된 리포좀은 바이오폴리머 (키토산 또는 펙틴)를 이용한 경우보다는 레시틴 (양이온 또는 음이온)을 이용하여 코팅한 경우에 더욱 작은 리포좀 입자를 생산하였다. 3) 서로 다른 온도 (25도, 37도, 60도)에서의 리포좀의 저장안정성을 살펴보면, 약 100 nm 이하로써 안정한 입자크기를 유지하였다. 3. 세 번째 연구는 지용성 물질 중 하나인 시나몬 오일을 가용화하여 실제 식품에 적용하기 위한 방법으로 나노에멀젼화 기술을 사용하였다. 본 연구의 목적은 시나몬 오일을 함유한 나노에멀젼의 최적공정을 확립하고, 실제 식품모델을 사용하여 음료(수박주스)의 유통기한을 연장하고자 하는 것이다. 생성된 시나몬 함유 나노에멀젼은 이화학적 (입자크기, 분산도, 전하, 포집효율, 시나몬 릴리즈 속도)및 미생물학적 분석(Salmonella typhimurium, Staphylococcus aureus, Escherichia coli의 저해효과)을 실시하였다. 1) 최종적으로 나노에멀젼 (< 200 nm)은 시나몬오일과 유화제 (Tween 20)를 1:3으로 혼합한 다음 고에너지 유화방법(10,000 rpm high-speed homogenization, 140 MPa high-pressure homogenization)을 통해 생산하였다. 2) 시나몬 포집효율은 70%이상으로 나타났으며, 시나몬 에멀젼 농도가 높을수록 시나몬의 릴리즈 속도가 증가하는 경향을 보였다. 3) 시나몬 에멀젼의 항균효과를 살펴보면, Salmonella typhimurium 와Staphylococcus aureus성장에서 저해효과가 나타났으며, 시나몬 에멀젼을 수박주스에 적용하였을 경우에도 유사한 효과를 보여주었다. 4. 네 번째 연구는 또 다른 지용성 물질인 피쉬오일을 가용화하고 외부로부터 안정하게 보호하기 위한 방법으로 다중층 에멀젼화 기술을 사용하였다. 피쉬오일을 함유한 다중층 에멀젼은 바이오폴리머 (키토산과 펙틴)와 함께 Layer-by-Layer electrostatic interaction 방법을 사용하여 제조되었으며, 제조된 다중층 에멀젼은 물리적 안정성 및 산화안정성을 평가하였다. 또한 햄버거 패티 (Pork patty)에 적용하여 냉장 유통기한을 연장하고자 하였다. 이들의 품질특성을 평가하기 위하여 다양한 이화학적 (pH, 색도, 보수력, 수분함량, 지방산패도)•물성학적 및 미생물학적 (총균수) 분석을 진행하였다. 1) 피쉬오일을 함유한 다중층 에멀젼의 최적조건은 1차 코팅막인 Tween 20 1.25%, 2차 코팅막인 키토산 0.1%, 3차 코팅막인 펙틴 0.2%로 산출되었다. 2) 최적 조건에서 생성된 1, 2, 3차 에멀젼은 모두 300 nm 이하의 입자크기와 좁은 입자분산도를 나타냄으로써 물리적으로 매우 안정한 상태를 유지하였다. 3) 실제 식품시스템인 햄버거 패티에 각각 최적 포물레이션인 피쉬오일 에멀젼 (1, 2, 3차 에멀젼)을 첨가하여 품질특성을 측정한 결과, 햄버거 패티의 물성학적 변화를 나타나지 않았다. 또한 코팅막의 수가 증가할수록 지방의 산패가 억제되는 경향을 보여주었으며, 총균수는 감소하는 것을 확인할 수 있었다.

In this study, solubilization methods were developed to improve bioavailability and ultimately the solubility of functional water-insoluble substances. The solubilization was demonstrated by 1) hydrolysis producing low-molecular-weight materials from high-molecular-weight materials and 2) encapsulation to emulsify and protect fat-soluble materials. As for the protein hydrolysis, in this study evaluated the ability of hydrothermal processing to produce protein hydrolysates from porcine skin. A custom-fabricated hydrothermal treatment system processed the porcine skin samples at high temperature (150–250°C) and pressure (350–3,900 kPa). The porcine skin hydrolysates (PSHs) were fractionated by ultrafiltration between molecular weight cut-offs (MWCOs) 1 and 10 kDa. The control (pretreatment porcine skin) showed free amino acid content of 2.67%, which increased to 18.77% at 250°C and 3,900 kPa, thus pointing to enhanced protein hydrolysis. During empirical model fitting, a synergistic effect of temperature and pressure on protein hydrolysis was confirmed. On protein gel electrophoresis, no obvious peptide band of >15 kDa was observed after treatment above 190°C and 1,100 kPa. According to these results, the optimal conditions for preparation of PSHs are 210°C and 2,100 kPa (hydrothermal processing, method PSH-IV), and the optimal collagen peptide isolated fraction (CP) was eventually identified: MWCO 1–3 kDa (fraction CP-II). PSH-IV and CP-II had higher antioxidant (possible antiaging) activity than the control, but there was no significant difference between PSH-IV and CP-II. This study showed the potential benefit of hydrothermal processing for hydrolysis of high-molecular-weight proteins from animal byproducts as a green and sustainable technology, in particular, PSH-IV has the potential for development into new health foods and cosmetic products. To encapsulate collagen peptides (water-soluble material), liposomes loaded with collagen peptides (L-CP) were assembled using a combination of thin film hydration and ultrasonication emulsification. The influence of a lipid charge (negatively or positively charged lipids), duration and power of ultrasonication, and collagen peptide concentration was evaluated. Layered liposomes loaded with collagen peptides (LL-CP) were prepared from a biopolymer (chitosan or low-methoxyl pectin) and charged liposomes. Droplet properties of L-CP were dependent on collagen peptide concentration and ultrasonication duration, and showed a smaller size (with the increasing ultrasonication duration) and >60% encapsulation efficiency. LL-CP with charged liposomal coating had a smaller particle size than those with a biopolymer. In addition, L-CP and LL-CP with charged liposomal coating were physically stable during storage (particle size <100 nm) regardless of storage temperature. To dissolve and encapsulate functional oils (fat-soluble material: trans-cinnamaldehyde; CIN), CIN emulsions were prepared and studied by examining physical and microbial properties. Moreover, CIN emulsions were applied to a real food system like watermelon juice. Nanosized (< 200 nm) CIN emulsions were produced by means of the optimal mass ratio of CIN and Tween®20 (TW, 1:3) and then high-energy emulsification (10,000 rpm high-speed homogenization and 140 MPa high-pressure homogenization) was carried out. CIN was encapsulated at over 70% efficiency, and its encapsulation efficiency was maintained at a higher level in 0.8 wt% CIN emulsions. As for the antibacterial activity, 0.8 wt% CIN emulsions corresponded to the lowest concentration that showed inhibition of Salmonella typhimurium and Staphylococcus aureus growth (except for Escherichia coli) in both pure water and water melon juice. As another fat-soluble material, fish oil (FO) was solubilized by emulsification, and then multilayered FO emulsions were created by the layer-by-layer electrostatic deposition method to further improve the physical or oxidation stability. To develop an FO emulsion formulation, the composition of the emulsifier and biopolymer for stable FO emulsions was determined using the modified critical micelle concentration principle. In this study, the selected concentrations of coating materials were 1.25% TW (primary layer), 0.1% chitosan (secondary layer), and 0.2% low-methoxyl pectin (tertiary layer). All FO emulsions were physically stable, resulting in small particles below 300 nm with a narrow size distribution. Furthermore, the oxidation stability of multilayered FO emulsions decreased with the decreasing number of membrane layers because FO was released from the layered emulsions. To apply these methods to a real food system, we determined the effects of a multilayered FO emulsion with or without CIN on pork patties. After this treatment, pork patties were stored for 20 days in a refrigerator (5°C) to study changes in quality. Lipid oxidation was stronger in treated pork patties than in control pork patties during the storage. In addition, lipid oxidation and total viable bacterial counts in pork patties decreased as the number of coating layers increased. Nonetheless, hardness, cohesiveness, and springiness of all samples showed no significant changes during storage as compared to fresh pork patties. According to these results, our study confirmed that FO emulsions do not affect the texture characteristics of fresh pork patties, indicating that this formulation may be used to improve the quality of pork patties by supplying high-quality fat such as unsaturated fatty acids.

In this study, the selected concentrations of coating materials were 1.25% TW (primary layer), 0.1% chitosan (secondary layer), and 0.2% low-methoxyl pectin (tertiary layer). All FO emulsions were physically stable, resulting in small particles below 300 nm with a narrow size distribution. Furthermore, the oxidation stability of multilayered FO emulsions decreased with the decreasing number of membrane layers because FO was released from the layered emulsions. To apply these methods to a real food system, we determined the effects of a multilayered FO emulsion with or without CIN on pork patties. After this treatment, pork patties were stored for 20 days in a refrigerator (5°C) to study changes in quality. Lipid oxidation was stronger in treated pork patties than in control pork patties during the storage. In addition, lipid oxidation and total viable bacterial counts in pork patties decreased as the number of coating layers increased. Nonetheless, hardness, cohesiveness, and springiness of all samples showed no significant changes during storage as compared to fresh pork patties. According to these results, our study confirmed that FO emulsions do not affect the texture characteristics of fresh pork patties, indicating that this formulation may be used to improve the quality of pork patties by supplying high-quality fat such as unsaturated fatty acids.
