Flavonoids from Chamaecrista nomame and Actinidia polygama and their free radical scavenging activities

In the search of new chemical compounds from nature with expectation of having profound biological activities, two plants namely Chamaecrista nomame and Actinidia polygama were phytochemically investigated. From their methanolic extract, 48 compounds were isolated through various types of chromatographic techniques. The chemical structures of isolated compounds were elucidated using different analytical methods like UV-VIS, Mass, IR, 1D, 2D NMR spectroscopy and polarimetry. Among isolated compounds, four compounds are new, named as isocassiaoccidentalin B (CN-8), nomaminoside (CN-22) obtained from C. nomame and 1-ketodehydroiridodial (AP-1), kaempferol 3-O-[α-L-rhamnopyranosyl (1→3)-(4-O-acetyl)-O-α-L-rhamnopyranosyl(1→6)-(2-O-acetyl)-O-β-galactopyranoside (AP-18) were isolated from A. polygama. The phytochemical groups of compounds isolated from C. nomame are flavanone glycoside (CN-1), flavanone (CN-7), phenyl propanoids (CN-2, CN-4, CN-24), phenyl propanoids glycosides (CN-17, CN-18), chalcone (CN-11), chalcone glycosides (CN-5, CN-21), anthraquinones (CN-13, CN-14, CN-15), flavonol (CN-23), isoflavone (CN-25), flavone (CN-12), and flavone glycosides (CN-3, CN-6, CN-8, CN-9, CN-10, CN-12, CN-18 ̶ CN-20, CN-22). CN-5, CN-11 were potent free radical scavenger, however compounds CN-13, CN-14, CN-15 and CN-25 expressed weak free radical scavenging activity measured by DPPH and ABTS assays. Nitric oxide release was significantly inhibited by compounds CN-12, CN-7 and CN-3 with negligible damage of cells, while compounds CN-4, CN-6, CN-8, CN- 13, CN-20, CN-22 were found inactive of nitric oxide inhibition. Compounds isolated from methanolic extract of A. polygama are iridoid (AP-1), phenolic acids (AP-2, AP-6, AP-7, AP-11, AP-12), phenyl propanoids (AP-6, AP-13, AP-22), norisoprenoid (AP-3) , coumarin (AP-4), chromene (AP-5), neolignans (AP-10, AP-19), flavanone (AP-8), flavonol (AP-9), flavone glycoside (AP-23) and flavonol glycosides (AP-14 ̶ 18, AP-20, AP-21). Compound AP-13 and AP-14 showed significant free radical scavenging activity in both DPPH and ABTS assays.
