Galla rhois is a widely used oriental medicine in East Asia for the treatment of various diseases such as diarrhea, hemorrhaging, inflammation of the intestinal mucosa, anaphylaxis, and bacterial or viral infection. However, the effect of Galla rhois on cancer metastasis and its molecular mechanisms are unclear. In this study, the anti-metastatic properties of the water extract of Galla rhois (GRWE) on metastatic colorectal cancer (CRC) cells were investigated. To confirm the effect of GRWE on the cell viability and apoptosis of CRC cells, WST-8 assay and Annexin V assay were carried out using colon 26 (CT26) cells. Western blotting and real-time RT-PCR were performed to elucidate the underlying molecular mechanisms which were involved in apoptosis and metastatic phenotypes such as epithelial-to-mesenchymal transition (EMT), respectively. The effects of GRWE on migration and invasion were examined using wound healing assay and matrigel invasion assay. The effect of GRWE on CT26 cell metastasis was further evaluated in experimental lung metastasis models in vivo. GRWE suppressed cell proliferation of CT26 cells by inducing apoptosis through caspase-3, caspase-8, caspase-9, and PARP cleavage, down-regulation of Bcl-2 and Bcl-xL, and up-regulation of Bax. In addition, the expression of EMT markers including E-, N-cadherin, vimentin, snail, slug and twist was regulated by GRWE treatment in CT26 cells. Moreover, non-cytotoxic concentrations of GRWE attenuated cell migration and invasion through inhibition of Matrix metalloproteinase (MMP)-2 and MMP-9 expression. In an expreimental lung metastasis model, GRWE significantly inhibited the lung metastasis of CT26 cells. Taken together, our study suggests that GRWE might be potential agent for colorectal metastasis.
