Bacterial infection has emerged as a serious problem in global public health. Moreover, many drug-resistant bacteria have been found with increasing the use and abuse of antibiotics. A number of advances such as improvement or change in structures of existing antibiotics are currently being made in order to get over the drug resistance. However, the issue of drug resistance has not been completely resolved. Thus, it is important to conduct the research on new antibiotics in order to get over the drug resistance. The purpose of this study is to determine antimicrobial activity and synergistic effect of SSH against MRSA with existing antibiotics such as Oxacillin and Ciprofloxacin against MRSA, and to identify the mechanism. The disc diffusion method was used with 50, 100, 200, 400 μg/ 20 μL of SSH. According to the results, the inhibition zone of diameter of 8 ~ 14 mm was observed in MRSA (ATCC 33591, CCARM 3095, DPS-1) strains. The MIC for SSH against MRSA was 125 ~ 250 μg/mL. When the SSH was combined with oxacillin or ciprofloxacin, it showed the synergy or partial synergy, respectively. When the SSH is combined with Tris, the antimicrobial activity was increased by 44 ~ 50 % because of increased membrane permeability. When the SSH is combined with NaN3, the antimicrobial activity was increased by 52~54 % because of the inhibition of ATP metabolism. Taken all together, SSH has the antimicrobial activity against MRSA. The results suggest that the SSH is the excellent antimicrobial substance in vitro when it is combined with antibiotics. We hope our data may contribute to develop new drugs to control the MRSA resistance.
