Since MCG ferments using the LAB can take advantage of both MCG and LAB, this study was examined the optimal fermentation conditions required to increase ginsenoside content as well as to increase antioxidative activity in ferments of MCG extracts using the LAB isolated from fermentation foods. Prior to the fermentation of MCG, LAB was isolated from fermented foods, and five different strains were selected by experimenting β-glucosidase activity. As the physicochemical properties and crude saponin content were examined in fermentation of MCG extracts, and the final strain was selected and identified. For the extraction condition of MCG, a central composite design was applied using extraction time and temperature as independent variables, and investigating antioxidative activity and an effective components of MCG extracts as dependent variables. In addition, to establish LAB's fermentation condition of MCG, the ginseng concentration, fermentation time, and the sub-material concentration were determined as independent variables, and physicochemical properties, antioxidative activity, and effective components of MCG ferments as dependent variables. Among the pure strains isolated from fermented foods, a strain with excellent β-glucosidase activity and MCG fermentation capacity was selected, identified, and named as Leuconostoc mesenteroides L1. Alcohol (50%) was added to ground MCG in the ratio of 10:1, and was experimented under each extraction condition. In the experiment, the optimum temperature was found to be 75-82℃, and the optimum extraction time was found to be 5.7-6.0 hrs. Extraction temperature and time were found to primarily affect MCG extraction yield and the antioxidative characteristics. The optimum extraction condition for crude saponin and ginsenoside content, was 78-81℃ for 5.5-6 hrs, which were primarily affected by extraction time. The Leuconostoc mesenteroides L1 was inoculated into the MCG extracts obtained under the optimum extraction condition of 80℃ for 6 hrs. The cultures was then fermented under each condition and the results was as followed. The optimum fermentation condition for antioxidative characteristics was MCG concentration of 6.0-6.3%, fermentation time of 60-80 hrs, and sub-material concentration of 1.3-1.5%, and antioxidative characteristics were primarily affected by MCG concentration and fermentation time. The pH, acidity, and LAB numbers of the ferments were primarily affected by sub-material concentration. The optimum fermentation condition for crude saponin and ginsenoside content were MCG concentration of 5.5-6.0%, fermentation time of 60 hrs, and sub-material concentration of 0.0-0.5%. The content were primarily affected by the concentration of MCG. With an increase in fermentation time and a higher intitial concentration of MCG concentrate, the content increased. Under the optimum fermentation condition for MCG, acidity increased approximately 6-fold, and LAB numbers increased up to 1,000-fold when compared to the initial numbers at fermentation. Furthermore, for the antioxidative characteristics, the totoal phenolic compound content was showed 1.9 times the content of MCG extracts, DPPH radical scavenging was 1.2 times, and ABTS radical scavenging was 1.3 times, with an increase particularly in total phenolic compound content. No significant change was detected in nitrite scavenging activitiy, FRAP, and reducing power, whereas pH and total flavonoid content decreased. Crude saponin content of MCG ferments increased about 2-fold when compared to that of MCG extracts.
