In order to analyze the effects of Limoninum tetragonum on melanogenesis and demonstrate its suggested mechanism of action, melanogenesis related experiments were performed in B16F10 melanoma cells. L. tetragonum forms a community in the Jeju beach and the western and southern coastal tidal flats of the upper part in Korea. Also, it is distributed along with the Zoysia sinica Hance, Suaeda japonica Suaeda maritima on the sea dyke. L. tetragonum is a type of halophyte, reported to possess biological activities such as anti-inflammatory, anti-obesity and inhibiting cancer cell growth-cancer metastasis effects. However, at present, relevant research on its anti-melanogenic effect remains insufficient. Therefore, in this study, anti-melanogenic effects of crude extract and its solvent fractions (n-hexane, 85% aq. MeOH, n-BuOH and H2O) from L. tetragonum on differentiating melanoma cells and its mechanism were evaluated. L. tetragonum was shown to inhibit melanin synthesis and tyrosinase acivity in a concentration-dependent manner. L. tetragonum inhibited the expression of MITF, tyrosinase, TRP-1, and TRP-2 in terms of mRNA and protein levels, respectively. Overall, anti-melanogenic activities by n-BuOH fraction, which was the most effective sample among tested extracts from L. tetragonum, were confirmed through an appropriate control of the inhibition of melanogenesis-related factors in the α-MSH induced B16F10 melanoma cells. Based on the results of the present study, it is suggested that L. tetragonum holds great deal of potential to be utilized as a promising source of cosmeceuticals with anti-melanogenic effect.
