SB-1493균주에 의해 생산된 SB-1493 조효소액은 AMM (alginate-minimal liquid medium) 액체배지를 이용하여 25℃에서 48시간 배양한 후 원심분리 및 여과를 통하여 제조하였다. 0.5% Na-alginate와 조효소액을 9:1로 첨가하여 40℃에서 60분간 반응하여 얻어진 가수분해물은 PTP1B 저해활성에 의해 IC50 값을 결정하였다. 0.5% Na-alginate 가수분해물의 항비만 효과(3.26±0.01 ㎍/㎖)는 대조구(6.70±0.25 ㎍/㎖)와 비교하여 약 2배 증가되었음을 확인하였다. 반면 조효소액에 의해 가수분해된 다시마 추출물의 항비만 효과(0.660±0.018 ㎍/㎖)는 다시마 추출물인 대조구(0.811±0.004 ㎍/㎖)와 비교하여 약 1.2배로 낮은 증가를 보였다. 다시마 추출물의 항비만 효과를 증진시키기 위해, 3종의 상업용 효소(Viscozyme® L, Celluclast® 1.5L, Saczyme®)와 SB-1493 조효소액을 단일 및 혼합 효소로 이용하여 6 brix 다시마 추출물을 가수분해(30∼70℃, 24시간)한 후 항비만 효과를 분석하였다. 그 결과 Celluclast® 1.5L와 SB-1493 조효소액으로 조합된 C+A 가수분해물(0.233±0.037 ㎍/㎖)의 항비만 효과는 대조구(0.811±0.004 ㎍/㎖)와 비교하여 약 3.4배 증가되었음을 확인하였다. 효소 처리된 가수분해물의 항비만 효과를 개선하기 위해 가수분해물이 첨가된 배지에 Lactobacillus brevis BJ-20을 접종하여 배양(37℃, 48 시간, 150 rpm)한 후 항비만 효과를 분석하였다. Celluclast® 1.5L에 의해 가수분해된 유산균 다시마 혼합발효물의 항비만 효과(0.990±0.046 ㎍/㎖)는 대조구인 다시마 추출물(3.24±0.243 ㎍/㎖)과 비교하여 약 3.3배 증가되었다. 또한 L. brevis BJ-20 외 9종의 유산균과 Celluclast® 1.5L 처리 다시마 추출물과 혼합발효를 수행하여 항비만 효과를 비교한 결과, Bifidobacterium longum, L. plantarum 및 L. sakei의 IC50값은 0.720±0.021 ㎍/㎖, 0.827±0.007 ㎍/㎖, 0.855±0.026 ㎍/㎖로 나타나 L. brevis 보다 개선되었음을 확인하였다. 3종 유산균의 다시마 혼합발효물은 이용한 항산화, bifidogenic growth 효과 및 항고혈압 효과를 분석한 결과, B. longum 혼합발효물은 상대적으로 높은 항산화와 bifidogenic growth 효과를 보였다. 또한 용매계통 추출을 실시한 결과, Celluclast® 1.5L 처리 다시마 추출물과 비교시 B. longum에 의한 다시마 혼합발효물은 ethyl acetate 층은 감소한 반면 dichloromethane과 butanol 분획 층에서 항비만 효과가 증가하였음을 확인하였다. 상기 결과로부터, Celluclast® 1.5L과 SB-1493 조효소액을 이용한 효소가수분해로부터 다시마 추출물의 항비만 효과를 개선을 확인하였으며 유산균을 이용한 혼합발효를 통하여 항비만 효과 외에 항산화, bifidogenic growth 효과 등 추가적인 기능성을 부여할 수 있음을 확인하였다.

The purpose of this study was to enhance the anti-obesity effect of sea tangle extracts, through techniques based on enzymatic hydrolysis and the addition of mixed fermentative lactic acid bacteria. To hydrolyze alginate, the main component of sea tangle, we assessed the activation of enzymes in 23 bacterial strains involved in alginate decomposition, and selected SB-1493 as an exceptional strain for our purpose. The crude enzyme solution produced by the bacterial strain SB-1493 was cultivated for 48 hours at 25°C using alginate-minimal liquid medium (AMM), and was then centrifuged and filtered to obtain the final product. For the hydrolysate produced from the reaction of 0.5% Na-alginate and the crude enzyme solution (9:1 ratio) for 60 minutes at 40°C, we determined the half-maximal inhibitory concentration (IC50) value based on the inhibitory activity of protein tyrosine phosphatase 1B (PTP1B). The anti-obesity effect of 0.5% Na-alginate hydrolysate (3.26±0.01 ㎍/㎖) was increased by two-fold compared to the control (6.70±0.25 ㎍/㎖). By contrast, the anti-obesity effect of seatangle extract hydrolyzed by the crude enzyme solution (0.660 ± 0.018 ㎍/㎖) was only increased by 1.2-fold compared to the control sea tangle extract (0.811±0.004 ㎍/㎖). To enhance the anti-obesity effect of sea tangle extract, we combined 3 commercial enzymes (Viscozyme® L, Celluclast® 1.5L, and Saczyme®) and the crude enzyme solution as single and mixed enzymes, hydrolyzed 6 brix sea tangle extracts (30–70°C, 24 hours), and analyzed the anti-obesity effect. The results indicated that the anti-obesity effect of the hydrolysate from the combination of Celluclast® 1.5L and crude enzyme solution (C+A hydrolysate; 0.233±0.037 ㎍/㎖) was increased by 3.4-fold compared to the control (0.811±0.004 ㎍/㎖). To enhance the anti-obesity effect of the enzyme-treated sea tangle hydrolysate, we cultivated Lactobacillus brevis BJ-20 (37°C, 48 hours, 150 rpm) in medium containing the hydrolysate, and then analyzed the anti-obesity effect. The anti-obesity effect of the sea tangle mixed fermentative product hydrolyzed by Celluclast® 1.5L (0.990±0.046 ㎍/㎖) was 3.3-fold higher than the control sea tangle extract (3.24±0.243 ㎍/㎖). Moreover, the sea tangle mixed fermentative product was obtained using 9 other bacteria (excluding L. brevis BJ-20), the sea tangle extracts were treated with Celluclast® 1.5L, and their anti-obesity effects were compared. The IC50 values of sea tangle mixed fermentive product with Bifidobacterium longum, Lactobacillus plantarum, and Lactobacillus sakei were 0.720±0.021 ㎍/㎖, 0.827±0.007 ㎍/㎖, and 0.855±0.026 ㎍/㎖, respectively, indicating greater enhancement compared to the L. brevis. Analysis of the antioxidative effect, influence on bifidogenic growth, and anti-hypertensive effect of the sea tangle mixed fermentative products produced by 3 different bacterial strains showed that the sea tangle mixed fermentative product from B. longum had a relatively enhanced anti-oxidative effect and effect on bifidogenic growth. Furthermore, solvent extraction showed that the sea tangle mixed fermentative product had a strong anti-obesity effect in the dichloromethane and butanol layers compared to the sea tangle extract treated with Celluclast® 1.5L. From the above results, we validated that enzymatic hydrolysis using Celluclast® 1.5L and crude enzyme solution was able to enhance the anti-obesity effect of sea tangle extract. Furthermore, we validated that the sea tangle mixed fermentative product obtained using lactic acid bacteria can provide additional beneficial effects, including an antioxidative effect and influence on bifidogenic growth.
