다시마는 7개의 당분해 효소와 5개의 단백질 분해효소를 사용하여 가수분해한 추출물들의 항산화활성을 전자스핀 공명기기를 이용하여 free radical 에 대한 소거 활성을 측정하였다.
다시마를 당분해 효소와 단백질 분해효소로 가수분해시킨 추출물에서 AMG로 가수분해시킨 추출물에서 가장 높은 hydroxyl 라디칼 소거활성을 나타내었으며, 특히 AMG로 가수분해시킨 추출물 2.0 mg/ml의 농도에서 87.6%로 높은 hydroxyl 라디칼 소거활성을 나타내었다
반면에 다시마를 다양한 단백질 및 당 가수분해효소들로 가수분해시킨 추출물들의 alkyl라디칼 및 superoxide 라디칼 소거활성에서는 DPPH 및 hydroxyl 라디칼 소거활성보다 전반적으로 낮은 활성을 나타내었다.
추가적으로, 다시마 효소적 추출물의 항산화 효과를 in vitro에서 hydroxyl 라디칼로 유발된 pBR322 DNA 손상에 대한 방어 여부를 이용하여 측정하였다.
트립신 추출물에서 H2O2로 유발된 DNA 손상에 대한 방어 효과를 보여주었으나 AMG 추출물에서는 어떠한 방어효과도 나타내지 않았다.
Trypsin 및 AMG 추출물을 이용하여 H2O2로 유발된 세포사멸에 대한 방어효과를 PC-12 셀 라인을 이용하여 측정하고, flow cytometer로 세포주기를 분석하였다.
AMG 및 trypsin 추출물은 PC-12 셀 라인과 대조하여 H2O2에 의해서 유발된 세포사멸을 보호하는 것으로 관측되었다.

Laminaria japonica was enzymatically hydrolyzed by seven carbohydrases (dextrozyme, AMG, promozyme, maltogenase, termamyl, viscozyme and celluclast) and five proteinases (flavourzyme, neutrase, protamex, alcalase and trypsin), and their free radical scavenging activities were evaluated on DPPH, hydroxyl, superoxide and alkyl radical by using an electron spin resonance (ESR) spectrometer. All hydrolysates showed DPPH radical scavenging activities, and hydroxyl radical scavenging activities of AMG and trypsin extracts from Laminaria japonica were the highest as comparison to those of the other carbohydrases and proteinases extracts, and their activities were 87.6% and 67.5% at 2.0 mg/mL, respectively. However, superoxide and alkyl radical scavenging activities of enzymatic hydrolysates were lower than DPPH and hydroxyl radical.
In addition, the ability of extracts (trypsin and AMG) to inhibit the oxidative damage of DNA was assessed, in vitro, by measuring the conversion of supercoiled pBR322 plasmid DNA to the open circular form. Trypsin extracts of Laminaria japonica showed protective effect on H2O2-induced DNA damage but not AMG extract. Additionally, the protection effect on hydroxyl radical-induced oxidative damage in PC-12 cells via propidium iodide staining using a flow cytometer were investigated. AMG and trypsin extracts of Laminaria japonica decreased cell death in PC-12 cells against hydroxyl radical-induced oxidative damage in a dose dependant manner. These results indicate that enzymatic extracts of Laminaria japonica antioxidant activity.
