The strip consists of four main components: sample pad, conjugate pad, membrane, and absorbent pad. The nitrocellulose membrane is separately coated with goat anti-rabbit antibodies (control line) and monoclonal anti-pepsin antibodies (test line). When the pepsin-containing samples are applied to the sample pad, pepsin initially reacts with polyclonal antibody-coated gold nanoparticles, and then reacts with the fixed monoclonal antibody on the membrane. These reactions result in the red change at the test line, whose intensity is proportional to the pepsin concentration. The design parameters examined in this work include the antibody amount loaded to the membrane for immunochromatographic action, and incubation time. To finally, confirm the analytical usefulness of the LPR immunostrip sensor, the sensor was tested for artificial and human (patient and non-patient) saliva samples.
