Herpes simplex virus type-1 is mainly associated with oral and facial infections, and establishes latent infections. In this study, we first investigated the molecular mechanism that links anti-viral and anti-inflammatory activities of quercetin using in vitro HSV-1 infection system. Raw 264.7 cells were infected with HSV-1 in presence or absence of various concentrations of quercetin and infected cell lysates were harvested after 24h. HSV plaque reduction assay, western blot (HSV-1gD, HSV-1 ICP0), and real time PCR analysis (HSV-1ICP0, HSV-1UL13, HSV-1UL52) were performed to confirm anti-HSV-1 effect of quercetin. Also, to explore anti-inflammatory mechanism of quercetin, western blot analysis (TLR-2, 3, 9, NF-kB, IRF3) was performed. In addition, the production of pro-inflammatory cytokine (TNF-α) was measured. Quercetin significantly inhibited HSV infectivity in Raw 264.7 cells and suppressed the expression of HSV proteins (gD, ICP0), which correlates with reduced HSV gene expressions (ICP0, UL13, UL52). Especially, quercetin downregulated the expression of TLR-3, as a result, downstream transcriptional factors including NF-kB and IRF3 activations were blocked. We suggest that anti-HSV-1 effect of quercetin is TLR-3-dependent in Raw 264.7 cells and expect quercetin might be potential therapeutic agent for treating HSV-1 infection. Next, we evaluated the anti-cancer effect of quercetin and isoliquiritigenin against EBV(+) human gastric carcinoma (SNU719) or EBV(-) human gastric carcinoma (MKN74) in vivo. Approximately 10% of gastric carcinoma are Epstein-Barr virus associated gastric carcinoma (EBVaGC). First, the tumor-bearing mouse model was established by subcutaneously inoculating suspension of SNU719 cells or MKN74 cells (5×106 cells in 0.1 ml physiological saline) into the dorsum next to the right hind leg of mice. Quercetin and isoliquiritigenin were orally administrated for 2 weeks, and tumor size was measured every other day using a standard caliper. The results showed that anti-cancer effect of quercetin was greater than isoliquiritigenin in mice injected with EBV(+) human gastric carcinoma (SNU719) cells. On the other hand, quercetin and isoliquiritigenin had similar anti-cancer effects in mice injected with EBV(-) human gastric carcinoma (MKN74) cells. Subsequently, we explored the molecular mechanisms responsible for their anti-cancer activities using tumor tissues derived from xenograft animals. Quercetin more effectively induced p53-dependent apoptosis than isoliquiritigenin in EBV(+) human gastric carcinoma. And quercetin induced the increased expressions of p21, PUMA, Bax, cleaved forms of caspase-3, -9, and Parp. In EBV(-) human gastric carcinoma, isoliquiritigenin as well as quercetin markedly increased p53, PUMA, Bax, and the cleaved forms of caspase-3, -9, and Parp. We conclude that quercetin is a good candidate for anti-cancer agent in EBV-associated human gastric carcinoma.
