최근 기초 과학, 의료 과학 및 기술의 발전은 중간엽 줄기세포를 이용하여 다양한 질환 치료를 가능하게 하였다. 환자의 안전과 복지에 있어 줄기세포 치료제의 품질 제어의 필요성에 대한 인식이 증가하고 있다. 따라서 줄기세포 치료제 제조 공정 개발을 통해 안전성 확보와 치료 효과의 최적화가 필요하다. 본 연구에서는 효과적인 줄기세포 치료를 위해 줄기세포 치료제의 제조 공정의 각 단계에 적용 할 수 있는 탯줄유래 생체 재료와 줄기세포의 활용 방안을 제시 하고자 하였다.
Part II는 줄기세포를 안전하고 효과적인 세포치료제로 활용하기 위한 방안을 마련하기 위해, 탯줄에서 콜라겐과 탯줄추출물을 분리하여 코팅 물질과 우태아 혈청 대체체로서의 활용 가능성을 기술하였다. 탯줄 추출물로 배양한 결과, 우태아 혈청을 포함하는 배지로 배양한 세포와 유사하게 세포 성장 능력과 줄기세포의 특성이 유지되는 것을 확인하였다. 결론적으로 의료용 폐기물인 탯줄로부터 줄기세포치료제의 자원 확보와 지방유래 줄기세포, 골수 유래 줄기세포, 섬유아세포를 포함하는 다양한 세포를 배양 할 수 있는 무이종성 배양 시스템을 확보할 수 있는 가능성을 확인하였다.
Part III는 줄기세포의 동결 보존을 위해 저장 단백질 2를 이용하여 저농도 DMSO와 무혈청 조건의 동결 보존액으로서의 활용 가능성으로 기술하였다. 5 mg/mL의 저장 단백질 2와 4% DMSO를 포함하는 동결보존 배지와 우태야 혈청을 포함하는 동결보존 배지를 이용한 줄기세포를 동결 보존 후 해동 하여 줄기세포의 특성을 비교하였다. 두 조건 모두에서 1년 동안 동결 보존 후에도 세포의 생존율이 유지됨을 확인하였다. 또한, 지방세포, 조골세포로의 분화능을 확인한 결과, 분화능이 유지됨을 확인하였다. 게다가, 지방유래 중간엽 줄기세포, 골수유래 중간엽 줄기세포 등의 다양한 중간엽 줄기세포의 동결 보존에도 적용할 수 있음을 확인하였다. 결과적으로 저장 단백질 2는 저농도의 DMSO와 무혈청 동결 보존제로서 사용할 수 있는 가능성을 확인하였다.

Recent advance in basic and medical science and technologies has realized the employment of mesenchymal stem cells (MSCs) for a variety of therapeutic indications including regenerative therapies. Stem cell therapy is a growing awareness of the need for enhanced quality control for these cells, with patient safety and well-being as the ultimate goal. Therefore, it is necessary to optimize therapeutic efficacy and to ensure safety over the cell manufacturing process. This study attempts approach for effective stem cell therapy using umbilical cord-derived materials and stem cells that are applicable for cell therapy manufacturing process.
Part II describes possibility of human umbilical cord extract (UCE) serving as a serum replacement and collagen purified from umbilical cord (UC-collagen) acting as an extracellulear matrix (ECM) for in vitro culture of MSCs derived from human umbilical cord (UC-MSCs). Importantly, the proliferation and stemness of the UC-MSCs cultured with the UCE media were similar to those cultured under fetal bovine serum (FBS) conditions on UC-collagen-treated plates for eight passages. Based on these results, I suggest that umbilical cords discarded as medical waste represent a viable alternative source of xeno-free biomaterials to replace animal-derived serum and ECM materials for the cultivation of various cell types, including UC-MSCs, adipose tissue-derived MSCs, bone marrow-derived MSCs and fibroblasts.
Part III describes storage protein 2 (SP2) as an alternative to establish an effective, low-dimethyl sulfoxide (DMSO) and FBS-free cryopreservation agent for the cryostorage of hMSCs. When the cell characteristics (cell viability and stemness) were analyzed after thawing, those obtained using 5 mg/mL SP2 were comparable to those obtained using a freezing medium with FBS. The stable cell viability and characteristics were shown even after 1 year cryopreservation. In addition, when the cells were differentiated into adipocytes and osteocytes, I confirmed that the differentiation behaviors of the thawed cells were well maintained. The positive results could be also obtained when SP2 were applied to other MSCs. The results clearly indicate that SP2 could be used as an alternative to FBS for a freezing medium with reduced DMSO.
Part IV describes the injectable hydrogel scaffolds that promote the regeneration of damaged or degenerative cartilages via arthroscopy. The injectable hydrogel, which is composed of cross-linked hyaluronic acid and UC-collagen, was constructed in different ratios, and the physical (adhesive strength and swelling/shrinking) and biological (degree of degradation and cytotoxicity) properties were evaluated to determine the effective ratio. Furthermore, UC-MSCs with hydrogel altered the secretion pattern of cytokines, which promote the migration of bone marrow-mesenchymal stem cells and demote the catabolic and inflammatory mechanisms of cartilage. Ten weeks after the transplantation, histological analysis clearly demonstrated significant cartilage regeneration without inflammation on the damaged site compared with the untreated group, in vivo. The results clearly indicate that hydrogel is an effective bioscaffold that can be used to treat degenerative cartilage via arthroscopic procedures.
