Here, the role of RHEB (Ras homolog enriched in brain) was investigated in tissue regeneration via modulating signaling mechanisms in various kinds of human cells including chondrocytes, mesenchymal stem cells, and cancer cells. In the first part of the study, the regulation of senescence associated signaling mechanisms in chondrocytes for cartilage regeneration has been reviewed. Adult articular chondrocytes undergo slow senescence and dedifferentiation during in vitro expansion, restricting successful cartilage regeneration. Rapid senescence or dedifferentiation not only results in the loss of the chondrocytic phenotype but also enhances production of inflammatory mediators and matrix-degrading enzymes. Upregulation of the genes interleukin 1 beta (IL-1β), p53, p16, p21, and p38 mitogen-activated protein kinase (MAPK) is responsible for the direct induction of senescence, whereas downregulation of the genes transforming growth factor-beta (TGF-β), bone morphogenetic protein-2 (BMP-2), SRY (sex determining region Y)-box 9 (SOX9), and insulin-like growth factor-1 (IGF-1), indirectly induces senescence. In the second part, the mechanistic underpinning of the RHEB gene was investigated in the control of senescence and dedifferentiation through the modulation of oxidative stress in the chondrocytes, a hallmark of osteoarthritis. Serial expansion of human chondrocytes led to senescence, dedifferentiation, and oxidative stress. RHEB maintained innate characteristics of chondrocytes by regulating senescence, dedifferentiation, and oxidative stress, increased the expression of COL2 via SOX9, and down-regulated the expression of p27 via MCL1. RHEB also decreased the expression of COL10. RHEB knockdown mimics in decreased expression of SOX9, COL2, and MCL1 while abrogating the suppressive function of RHEB on p27 and COL10 in chondrocytes. RHEB-overexpressing chondrocytes successfully formed cartilage tissue in vitro as well as in vivo with increased expression of GAG matrix and chondrogenic markers. RHEB induces distinct gene expression signature that maintained the innate chondrogenic properties for a long period of time. In the third part, role of RHEB was investigated in the differentiation of human adipose derived stem cells (ASCs) into osteogenic, chondrogenic and adipogenic lineage. RHEB play crucial role in the differentiation of ASCs while promoting osteogenic and chondrogenic differentiation and impairing the adipogenesis. Results findings implicate that osteogenesis and chondrogenesis is critically dependent on RUNX2 and SOX9. In contrast, decreases in differentiation of ASCs into adipocytes was due to downregulation of C/EBPβ. RHEB may be working as a molecular rheostat between the RUNX2 and C/EBPβ to fine-tune the balance between osteogenesis and adipogenesis. In the fourth part, RHEB role was investigated in 293T cancer cell line. RHEB play crucial role in morphological transition of 293T cells from epithelial to mesenchymal phenotype. RHEB depressed the E.Cadherin expression while increased the N.Cadherin expression. Further, RHEB expressing 293T cells showed more potential in wound healing. Hydrogels fabricated with environmentally sensitive polymers can impart them extra property like thermo responsiveness that exhibit phase transition from gel to solution and vice versa upon changes in the temperature. The property of thermo-responsiveness makes hydrogels interesting for drug delivery, gene therapy, cell culture and tissue engineering approaches and therefore, considered to be a first biomaterial have been developed for the use of humans. This review focuses on the mechanism of phase transition in thermo responsive hydrogels, the factors affecting the behavior of gel and in particular the role of phase transition of PNIPAM in drug delivery and tissue engineering.
