DNA 메틸화(DNA methylation)는 주로 유전자의 발현을 억제하며, 히스톤 변형(Histone modification)은 히스톤 단백질의 N잔기(N-terminal)에 메틸화(methylation), 아세틸화(acetylation), 인산화(phosphorylation), 수모화(sumoylation)등과 같은 변형이 일어나 유전자의 발현을 조절한다. 이러한 번역 후 변형(post-translational modification)은 히스톤(histone)뿐만 아니라 비히스톤(non-histone)단백질에도 일어나 유전자 발현(gene expression)을 조절하거나 활성(activity)을 조절한다. 또한 다양한 후성유전학 조절자들(epigenetic modifiers)이 존재하며 유전자 발현과 활성을 조절 함으로써 여러 질병모델과 생물학적 기능에 중요한 역할을 하고 있다는 보고가 많다. 그 중 최근 들어 밝혀진 메틸기를 제거해 주는 역할을 하는 히스톤 탈메틸화 효소들(histone demethylases)은 작용기전에 따라 크게 두 그룹으로 나뉜다. 본 연구에서는 히스톤 탈메틸화 효소들의 각 그룹에 속해 있는 주몬지(JmjC) 도메인을 갖는 히스톤 탈메틸화효소4A(Histone demethylase 4A, KDM4A)와 라이신 탈메틸화효소(Lysine demethylases 1, LSD1)의 암 후성유전학(cancer epigenetics)에서의 역할과 분자조절 기전을 밝히고자 하였다. 첫 번째로 탈메틸화효소4A(KDM4A)의 수모화(sumoylation)에 의한 번역 후 변형(post-translational modification)이 일어나는지 확인하고 수모화에 의한 KDM4A의 기능들과의 상호작용에 대해 연구하였다. KDM4A가 SUMO1 단백질에 의해 수모화에 의한 번역 후 변형이 일어나며 KDM4A의 라이신 471번째에 수모화가 일어나는 것을 확인하였다. 또한, KDM4A의 수모화는 KDM4A의 세포 내 위치(subcellular localization), 히스톤 탈메틸화 효소 활성(histone demethylase activity) 그리고 KDM4A의 안정성(stability)에는 영향이 없는 것을 확인하였다. 그런데 KDM4A의 수모화는 암 억제 유전자(tumor suppressor gene) p53과의 결합에 필요하다는 것을 알아냈다. 그러므로 이러한 결과는 암 후성유전학에서의 KDM4A의 수모화에 의한 새로운 기전을 제시해 줄 것으로 생각된다. 두 번째로, 유방암과의 관련성이 매우 높다는 보고는 되어있었으나 분자 조절기전은 알려진 바가 없었던 S100A7의 단백질이 탈메틸화효소1(LSD1)에 의해 조절되는지 연구하였다. 먼저, 유방암 세포에서 에스트로겐을 장시간 처리하였을 때 S100A7의 mRNA, 단백질(protein)발현이 높아지는 것을 확인을 하였다. LSD1의 억제제(inhibitor) 또는 넉 다운(knockdown)으로 LSD1의 발현 양을 감소시켜 주었을 때 앞선 에스트로겐에 의해 높아졌던 S100A7의 발현이 매우 감소하는 것을 알았다. 또한 이러한 S100A7의 발현이 LSD1의 탈메틸화 기능이 없는 돌연변이에 의해서는 S100A7의 발현에 영향이 없는 것을 확인하였으며 CHIP 실험을 통해 S100A7의 프로모터 부분에 LSD1이 결합되어있는 것을 확인하였다. 게다가 S100A7 넉다운으로 인한 S100A7의 발현 양을 감소시켜 주었을 때 에스트로겐에 의한 유방암 세포의 성장이 증가하는 것을 확인할 수 있었다. 앞선 결과들을 종합해보면 히스톤 탈메틸화효소 LSD1에 의한 S100A7의 발현 조절은 유방암 세포의 성장을 감소시켜주는데 역할을 할 것이라고 생각되며 유방암에서 S100A7은 암억제유전자로서의 역할을 하고 있을 것이라 예상할 수 있다.

Recently, epigenetic modifiers, two classes of Histone lysine demethylase (KDM) have been reported: the amine-oxidase type lysine-specific demethylases (LSD) and the JumonjiC (JMJC) domain-containing histone demethylases. This work aimed to define the role and molecular mechanisms of the human histone lysine demethylases (LSD1 and KDM4A) in cancer epigenetics. First of all, histone demethylase 4A (KDM4A, known as JmjD2A) has diverse functions such as gene regulations, development, cell cycle and plays an oncogenic role in various cancer cells. The activities of KDM4A are modulated by post-translational modifications, such as ubiquitination. Here, I demonstrated that post-translational modification of KDM4A with SUMO1 (small ubiquitin-related modifier 1) is a novel regulatory switch for the tumor suppressor p53. I found that KDM4A is SUMOlated at lysine residue 471. Furthermore, the SUMOylated KDM4A does not alter its own subcellular localization, histone demethylase activity and its protein stability. Surprisingly, co-immunoprecipitation data revealed that the SUMOylation of KDM4A at lysine 471 allows binding of KDM4A to p53. Therefore, these observations may provide a novel insight into understanding of SUMOylation-mediated control of KDM4A function. Secondly, S100A7, a member of S100 calcium binding protein family, is highly associated with breast cancer. However, the molecular mechanism of S100A7 regulation remains unclear. Here I show that long-term treatment with estradiol stimulated S100A7 expression in MCF7 breast cancer cells at both the transcriptional and translational levels. Both treatment with a histone demethylase LSD1 inhibitor and shRNA-based knockdown of LSD1 expression significantly decreased 17b-estradiol (E2)-induced S100A7 expression. These reduced E2-mediated S100A7 expression are rescued by the overexpressed wild-type LSD1 but not by its catalytically inactive mutant. My data showed in vivo association of LSD1 with S100A7 promoters, confirming the potential role of LSD1 in regulating S100A7 expression. S100A7 knockdown increased both normal cell growth and estrogen-induced cell proliferation, suggesting a negative influence by S100A7 on the growth of cancer cells. Together, my data suggest that estrogen-induced S100A7 expression mediated by the histone demethylase LSD1 may downregulate breast cancer cell proliferation, implying a potential tumor suppressor-like function for S100A7.
