This process is called photoaging, which is an important cause of skin cell immunosenescence. Although many studies on skin aging have been carried out to date, there are insufficient measures for skin aging in vivo due to lack of research on skin immunity deterioration and mechanism due to aging. The skin cell senescence model is known to be very useful for studies on damage accumulation and growth retardation due to cell growth arrest. It had been reported that continuously passaged (replicative senescence-induced) senescent cells, stress induced aging cells, and Hutchinson–Gilford progeria syndrome (HGPS) fibroblast cells were suitable as models of skin cell immunosenescence. The purpose of this study is to investigate the main factors of skin immune regulation using various skin cell senescence models and to provide skin immunosenescence biomarker. The above-mentioned cells were used to compare cell responses to UVA and UVB. First, QuantSeq 3'mRNA sequencing was performed to evaluate the differential gene expression (DEGs) of UV-irradiated fibroblasts. Senescent cells secreted higher level of matrix metalloproteinases (MMPs) and inflammatory chemokine IL-8. It was confirmed that the toll-like receptor pathway genes, such as TLR4, MyD88, and CXCL8 (IL-8) were highly expressed in senescent cells as compared with normal cells. Therefore, these data suggest that increased expression of toll-like-receptor (TLR) pathway genes may contribute to upregulation of MMP and IL-8, which promotes skin senescence. HGPS is a rare genetic disease, which causes accelerated aging due to the mutation of Lamin A (LMNA) gene and skin fibroblasts from HGPS patients show distinct aging cellular phenotypes. Metformin, a drug used in the treatment of type 2diabetes similar to rapamycin's ability to affect aging improvement through inhibition of mTOR, has been investigated to improve the aging phenotype. The data show that metformin treatment can has been shown to reduce reactive oxygen species (ROS) and to partially regain abnormally nuclear phenotype and delay senescence in HGPS fibroblasts. Collectively, this study characterized cellular responses of the non-senescent cells and senescent cells to UV irradiation, and provided a transcriptional analysis of the DEGs involved in a specific pathway to UV. In addition, it shows that metformin, a diabetic drug, restores nuclear defects and alleviates the senescence phenotype in the skin cells of HGPS patients. These findings provide a framework for studying the basic molecular mechanisms of skin immunosenescence.
