본 연구에서는 혼합균주를 접종처리하여 아로니아 (Aronia melanocarpa) 특유의 떫은맛을 저감화시키고, 식품가공과정에서 부산물로 폐기되는 아로니아 박으로부터 펙틴을 추출하여 구조적 특성 및 유화적 특성을 분석하고자 하였다. 아로니아는 3가지의 균주 (Lactobacillus bulgaricus, Streptococcus thermophiles, Saccharomyces cerevisiae)를 혼합하여 접종하였고, 이러한 혼합균주의 비율을 각각 0.00005% (w/w), 0.00010% (w/w), 0.00015% (w/w)로 달리하여 접종하지않은 대조군을 포함한 총 4가지의 시료 (NA, AI05, AI10, AI15)로 제조하였다. 화학적 특성 실험결과, 혼합균주의 접종처리 이후 생성된 부산물인 아세트알데히드로 인하여 당도 (total soluble solid)가 감소하였음을 알 수 있었고, 증가된 유기산 함량으로 pH 또한 감소한 것을 확인할 수 있었다. 총 폴리페놀 함량 (total phenolic content), 총 플라보노이드 함량 (total flavonoid content), 총 안토시아닌 함량 (total anthocyanin content)의 수치도 감소하였는데, 이는 혼합균주로 인하여 생성된 아세트알데히드 및 β-glycosidase의 영향인 것으로 추측된다. 한편 아로니아의 떫은맛을 일으키는 주요 성분인 탄닌 함량의 변화를 관찰하기 위하여, bovine serum albumin (BSA) 침전능 분석, 축합형 탄닌 분석, 아세트알데히드 분석을 실시하였다. 아로니아에 함유된 탄닌의 BSA 침전능을 분석한 결과, 혼합균주를 접종하지않은 대조군의 경우 0.13 mg TAE/mL이었으나 접종시킨 시료군 전체에서 그보다 낮은 0.11 mg TAE/mL의 동일한 값을 나타내었다. 이는 접종처리 이후 아로니아의 탄닌 함량이 감소하여 단백질과 결합할 수 있는 탄닌 함량 또한 감소한 것으로 사료된다. 실제적으로 식품의 탄닌은 입 안의 침 단백질과 결합하여 축합반응을 일으키고 그 반응물이 침전되어 떫은맛을 유발하기 때문에, 본 연구에서 확인된 탄닌의 단백질 침전능 감소는 결과적으로 혼합균주의 접종을 통하여 아로니아의 떫은맛을 저감화시킬 수 있는 것으로 사료된다. 접종처리방법을 통한 탄닌 함량의 감소는 처리과정 중의 중간산물로 생성된 아세트알데히드가 아로니아 내의 탄닌과 결합하여 변성 탄닌을 형성함으로써, 그 결과 단백질과 결합할 수 있는 유리 탄닌이 감소하였기 때문인 것으로 사료된다. 본 연구에서는 이를 입증하기 위하여 축합형 탄닌 분석과 아세트알데히드 분석을 실시하였다. 실험 결과, 혼합균주의 농도가 증가할수록 축합형 탄닌 함량이 대조군 (30.04 mg CE/mL)보다 접종시킨 시료군에서 유의적으로 낮은 수치인 26.60 mg CE/mL (AI05), 25.49 mg CE/mL (AI10), 25.04 mg CE/mL (AI15)인 것으로 나타났으며, 아세트알데히드 분석에서는 대조군 (2.62 mg/mL)에 비하여 접종시킨 시료군에서 보다 높은 수치(2.76, 2.78, 2.90 mg/mL)인 것으로 확인되었다. 이러한 결과는 BSA 침전능의 경향과 일치하고 있었으며, 이러한 경향성은 관능적 품질특성 평가에서도 동일하게 나타났다. 50명의 패널을 대상으로 ‘전반적 기호도, 떫은맛, 쓴맛, 단맛, 신맛, 베리향미’의 총 5가지 평가항목에 대하여 소비자 평가를 실시한 결과, 대조군의 경우, 5가지 항목 모두에서 가장 낮은 기호도를 보이고 있었다. 순위법 (Rankingdescriptive analysis) 관능평가는 ‘떫은맛, 쓴맛, 단맛, 신맛’의 평가항목에 대하여 수 주간 훈련된 전문패널을 대상으로 실시하였으며, 떫은맛 평가항목 결과에 따르면 혼합균주의 비율이 증가할수록 유의적으로 아로니아의 떫은맛이 감소하는 것으로 조사되었다. 한편 본 연구에서는 아로니아 박으로부터 추출된 펙틴 (aronia press cake pectin; APP)의 구조를 밝히기 위하여, 총당 함량, galacturonic acid (GalA) 함량, degree of esterification, 단당류 조성, fourier transform infrared (FT-IR) spectroscopy, X-ray diffraction (XRD), 1D Nuclear magnetic resonance (NMR) spectroscopy (1H NMR and 13C NMR), 분자량 분포 분석을 실시하였다. FT-IR, XRD, 분자량 분포 분석은 보다 정확하고 신뢰성 있는 분석을 위하여 펙틴 표준물질 (pectin standard; PS)과 비교하여 측정하였다. XRD 기기분석 결과, APP는 펙틴물질에서 특징적으로 발견되는 peak (2θ = 20.9 ˚)를 나타내었으며, FT-IR 결과 (1735-1750 cm-1, 1600-1650 cm-1)와 NMR 결과 (1H NMR: 5.05, 4.93, 5.19 ppm; 13C NMR: 98.05, 100.38, 95.86 ppm)를 통하여 APP가 α-1, 4 결합으로 연결된 D-갈락투론산을 주쇄로 하며 α-1, 2, 4 결합으로 된 L-람노오스와 D-자일로오스를 잔기로 가진 고메톡실 펙틴인 것으로 밝혀졌다. 또한 분자량 분포 분석 결과에서는, APP (Mw = 1335 × 1010 kDa)가 PS (Mw = 26 × 1010 kDa)에 비하여 높은 평균 분자량을 가진 것으로 측정되었다. 또한 APP의 유화적 특성 분석을 위하여 조건을 달리하여 제조한 3가지의 시료를 비교하였고, 준비된 시료는 각각 유화제가 미첨가된 대조군, APP를 유화제로 사용한 유화액, PS를 유화제로 사용한 유화액이었다. 실험 결과, APP가 PS에 비하여 유의적으로 높은 수준의 유화능 (59.23%)과 유화안정성 (54.66%)을 보이고 있었다. 이는 현미경 분석과 입도 분석의 결과에서 확인된 바와 같이, APP가 다른 시료에 비하여 유의적 수준에서 유화액의 droplet size를 확연하게 감소시켰기 때문인 것으로 사료된다. 이러한 droplet size의 효과적인 감소는 APP에 다량으로 함유된 단백질 함량 (7.41%), 높은 수준의 평균 분자량과 점도에 기인한 것으로 사료된다. 기존 연구에 따르면 단백질이 가진 소수성 특성이 유화액의 품질에 크게 영향을 끼치므로, 우수한 유화제의 조건 중 단백질 함량은 가장 중요한 요소로 간주되고있다. 일반성분 실험에서도 확인된 바와 같이, atomic force microscopy (AFM) 분석에서도 동일하게 APP에서 더 높은 단백질 함량을 보이고 있는 것으로 밝혀졌다. AFM 실험결과, PS와 달리 APP에서 응집된 구조가 상대적으로 다수 발견되었는데, 이러한 구조는 AFM과 관련된 기존의 연구결과에 따르면 펙틴의 단백질 구조로 인식되는 것이었다. 그 외에 분자량 분포의 결과로도 APP의 효과적인 유화제로서의 기능을 설명할 수 있었다. APP의 높은 분자량 수치는 APP가 PS보다 군집된 구조를 이루고 있다는 것을 나타내며, 이러한 구조로 인하여 유화액의 구조적 안정화를 가져올 수 있기 때문이다. 또한 레올로지 연구 결과, APP가 첨가된 유화액이 PS가 첨가된 유화액보다 더 높은 G'과 G"값을 나타내었는데, 이는 유화액의 오일층과 수용액층 사이에 존재하는 표면에 높은 점탄성을 가진 APP가 PS보다 더 두꺼운 막을 형성하여 droplet의 응집을 방해하기 때문인 것으로 사료된다. 그러므로, 본 연구에서 조사된 결과에 의하면, 혼합균주의 접종처리는 아로니아의 화학적 및 관능적 품질 특성 측면에서 떫은맛을 효과적으로 저감화시킬 수 있으며, APP의 경우 향후 식품산업에서 고품질의 유화제로서 활용 가능할 것으로 판단된다.

The aims of this study were (1) to investigate the effect of mixed inoculation on blocking the aronia astringency in respects of the chemical and sensory properties of aronia and (2) to verify the structural characteristics and possibilities on application as an emulsifier of aronia press cake pectin (APP). The mixed inoculation treatment was undertaken with the inoculation of mixed starter culture (Lactobacillus bulgaricus, Streptococcus thermophilus, and Saccharomyces cerevisiae), and then the four types of aronia concentrates were produced depending on the different ratio of mixed starter culture (NA, non-inoculated aronia concentrate; AI05, aronia concentrate inoculated with 0.00005% (w/w) mixed culture; AI10, aronia concentrate inoculated with 0.00010% (w/w) mixed culture; AI15, aronia concentrate inoculated with 0.00015% (w/w) mixed culture). The slightly decreased total soluble solid, pH, total phenolic content, total flavonoid content and total anthocyanin content were determined in mixed inoculated groups, and it seemed to be due to several possible by-products, such as acetaldehyde, organic acid and β-glycosidase, produced after the mixed inoculation. In particular, the astringency moderation after mixed inoculation was demonstrated in three intimately reciprocal tests: bovine serum albumin (BSA) precipitation assay, condensed tannin analysis and acetaldehyde measurement. Increased acetaldehyde components, ranging from 2.62 mg/mL to 2.90 mg/mL produced after mixed inoculation, could react with condensed tannin in aronia to form a new product, namly modified tannin, resulting in decreased condensed tannin contents from 30.04 mg CE/mL to 25.04 mg CE/mL. This trend was supported by the reduced aggregation of tannin-protein complex, directly responsible for astringency sensation in mouth, from 0.13 mg TAE/mL to 0.11 mg TAE/mL after mixed inoculation. Similarly, sensory analyses (consumer testing and ranking descriptive analysis) showed the moderated astringency perceptions after mixed inoculation. For consumer testing, taste scores of hedonic test with 50 consumers were assessed by six sensory attributes (overall liking, astringency, bitterness, sweetness, sourness and berry taste), and the scores of all sensory attributes in mixed inoculated samples were slightly higher than those of NA. The ranking descriptive analysis was evaluated by selected panelists received a common descriptive sensory training over several weeks, and there were four sensory attributes (astringency, bitterness, sweetness and sourness). The findings indicated the significantly decreased scores at astringency attribute after mixed inoculation. In secondly performed works of the present study, the structure of APP was characterized via chemical properties determinations [i.e. total sugar contents, galacturonic acid (GalA) contents, degree of esterification and monosaccharide composition], fourier transform infrared (FT-IR) spectroscopy, X-ray diffraction (XRD), 1D Nuclear magnetic resonance (NMR) spectroscopy (1H NMR and 13C NMR) and molecular weight distribution. It was indicated that APP had the pronounced quantity of protein (7.41%), relatively high amounts of total sugar (79.51%), GalA (57.26%), average molecular weight (i.e. Mw × 1010 = 1335 kDa), and the various neutral sugar contents (xylose, rhamnose and arabinose). For the results from degree of esterification value (63.38%) and FT-IR (1735-1750 cm-1 and 1600-1650 cm-1), APP was identified as one of the high methoxyl pectin, supporting by the observed specific peaks of XRD (at 2θ = 20.9 ˚) and NMR (at 5.05, 4.93 and 5.19 ppm in 1H NMR; at 98.05, 100.38 and 95.86 ppm in 13C NMR) analyses. Moreover, to examine the potential of APP as an emulsifier, three types of samples were prepared: (1) control; (2) emulsion (o/w) stabilized by 2% (w/w) of APP solution; (3) emulsion (o/w) stabilized by 2% (w/w) of PS solution. In detail, from the microscopy and particle size distribution measurements, it was revealed that APP more significantly decreased the droplet sizes (D[4,3] = 17.19 μm) in emulsion than PS (D[4,3] = 70.56 μm) and control (D[4,3] = 588.21 μm). Furthermore, APP showed significantly lower surface tension (from 68.05 mN/m to 56.66 mN/m) than that of PS (from 69.74 mN/m to 61.77 mN/m). Emulsion capacity and stability of APP (59.23% and 54.66%) were higher than those of PS (51.79% and 47.11%) at significant level (p < 0.05). The effective emulsifying properties of APP was likely to be encouraged by some factors depending on these mechanisms: (1) the large contents of protein in APP, considered as the major factor and observed as dense clusters by AFM, seemed to have the ability to strongly adsorb to oil and water interface owing to hydrophobicity; (2) the possible long and entangled polymer chains, sufficiently expected by molecular weight distribution of APP, may provide efficient steric stabilization in emulsion; (3) the great viscoelasticity of emulsion prepared by APP can form thick interfacial layers, thus restricting droplet movement and flocculation in emulsion. In summary, these results suggested that the mixed inoculation can be used to moderate astringency level of aronia in both chemical and sensory aspects, and further APP may become a highly promising pectin with effective emulsifying actions in various fields of food industry.

In secondly performed works of the present study, the structure of APP was characterized via chemical properties determinations [i.e. total sugar contents, galacturonic acid (GalA) contents, degree of esterification and monosaccharide composition], fourier transform infrared (FT-IR) spectroscopy, X-ray diffraction (XRD), 1D Nuclear magnetic resonance (NMR) spectroscopy (1H NMR and 13C NMR) and molecular weight distribution. It was indicated that APP had the pronounced quantity of protein (7.41%), relatively high amounts of total sugar (79.51%), GalA (57.26%), average molecular weight (i.e. Mw × 1010 = 1335 kDa), and the various neutral sugar contents (xylose, rhamnose and arabinose). For the results from degree of esterification value (63.38%) and FT-IR (1735-1750 cm-1 and 1600-1650 cm-1), APP was identified as one of the high methoxyl pectin, supporting by the observed specific peaks of XRD (at 2θ = 20.9 ˚) and NMR (at 5.05, 4.93 and 5.19 ppm in 1H NMR; at 98.05, 100.38 and 95.86 ppm in 13C NMR) analyses. Moreover, to examine the potential of APP as an emulsifier, three types of samples were prepared: (1) control; (2) emulsion (o/w) stabilized by 2% (w/w) of APP solution; (3) emulsion (o/w) stabilized by 2% (w/w) of PS solution. In detail, from the microscopy and particle size distribution measurements, it was revealed that APP more significantly decreased the droplet sizes (D[4,3] = 17.19 μm) in emulsion than PS (D[4,3] = 70.56 μm) and control (D[4,3] = 588.21 μm). Furthermore, APP showed significantly lower surface tension (from 68.05 mN/m to 56.66 mN/m) than that of PS (from 69.74 mN/m to 61.77 mN/m). Emulsion capacity and stability of APP (59.23% and 54.66%) were higher than those of PS (51.79% and 47.11%) at significant level (p < 0.05). The effective emulsifying properties of APP was likely to be encouraged by some factors depending on these mechanisms: (1) the large contents of protein in APP, considered as the major factor and observed as dense clusters by AFM, seemed to have the ability to strongly adsorb to oil and water interface owing to hydrophobicity; (2) the possible long and entangled polymer chains, sufficiently expected by molecular weight distribution of APP, may provide efficient steric stabilization in emulsion; (3) the great viscoelasticity of emulsion prepared by APP can form thick interfacial layers, thus restricting droplet movement and flocculation in emulsion. In summary, these results suggested that the mixed inoculation can be used to moderate astringency level of aronia in both chemical and sensory aspects, and further APP may become a highly promising pectin with effective emulsifying actions in various fields of food industry.
