The present study characterized the Gouda cheese made from pasteurized and raw milk during ripening period; chemical and physical properties, microbiological communities, peptide profiling and fatty acids composition. Two batches of Gouda cheese; pasteurized (P-GC) and raw milk cheese (R-GC). The parameters of protein and fat content in two cheese groups were determined to be 22.58-28.33% and 33.51-41.52%, in P-GC and R-GC, respectively, and these contents were increased compared to moisture content decreased throughout the ripening period. As the moisture content of cheese decreased during ripening, the level of texture properties such as harness, cohesiveness, gumminess and chewiness was significantly increased (p<0.05). Also, the levels of harness, cohesiveness, gumminess and chewiness in P-GC were higher than those of R-GC at the early ripening period. Two cheese groups showed increase in pH values depending on the levels of acetic acid, L-lactic acid and D-lactic acid were significantly decreased as ripening progressed (p<0.05). Major fatty acids of the groups were determined to be myristic acid (11.14-11.92%), palmitic acid (37.96-41.46%), stearic acid (13.92-15.24%), and these fatty acids ratio in R-GC were significantly higher than those of P-GC at the beginning period; the ratio of saturated fatty acid (SFA) was determined to 64.28-68.04% (R-GC) and 63.02-67.96% (P-GC) during ripening. However EPA, nervonic acid and DHA in the two cheese groups were determined to be at minor levels. Peptide profiles differed by manufacturing using milk with pasteurized or not. In particular, one peptide fragment is identified only in Gouda cheese produced from raw milk. The level of lactic acid bacteria (LAB) in R-GC was higher than those of P-GC, the two groups were significantly decreased during ripening period (p<0.05). The count of total coliforms was gradually decreased from 2.20 to 0.87 log CFU/g and 2.39 to 0.33 log CFU/g until 24 weeks of ripening, in P-GC and R-GC respectively. E. coli counts were identified from Gouda cheese made from raw milk at the beginning period, only. The counts of yeast and mold were determined from 3.51 to 4.87 log CFU/g from in P-GC and R-GC as ripening progressed. Specifically, the result of taxonomic classification of cheese bacteria at genus and species level revealed that Lactococcus and Leuconostoc were dominantly present at higher concentrations in all cheese samples. In the case of R-GC, interestingly, the proportion of coliform bacteria such as Escherichia fergusonii, Leclercia adecarboxylata, Raoultella ornithinolytica, Pseudomonas azotoformans and Pseudomonas geniculate, tended to decrease for ripening period. The canonical correlation analysis (CCA) results of microbial communities related to the layer part (Center and surface) of cheese showed no discernible trend during ripening. However, the microbial diversity of P-GC and R-GC has a significant difference as ripening period.
