In this experiment, we evaluated the moisturizing effect of liposomal serine, serine loaded solid lipid nanoparticles (S-SLN) and polysaccharide-rich reed root (Phragmites communis) extract (RRE) incorporated in hydrogel bases on human skin by using corneometer and skin surface hygrometer. The values of the area under the normalized conductance-time curve (AUCC) and the area under the moisture content-time curve (AUMCC) were developed and compared as a parameter for the water holding capacity of the skin. Hydrogels with liposomal serine (1%) exhibited 1.62∼1.77 times greater moisturizing effect than those of hydrogel and hydrogel with serine (1%). On the other hand, hydrogels with S-SLN at 5% and RRE at 0.25% exhibited greater hydration effect as compared to blank hydrogel and were estimated as (937.03 ± 438.58%) x h, (1277.29 ± 579.97%) x h, and (758.44 ± 165.39%) x h, respectively. Significantly enhanced moisturizing effect was observed with the hydrogel containing 0.25% RRE and 3% S-SLN, with AUCC increased 2.21 times compared to blank hydrogel. The results imply that effective delivery of serine into the skin is possible using lipid-based nanocarriers and RRE, which could be a promising strategy to moisturize the skin effectively. The penetration enhancing effect of two different classes of fatty acids: mono-unsaturated (Glycerol monooleate) and saturated (lauric acid and capric acid) through the SC barrier was investigated by FTIR spectroscopy using tape stripping method, which is a non-invasive technique to remove the SC layers. The lipids were applied to the skin by solubilizing in propylene glycol (PG) and after that, a piece of adhesive was used to remove the SC effectively for monitoring the molecular vibration of SC barrier during FTIR measurement. The results showed that GMO significantly changes the frequency shift from 2918 cm-1 to 2920.39 cm-1 and 2850.99 cm-1 to 2852.04 cm-1 during FTIR measurement compared to SC treated with lauric acid, capric acid, PG and untreated SC. This indicates that higher carbon chain length (C21) of GMO and more importantly, it contains unsaturated bond, which can easily disorganize the lipid chains of SC due to the central location of double bond conditions the formation of "Kink" or "Bend" within the barrier structure, thus shifting the frequencies at 2920 cm-1 and 2850cm-1 region of SC from lower wave number to higher wave number. We also demonstrated the moisturizing efficacy of hydrogels with GMO based S-SLN and RRE on human skin by using tape stripping method. The changes in skin moisture content were monitored by comparing the hydration values of tape stripped skin with that of before stripping of the skin after application of the formulations. The lowest AUCC values obtained by hydrogel with mixture of RRE (0.25%) and S-SLN (5%) of tape stripped skin indicating the synergistic moisturizing effect in the skin. Increased moisture content in the skin leads to increase the adhesion forces between the SC and adhesive tape, thus effectively removing the SC layer. Therefore, less hydration values are exhibited of the tape stripped skin. The results of this study signify the importance of SC for maintaining the natural moisture balance of the skin.
