The purpose of this study was to investigate the correlation between norovirus and hygienic indicator microorganisms by analyzing contamination status and contamination level of norovirus and hygienic indicator microorganisms in domestic agricultural products and environmental samples. On the other hand, The purpose of this study was to analyze the inactivation level of Norovirus substitute according to temperature and humidity conditions for consider about the proper use and storage method of food container for prevent Norovirus in real life. From July 2015 to July 2016, total 560 samples, which were 85 cases of agricultural products, 85 cases of soils, 85 cases of human feces, 85 cases of agricultural water and 85 cases of river water near the cultivation area and another 135 cases of river water samples were investigated pollution degree of norovirus and indicator microorganisms such like Coliform bacteria, Escherichia coli and Male Specific Coliphage (MSC). Through semi-nested PCR and sequencing, six types of GI and six types of GII-type norovirus were detected from 40 samples, two in agriculture water samples and 38 in river water samples, respectively. In some cases, both GI type and GII type were detected simultaneously in one sample. Quantitative analysis of positive samples using real-time RT-PCR showed approximately 104 copies per liter. 77.5% of positive samples were detected in January and February, but each one sample was detected in June and September, indicating that Norovirus is highly resistant to the environment. And approximately 40% of the positive samples were detected at less than 4℃. Coliform and Escherichia coli were quantitatively analyzed by MPN using an enzyme - based kit. In the case of coliform, most of the samples were contaminated. In the case of E. coli, about 85% of samples except river water samples showed low contamination level of less than 10 MPN. As a result of the correlation analysis between coliform and E. coli for the environmental samples collected with agricultural products, the degree of E. coli contamination in the river water had a significant effect on the coliform contamination of agricultural products and agricultural water. The male-specific coliphage, which is considered to be a hygienic indicator microorganism of the virus, was quantitatively analyzed by plaque assay by one layer method. As a result, only 15 samples were collected from river water samples taken separately and showed a contamination level ranging from 102 to 105 PFU/L. Also, it was detected in samples with high contamination levels of coliform and E. coli. As a result of the Pearson correlation analysis of the SPSS statistical program for analyzing the correlation between norovirus and hygienic indicator microorganisms, the coliform group was 0.290**(P = 0.007 <0.01) for the genotype G I in agricultural water and river water samples detected in agricultural water samples, and E. coli was 0.507**(P = 0.000 <0.01), and the coliform group, E. coli and MSC were 0.231**(P = 0.007 <0.01), 0.549**(P = 0.000 <0.01) and 0.884**(P = 0.000 <0.01), respectively. The degree of inactivation of MS2 bacteriophage, which is a norovirus surrogate for Stainless steel, Glass, Plastic, and Bronze Alloy Yugi, which is the surface material of food containers, is 40% and 75% The relative humidity of MS2 phage did not seem to affect the activity of MS2 phage. MS2 phage seems relatively stable at 4℃ due to the large rate of decrease at 25℃ than at 4℃. In addition, the degree of inactivation of MS2 phage on the Bronze Alloy Yugi was relatively higher than other materials according to the conditions. Therefore, the present study was conducted to detect norovirus from domestic agricultural and environmental samples and to analyze the correlation between norovirus and hygienic indicator microorganisms. As a result, it was found that many river water samples were contaminated with norovirus and correlated with the contamination level of hygienic indicator microorganisms. However, more samples need to be analyzed to obtain more reliability. In addition, it is important to control the temperature condition rather than humidity in order to prevent and control such Norovirus. In the case of handling large amounts of fish and shellfish such as oysters, using Bronze Alloy Yugi containers is consider that better than using other surface materials for control of norovirus.
