통, 오한 및 근육통을 유발한다. 유전학적, 면역학적으로 매우 다양한 바이러스라 현재 항바이러스제가 없는 상태이며 일반적인 대증요법으로 치료하고 있는 실정이다. 노로바이러스 식중독이 국내외에 지속적으로 발생함으로써 사회·경제적으로 큰 문제를 야기함으로써 보다 근원적인 해결을 위한 종합적이고 통합적인 예방연구 강화가 필요하다. 현재까지 진행된 유통식품, 식품용수(지하수 등)의 실태조사에서 1차 오염원으로 작용할 수 있는 농업용수, 해수, 농수산물에 대한 안전성 조사로의 전면 확대를 통해 전방위적인 식중독 예방 연구의 필요성이 제기된다

노로바이러스 식중독 예방의 근원적인 예방을 위해서는 크게 다음의 세가지 연구가 필요하다.
1. 농업용수, 해수, 농산물에서 노로바이러스 오염원 조사 및 분석
2. 노로바이러스 살균물질 탐색 및 재평가, 현장적용 검출 기술의 개발
3. 노로바이러스 감염성 평가 동물 적용모델의 개발
본 연구 사업단은 3중단위 7세부의 추진 체계를 갖추고 다음의 연구를 진행하였다.
- 1중단위
노로바이러스에 오염된 수자원 (지하수, 농업용수, 육상수, 해수)과 농산물에 대한 실태와 분석이 수행되지 않아 1차적 식중독 오염원으로 작용할 수 있는 요소에 대한 연구가 필요하며, 국내 농수산물 생산지 오염원을 조사하여 관리방안 마련을 위한 현황자료의 확보 또한 필요하다. 이를 위해 정착성수산물 생산지역 해수 및 육상 유입수, 주요 농산물 생산지의 농산물, 농업용수를 채취하여 노로바이러스 및 지표 미생물을 정성, 정량적으로 분석하였다.
정착성수산물 생산해역 (4개 해역)에서 노로바이러스 및 지표 미생물 오염을 모니터링하고 오염원 및 오염 경로를 분석한 결과 노로바이러스는 연구 해역에서 전반적으로 오염되어 있음을 확인하였고 (오염 지도 확보), 겨울철에 검출률이 높음을 확인하였다. 육상 유입수의 연구를 통한 오염원 추적을 통해 노로바이러스 오염은 육상 유입수 주변 주거/상업시설의 존재 및 규모에 의해 결정되는 것으로 추정되었다. 이와 더불어 생산지역의 하수처리장에서 노로바이러스 제거효율을 확인한 결과 하수처리장 마다 노로바이러스 제거 효율이 차이가 나는 것을 확인하였으며, 이는 처리공법의 차이로 추정할 수 있었다. 따라서 해수의 오염을 방지하기 위해서는 해수로 이동하는 육상 유입수 이동경로를 고려하여 노로바이러스 제거에 효율적인 정화시설 (하수처리장 등)이 필요할 것으로 여겨진다. 해수에서 노로바이러스 오염도는 대부분 기상학적, 수질학적 인자와 관련성이 없거나 매우 약하며, 기존의 지표 미생물 (대장균군, 파지 등)과도 관련성이 없는 것으로 파악되었다. 오직 15도 이하의 수온에서 노로바이러스 검출이 높아지는 것만이 확인되었다. 차세대 염기서열 분석을 통해 조사 해역 육상 유입수와 해수 모두에서 노로바이러스와 관련성이 있는 세균 마커를 찾아내었으며, 이 마커를 정량화하고 검출할 수 있는 real-time PCR법을 개발하였다.
국내 주요 농산물 생산지에서 농산물, 농업용수, 주위환경 (하천수, 토양, 분변 등)을 3차년도 동안 채취하였으며 노로바이러스 오염도 및 오염경로를 분석하였다. 하천수를 제외하고는 농산물, 농업용수, 주위환경 시료에서 노로바이러스 검출률은 낮았다 (2.3% 미만). 1~4월 (겨울~초여름)에 시료에서 노로바이러스가 대부분 검출되었으며, 특정 농작물에서 노로바이러스 검출률이 높았다.
농산물의 경우 노로바이러스 오염은 대장균균과 관련성이 있었으며 농업용수의 경우는 대장균/대장균군의 검출과 높은 관련성이 있었다. 월평균기온 약 20도 이하에서 노로바이러스가 검출되었으며, 관수법, 농업용수 종류, 농장형태, 화장실 형태와 대부분의 기상학적 요인은 노로바이러스 검출과 관련성이 없었다. 농산물의 노로바이러스가 오염된 경우 인체분변으로부터의 오염 추정사례가 가장 많았다.
노로바이러스 감염병 및 환경매체 검출에 영향을 미치는 잠재적 요인으로서 지형, 기후, 인문사회적 요인에 대한 전국단위의 공간정보를 구축하고, 과거 전국단위의 노로바이러스 감염병 발병 자료와 본 연구를 통해 얻어지는 환경매체 (농업용수, 하천수, 농산물, 인체분변) 검출자료를 공간정보화 함으로써 공간분석 기반 노로바이러스 오염원 추적 연구의 기반을 마련하였다. 이러한 공간정보를 토대로 공간 군집 분석을 수행함으로써 감염병이나 노로바이러스 검출률에 대한 공간적 분포를 파악하고 발생 빈도가 높은 hot spot지역을 분석하였으며 이 지역을 중심으로 미시분석을 실시하여 노로바이러스 검출에 미처리 생활하수와 인근 하수처리장의 유출수가 잠재적 오염원으로 파악되었다. 또한 본 연구단에서 실시한 모니터링 자료를 이용하여 노로바이러스 검출 지점에 대하여 공간 검색 통계 방법을 이용하여 환경 시료(농업용수, 농산물, 토양, 인체분변, 하천수)별 공간군집분석(공간검색통계)을 실시하였으며 군집이 형성된 지역을 중심으로 군집의 특징적 환경요인을 분석하였다.
수치적으로 계산된 환경요인을 검색 통계를 이용하여 전국적으로 적용하여 군집의 환경요인과 유사한 환경 조건은 갖는 지역들을 검색하여 노로바이러스 검출이 예상되는 지역을 선정하였고 과거 감염병 발병 지역과 비교분석을 실시한 결과 검출 예상 지역의 대부분이 과거 노로바이러스 감염병 발병 이력을 갖고 있음을 파악하였다. 통계적 기법에 기반하여 노로바이러스 검출 또는 감염병 발병에 영향력이 상대적으로 높은 환경요인을 추출한 결과 온도, 습도, 환경기초시설(마을하수도, 산업폐수처리장 등)이 설명변수로 선정되었으며 이를 이용하여 통계적 예측모델을 개발하였으며 아울러 일반 사용자가 노로바이러스 검출 확률 예측 모델을 ArcGIS상에서 손쉽게 구현할 수 있는 모듈을 개발하였다.
- 2중단위
노로바이러스 검지 키트는 유전자 변이가 심한 노로바이러스의 특징상 외국에서 검출된 바이러스를 기초로 한 연구결과 및 키트를 국내에 그대로 적용시킬 수 없으며, 낮은 민감도나 복잡한 과정을 통해 노로바이러스를 검출하므로 식품 현장에 적용하기가 어려운 실정이다. 또한 기존 미생물 저감화 기술 중 노로바이러스에 효율적이지 않은 기술이 많아 가장 효율적인 저감화 기술의 탐색이 필요하다. 이와 더불어 기존의 화학적 소독제, 기기적 소독장치를 통한 노로바이러스 저감화 외에 식품에 직접 사용가능한 자생 산약초 천연물, phytochemical 및 일상적 식품 소재를 대상으로 천연 노로바이러스 감염력 저해 및 노로바이러스 유전자 복제 저해 효능 물질을 발굴하고 실제 식품 적용가능성을 검증함으로써 항노로바이러스 식품 소재를 개발하는 것도 필요하다.
이를 위해 국내 현장에서 즉시 적용 가능한 키트 플랫폼을 개발하고 표준화된 검출법을 매뉴얼화함으로써 향후 노로바이러스 검출과 제어에 있어서 효과적인 방안들을 제시하고자 하며, 기존가열 저감화 기술 및 비가열 저감화 기술의 인간 노로바이러스 및 노로바이러스 surrogate에 대한 불활성 효과를 비교 검증하고자 한다. 또한 식품, 세척용수 및 기구에 오염된 노로바이러스를 저감화하기 위한 최적 처리 조건을 확립하고 이를 통해 저비용·고효율 현장이동형 노로바이러스 제어장비 및 오염원 차단을 위한 세척 용수 내 노로바이러스 제어 관리 시스템을 개발하고, 개발된 장비를 현장에 적용하여 효율성을 평가하고 이를 통한 현장용 표준 매뉴얼을 마련하고자 한다. 이와 더불어 천연물질을 이용한 다소비 식품에서 노로바이러스 제어 방법론을 수립하고 식품에 적용방안을 개발하고자 한다.
면역학적 방법을 이용한 노로바이러스 검지법을 개발하였다. 단시간 내로 높은 민감도를 가지고 노로바이러스를 검출할 수 있었으며, 사용법이 간단하여 식품현장 등지에서 일반인들도 사용 가능할 것으로 여겨진다. 또한 현장시료에 적용 가능한 유전학적 검출법 (RT-LAMP)를 개발하였으며, 이 검출법은 기존에 주로 사용되는 real-time PCR과 비슷한 민감도를 가지면서 보다 간편하게 노로바이러스를 검출할 수 있었다. 식품현장에서 노로바이러스를 회수할 수 있는 최적 스왑 샘플링법을 확립하였으며, 유전학적 검출법에서 증폭을 방해하는 저해물질을 제거할 수 있는 방법도 확립하였다.
기존 가열 및 비가열 저감화기술의 노로바이러스에 대한 불활성 능력 비교 측정하기 위해 인간 노로바이러스 및 노로바이러스 surrogate (뮤린 노로바이러스, feline calicivirus, MS2)를 활용하여 기존 가열 저감화 소독장치에 대한 노로바이러스 감염성 상실 효과를 측정하였고, 식품, 세척용수 및 기구 내 노로바이러스 저감화에 적용하여 효율성을 검증하였다. 염소계, 4급 암모늄계, 요오드계, 알코올계, 오존 살균소독제 등 상업용 살균소독제 종류 및 처리 조건별 인간 노로바이러스 및 노로바이러스 surrogate에 대한 불활성 효과도 비교 평가하였고, 주방용기, 식품 접촉 표면, 식품취급 작업자의 손, 식품, 수처리 등에 적용하여 노로바이러스 불활성화 효과 평가/소독 모델링 적용을 통한 저감화 소독방법의 노로바이러스 불활성화를 정량적으로 평가하였다. 각 저감화 기술의 식품 및 기구에 존재하는 노로바이러스 제어 효율을 높이기 위한 최적의 처리 조건을 확립하였고 저감화 기술 병행 처리를 통한 제어 효율에 대한 극대화를 연구하였다. 최종적으로 식품 산업 현장 및 급식소에 적용 가능한 저비용·고효율 이동형 노로바이러스 제어장비를 개발하고 오염원 차단을 위한 세척 용수 내 노로바이러스 제어 관리 시스템을 개발하였으며, 처리공정, 관리공정 등에 대한 현장 교육 매뉴얼 작성을 작성하고 홍보하였다.
신규 천연 노로바이러스 저해 효능 물질을 발굴하고, 동물실험을 통해 효능을 재확인하였다. 천연 항노로바이러스 물질의 식품 적용성을 검증하여 식품 소재로 개발하였다. 식품적용성 검증을 위해 다소비 식품 및 금속표면에서 노로바이러스 제어 효과를 확인하고 분무형 살균제 및 샐러드드레싱 시제품을 제작하였다.
- 3중단위
노로바이러스를 효과적으로 배양할 수 있는 세포배양 및 동물 모델 개발의 부재는 노로바이러스의 가장 중요한 장애물이다. 현재 노로바이러스 검사에 사용되고 있는 분자생물학적 유전자 증폭법은 노로바이러스의 존재 유무를 확인할 수 있으나, 생존성 여부를 판단하할 수 없는 문제점이 있다. 따라서 3중단위는 노로바이러스 예방을 위한 과학적 기반 강화 연구를 수행하였다. 특히 노로바이러스에 감수성을 나타내는 감염동물의 발굴은 노로바이러스 증식 기작을 규명하고, 환경저항성, 생존성 평가를 위한 도전적인 연구라고 할 수 있다.
3중단위 연구는 무균돼지, 미니돼지를 사용한 노로바이러스의 감염성 평가, human hFUT2-transgenic mice 모델 개발, 노로바이러스 생존성 평가를 위한 분자생물학적기술 개발로 수행되었다. 노로바이러스 감염량을 달리하여 공격접종한 무균돼지, 미니돼지에서 분변 및 조직내 바이러스 검출, 임상적 징후 등을 관찰하고, 병리조직학적병변과 면역조직화학법, 조직내교잡법 등을 활용하여 노로바이러스의 항원과 핵산의 분포를 조사하여 감염 표적세포를 확인하였다.
추가적으로 돼지 장 조직 explant model을 개발하여, 노로바이러스의 생존성을 상대적으로 용이하게 확인할 수 있는 검사법으로 제시하였다.
노로바이러스의 수용체로 알려진 human fucosyltransferase 2 (hFUT2)유전자를 삽입한 hFUT2-transgenic mice 중에서 백색계열인 Balb/c strain보다 흑색계열인 C57B6 strain에 삽입한 9880 line이 노로바이러스에 감염되는 것으로 확인할 수 있었다. 노로바이러스 최소감염량은 104 virions으로 확인되었다. 2중단위 연계 연구로 쥐노로바이러스를 사멸하는 자외선 조사조건으로 사람노로바이러스를 불활화 처리한 경우 9880 line transgenic mice에서 감염되지 않음을 확인하였다.
Ethidium monoazide (EMA) 또는 Propidium monoazide (PMA)를 적용한 분자생물학적 생존성 평가법을 개발하고, 쥐노로바이러스와 사람노로바이러스에 다양한 물리화학적 처리를 하고 플라크 측정법과 비교하였다. 가열처리, 에탄올, 자외선 조사 등 일부 조건에서는 EMA/PMA를 적용한 분자생물학적 검출법이 생존성 판별을 할 수 있었으나, 다양한 표면과 식품 적용에는 제한점이 많을 것으로 판단되었다.
3중단위에서 개발된 무균돼지, 미니돼지 모델은 노로바이러스 감염을 확인하는데 용이한 모델로 임파구, 대식구 등의 표적세포를 확인할 수 있었다. 또한 개발된 hFUT2-transgenic mice는 노로바이러스 생존성 평가에 활용할 수 있을 것으로 확인하였다. 후속연구를 통하여 노로바이러스를 배양할 수 있는 hFUT2-transgenic 세포주 개발에 기여할 수 있을 것이다.

Research objectives
