최근 환경 문제에 대한 사회적 우려가 증가함에 따라 친환경적인 섬유제품개발이 주목받고 있다. 천연 추출물인 티트리 오일은 일반적으로 호주에서 자라는 식물인 티트리(Melaleuca alternifolia)로부터 추출한 것으로, 항균 작용 및 여드름 염증 완화 기능을 가지는 것으로 알려져 있다. 에멀젼 전기방사(Emulsionelectrospinning)는 에멀젼을 사용하여 기존의 단일 노즐전기방사 장비로 비교적 용이하게 이중 구조의 나노섬유를 제조할 수 있다. 상기 방사법은 혼합되지 않는 친수성 물질과 소수성 물질을 전기방사할 수 있게 하므로 주로 바이오메디컬 분야에서 친수성 및 소수성 약물이 필요한 부위에 일정한 양으로 방출하게 하는 약물전달시스템을 개발되는 데에 활용된다. 본 연구에서는 에멀젼 전기방사를 활용하여 여드름 염증 완화 기능을 가진 티트리 오일을 함유한 core/sheath형 이중 구조 나노섬유를 제조하고 티트리 오일의 방출거동 및 항균성능을 평가하고자 한다. 에멀젼 용액을 제조하기 위하여 수상층의 PVA 10~14 wt%, 유상층의 티트리 오일 3.57~5 wt%, 계면활성제(Tween 80) 0.71~1 wt%로 구성된 다양한 에멀젼 용액을 전기방사하여 core/sheath형 이중 구조 복합나노섬유를 제조하기에 적합한 에멀젼 농도 및 최적의 전기방사 공정조건을 규명하였다. 티트리 오일과 PVA가 core/sheath형 이중 구조를 형성하는지 살펴보기 위하여 섬유 내부를 투과전자현미경(Transmission Electron Microscope)을 통해 관찰하였다. 티트리 오일을 형광염료인 Nile Red로 염색시켜 복합나노섬유 내부(core)에 봉입한 티트리 오일의 분포 양상을 공촛점현미경(Confocal Laser Scanning Microscope)을 통해 심층 분석하였다. 수용성 고분자 PVA와 티트리 오일을 함유하는 복합나노섬유의 내수성을 개선하기 위하여, 복합나노섬유의 열처리 조건을 규명함으로써 수분에 대한 안정성을 높이고자 하였다. 티트리 오일이 core/sheath형 복합섬유 내에 봉입된 상태에서 티트리 오일이 방출되는지 알아보고자, 열탈착식 가스크로마토질량분석기 (TDS-GC-MSD)를 통해 복합나노섬유에서 방출되는 티트리 오일의 방출거동을 분석하였다. 또한 티트리 오일을 함유한 복합나노섬유의 항균성능을 알아보고자 여드름 유발균으로 알려진 프로피오니박테륨 아크네(Propionibacterium acnes)와 2차 감염균인 황색포도상구균(Staphylococcus aureus)에 대하여 항균성능 평가를 실시하였다. 본 연구의 결과는 다음과 같다. 티트리 오일을 함유한 core/sheath형 복합나노섬유를 제조하기 위하여 도출된 에멀젼 농도는 PVA 14 wt%, 티트리 오일 5 wt%, 계면활성제(Tween 80) 1 wt%였다. 상기 에멀젼 농도에서 최적 방사 조건은 용액 공급 속도 0.8 ㎖/hr, 니들 게이지 23, 전압 25 kV, 방사거리 20 cm였다. 티트리 오일을 함유한 복합나노섬유의 형태를 주사전자현미경을 통해 관찰한 결과, 비드가 없고 직진성을 가진 나노섬유가 제조되었으며 섬유의 직경 범위는 약 155~295 nm였다. 투과전자현미경을 통해 섬유의 내부 구조를 분석한 결과, 섬유 내부에 심물질이 위치해 있는 이중 구조가 형성되었음이 관찰되었다. 공촛점현미경을 통해 내부에 봉입된 티트리 오일의 분포 양상 및 섬유 내부 구조를 심층 분석한 결과, 형광 물질 Nile Red(NR)로 염색된 NR-티트리 오일이 섬유 내부 심물질로 고르게 분포되었음을 확인하였다. 습윤 상태에서의 core/sheath형 복합나노섬유의 내수성을 개선하고자 열처리 조건을 규명한 결과, 170 ℃에서 1분간 열처리를 실시한 복합나노섬유의 경우 습윤상태에서 복합나노섬유의 형태 변화가 없어 수분 안정성이 높게 나타났다. 메디컬 나노텍스타일로의 최종 사용 용도를 고려하여 체온과 동일한 온도인 36 ℃에서 복합나노섬유에서 방출되는 티트리 오일의 방출거동을 분석한 결과, 건조 및 열처리 과정 후에도 티트리 오일의 구성 물질이 지속적으로 방출되는 것으로 나타났다. 또한 14일 간의 방출거동 평가를 통해 복합나노섬유 내에 위치한 티트리 오일의 구성성분 Terpinen 4-ol, γ-terpinene, Terpinolene이 14일까지 지속적으로 방출되는 것으로 나타났다. 복합나노섬유에 대해 항균성능 평가를 실시한 결과, 여드름 유발균인 프로피오니박테륨 아크네와 2차 감염균인 황색포도상구균에 대해 우수한 항균성능을 나타냈다. 이상의 연구 결과를 바탕으로 티트리 오일을 함유한 core/sheath형 복합나노섬유는 메디컬 나노텍스타일 및 항균 나노섬유제품으로 높은 활용 가능성이 있을 것으로 전망된다.

This research investigates incorporating tea tree oil into a polymeric matrix via emulsion electrospinning for the development of environmentally friendly, antimicrobial textile materials. Tea tree oil, a natural extract from the Australian native plant Melaleuca alternifolia (tea tree), is widely used due to several advantages, including its biosafety, environmental friendliness and antimicrobial activity. Recently, studies about the antimicrobial effects of tea tree oil on micro-organisms related to acne have been widely reported. Emulsion refers to a mixture of two or more liquids which are normally unblended, such as an oil phase and a water phase. Core/sheath structured nanofibers can easily be fabricated by means of emulsion electrospinning with a single nozzle (or needle) electrospinning set up, and no additional equipment is required as is needed when using coaxial electrospinning. Emulsion electrospinning is highly valued in biomedical areas, such as in drug delivery systems that provide controlled release of drugs. In this study, poly(vinyl alcohol) (PVA), known to be a biodegradable polymer and used as a medical material, was combined with tea tree oil to fabricate core/sheath structured nanofibers via an emulsion electrospinning technique. The main objectives of this study are to propose a method for the manufacture of core/sheath structured nanofibers containing tea tree oil and PVA via emulsion electrospinning, and to assess their release profile and antibacterial effects of the nanofibers containing tea tree oil for Propionibacterium acnes (P. acnes) and Staphylococcus aureus. An oil-in-water (O/W) type of emulsion composed of tea tree oil as the oil phase and aqueous PVA solution as the water phase was prepared. To determine appropriate concentrations to fabricate core/sheath structured nanofibers with a bead-free shape and a uniform diameter, the concentration ranges used were 3.57-5 wt% of tea tree oil, 10-14 wt% of PVA and 0.71-1 wt% of a surfactant (Tween 80). Adding the surfactant helped to stabilize the emulsion with the continued mixing and emulsification of the oil phase and the water phase. The morphology of nanofibers containing tea tree oil was observed by means of field emission scanning electron microscopy (FE-SEM), transmission electron microscopy (TEM), and confocal laser scanning microscopy (CLSM). To improve the stability of PVA-based nanocomposite fiber in water, heat treatment was used without any additional chemical method. The release profile of tea tree oil from the core of nanofibers was examined. Antibacterial effects of the nanofibers were assessed against P. acnes and Staphylococcus aureus. As a result, the optimum concentration to fabricate core/sheath structured nanofibers was 5 wt% tea tree oil, 14 wt% PVA, and 1% of a surfactant. The spinning conditions to fabricate core/sheath structured nanofibers were as follows: solution feed rate of 0.8 mL/hr, a needle gauge of 23, voltage of 25 kV and a tip-to-collector distance of 20 cm. The SEM image showed that bead-free, uniform nanofibers were fabricated via emulsion electrospinning. We observed a well-aligned core/sheath structure of the nanofibers containing tea tree oil in the core by means of TEM and CLSM. This confirms that core/sheath structured nanofibers were successfully fabricated via emulsion electrospinning. The optimum heat treatment condition to improve the aqueous stability of the nanocomposite fiber was 1 min at the temperature of 170 ℃. We investigated the release profile of tea tree oil emitted from the core/sheath structured nanofibers over 14 days and also examined the effect of drying and heat treatment on the release profile. Although the drying and heat treatment after electrospinning reduced the volatile organic compound(VOCs) emission, the release profile of Terpinen 4-ol emitted from the core/sheath structured nanofibers showed continuous emission over 1 to 14 days through TDS-GC-MSD analysis. Antibacterial activity of core/sheath structured nanofibers containing tea tree oil showed strong antibacterial effects against P. acne and a 99 % reduction rate against Staphylococcus aureus. In summary, we showed that an emulsion electrospinning technique can be used to fabricate core/sheath structured nanofibers containing tea tree oil without the need for an intermediate encapsulation step or an extra nozzle. We observed a well-aligned core/sheath structure of the nanofibers and identified the associated optimum concentrations and spinning conditions. The results showed that bead-free-shaped, uniform core/sheath structured nanofibers can be successfully fabricated via emulsion electrospinning. We conducted in vitro test of the antimicrobial activity of these nanofiber webs containing tea tree oil to explore the potential application to medical nanotextiles and healthcare textiles. Nanofibers containing Terpinen 4-ol, which has an antimicrobial effect on acne and helps to ease inflammation associated with acne, would expand the potential applications of medical nanotextiles and antimicrobial nanofibers. These materials may provide an opportunity to produce high-value-added nanotextile products and environmentally friendly items.
