To screen for genes regulating the root development of Arabidopsis, UAS activation tags were introduced into the background of Q2610 where GAL4 transcription factors are highly expressed leading to the random overexpression of genes neighboring to the tags. A dominant mutant developing short and twisted root was isolated and designated as defective root development 2-D (drd2-D). The T-DNA containing the UAS tag was localized at the 133 base-pair upstream of the putative start codon of At1g02030 (ZAT1) by using thermal asymmetric interlace-PCR. ZAT1 encodes a zinc finger (ZF) protein containing three C2H2 ZF motifs. The reverse transcription-PCR (RT-PCR) analysis revealed that the ZAT1 expression was upregulated in drd2-D. In addition, Q2610>>ZAT1 transgenic lines recapitulated the drd2-D phenotype. These results suggest that drd2-D was induced by ZAT1 overexpression. The expression of reporter genes driven by the ZAT1 promoter was observed in the tissues undergoing maturation such as border cell of root cap, endodermis, vasculature, and hydathodes. The border-cell specific expression pattern of ZAT1 was not altered in sombrero (smb) accumulating additional root cap layers and SMB-overexpressing mutant suggesting that ZAT1 expression is independent of SMB. WOX5 and CYCB1;1 expression was down-regulated in ZAT1-overexpression lines suggesting that ZAT1 compromises the identity of quiescent center and suppresses cell divisions. On the other hand, SMB and CEL5 expression pattern was not altered in ZAT1-overexpression background suggesting that ZAT1 could not change the identity of root cap tissues.
