Hypertension is one of the major risk factors for the development of cardiovascular diseases, including heart failure, stroke, atherosclerosis. The vascular endothelial cells are crucial in vein function and the formation of new vessels. Also vascular endothelial cell uses nitric oxide (NO) to signal the surrounding smooth muscle to relax, thus resulting in vasodilation and increased blood flow. Endothelial nitric oxide synthase, present in endothelial cells, catalyzes the production of NO from L-arginine and it's a activity is regulated by protein kinases and protein phosphatases. NO is considered to be an antihypertension drug as it causes vasodilation. Most cardiovascular diseases are associated with vascular dysfunction which is mostly a result of reduced NO production in endothelial cells. This study aimed to investigate the effects of Melanterite on the NO production in human umbilical vein endothelial cells. Melanterite increased NO production in HUVECs compared to the control group, but did not produce NO in RAW 264.7 macrophages. In conclusion, studies show effect of Melanterite on vascular dysfunction via increasing NO production in endothelial cells. These results suggest that Melanterite is a useful natural product and potential of a new hypertension treatment with vascular relaxation activity.
