The composition and content of phenolic compounds in soybean can be varied according to genetic and environmental factors, and also the various food processing such as fermentation, heating, pressuring, roasting, and drying can affect the composition of phenolic compounds in soy foods. Thus the content and composition of phenolic compounds in raw and roasted soybean flour during storage for 1 year at three different conditions of storage were analyzed by using LC-MS/MS in an attempt to figure out the effects of condition and duration of storage on the profile of phenolic compounds in soybean flour. Through the analysis of 56 selected phenolic compounds and free amino acids, 20 kinds of phenolic compounds were detected in the raw and roasted soybean flour. The content of total phenolic compounds was 301.59±9.48 µg/g in initial raw soybean flour and 257.47±14.42 µg/g in initial roasted soybean flour. Among phenolic compounds, 8 types of phenolic acid were found in the samples. Among the flavonoid groups, compounds that belong to isoflavone groups were only detected. After 12 weeks of storage, the content of total phenolic acids in raw soybean flour was significantly different depending on the storage temperatures. Also, after 24 weeks and 48 weeks of storage, the content of total phenolic acids in raw soybean flour was significantly higher in all three storage conditions than before. The changes in proportion of each isoflavone group depending on the storage condition and duration were measured. As the storage continued, the percentage of total malonyl glucosides over the sum of total isoflavones, accounting for 91.2 % of the sum of isoflavones, decreased in the raw soybean flour. However, the percentage of total glucosides increased following the storage duration. In roasted soybean flour, the percentage of total acetyl glucosides accounted for the highest proportion and that of total glucosides was the second at the initial. However, the percentage contribution of acetyl glucosides and glucosides to the total isoflavone contents was reversed after 2 weeks in all three storage conditions. The amount of phenolic acids and isoflavones in soybean flour increased as the storage temperature became higher and the storage duration became longer. This is thought as a consequence of the better release of phenolic compounds from soybean matrix or structural substances. This study showed that the composition and content of phenolic compounds in raw and roasted soybean flour were affected by the storage condition and duration, and the above results will be able to provide the basic information about the changes of beneficial phenolic compounds in soybean flour according to the storage condition and duration.
