PP300 투여군에서 고지방식이군보다 체중 및 지방량이 유의적으로 감소하였다. 간 내 염증인자, 지방생합성인자 및 콜레스테롤 대사 관련 mRNA 발현 정도를 본 결과 염증인자와 지방생합성인자는 정상식이군에 비해 고지방식이군에서 증가하였고, 고지방식이군에 비해 PP 투여군에서 감소하였으며, 콜레스테롤 대사 인자인 CYP7A1 및 담즙산 조절인자인 FXR은 증가하였고 간 내 지방생성 조절인자인 LXRα는 감소하였다. 3T3-L1 지방전구세포를 이용하여 OE 및 OF의 지방생성 억제효과를 관찰하였고, 지방생합성 관련 전사인자와 browning 관련 mRNA 발현을 통해 항비만 효과를 평가하였다. In this study, the anti-obesity effects of natural products containing flavonoids was evaluated separately. Anti-obesity effect of Pear pomace extract (Part 1), Onion extract (Part 2), and Quercetin (Part 3) was studied. In this study, effects of Pear pomace (PP) extract were first examined in 3T3-L1 preadipocytes followed by its effect in weight loss, fat reduction, glucose tolerance and lipid metabolism in high fat diet (HFD)-induced obese mice. Protein expression levels of adipogenesis related transcription factors were examined from 3T3-L1 cells. The lipid contents and adipogenesis related transcription factors expression levels were decreased in 3T3-L1 cells with PP. Then, effect of PP in HFD-induced obese mice was observed by orally administrating 150 mg/kg or 300 mg/kg PP in PBS once a day for 12 weeks. The body and fat weights of PP300 group were significantly lower than those of HFD group. Similarly, inflammatory factors and lipogenic factors were decreased in the PP-treated HFD group. PP significantly increased CYP7A1 in HFD-induced obese mice suggest that PP might reduce cholesterol. In order to investigate whether onion extract (OE) and onion fraction (OF) inhibit adipogenesis and browning related gene expression, we analyzed the expression of mRNA levels in 3T3-L1 cells. The mRNA levels of adipogenesis related transcription factors were significantly decreased in both OE and OF. mRNA levels of UCP1 were increased in OE treated mature adipocytes. To investigate the effects of quercetin (Q) the flavonoid rich onion on adipocyte differentiation, lipid contents and mRNA levels of adipogenesis related transcription factors were investigated in 3T3-L1 cells. Lipid accumulation was decreased in mature cells treated with Q. however mRNA expression levels of adipogenesis related transcription factors was not significantly decreased in Q treated cells. To compare the efficacy of the Q versus formulation qurecetin (FQ) on obesity improvement, we measured body weights in aqueous dispersion of quercetin or FQ-treated HFD mice. FQ-treated HFD group had significantly lower body weights compared with the HFD group, whereas the Q group did not differ from the HFD group. No significant change was observed in ALT and AST compared to normal chow, suggesting there was no FQ induced hepatotoxicity. In conclusion, PP and Q inhibits adipogenesis in adipocytes and showed anti-obesity effects by decreasing body and fat weights, and improving lipid metabolism in animal model. We suggest that flavonoid containing natural product may be a useful candidate for the treatment of obesity.
