TEM 분석으로 직경이 10~50nm인 대략 구형의 입자 형성을 확인할 수 있었다. 또한 은 나노입자가 로딩된 키토산/젤라틴 지지체의 항균 활성을 그람 양성균이 황색 포도상구균(S. aureus)을 이용하여 the disc-diffusion 방법으로 평가 하였다. 은 나노입자를 함유한 키토산/젤라틴 지지체의 항균활성은 은나노입자의 농도가 증가함에 따라 증가하였으며, 200 μg/Ml의 최고 농도에서 18mm의 최대 억제 영역을 나타냈다. 은나노 입자가 함유된 키토산/젤라틴 스캐폴더의 세포 독성을 분석하기 위하여, 마우스 유래인 NIH-3T3 Cell 로 MTT를 시행하였으며, 대조군 및 은 나노입자 미 함유 지지체를 비교한 결과 15일에 높은 생체적합성을 보였다. 그러므로 저비용-고효율적이고 환경친화적인 방법으로 생산된 은 나노입자는 생체 내 및 생체 외 응용분야에서 다양한 생체재료로 적용 할 수 있을 것이다.

Silver nanoparticles (Ag-NPs) have been successfully synthesized using a green approach via fruit extract of Lycium barbarum. The as-synthesized Ag-NPs were characterized by XRD, FT-IR, UV-VIS, SEM and TEM techniques. UV-VIS spectra of Ag-NPs formed at various reaction times showed an increase in the intensity of SPR band centered at 430 nm. The result of SEM clearly showed the Nano size of the silver particles, and EDS spectra showed the presence of silver element in the sample. XRD spectra revealed the face-cubic centered (fcc) crystal structure of silver nanocrystals, and the FT-IR spectrum revealed the presence of carbonyl, amide and hydroxyl groups from fruit extract, which bonded with silver nanoparticles for its stability. TEM analysis showed the formation of roughly spherical shaped particles with the diameter of 10-50 nm. Furthermore, the antibacterial activities of Ag-NPs-loaded chitosan/gelatin scaffolds were evaluated against Gram-positive bacterium Staphylococcus aureus by the disc-diffusion method. The antibacterial activity of Ag-NPs loaded chitosan/gelatin scaffold increased with an increase in the concentration of Ag-NPs, and produced a maximum zone of inhibition of 18 mm at the highest concentration of 200 μg/mL. The cytotoxicity of Ag-NPs-loaded chitosan/gelatin scaffolds were analyzed by MTT assay on mouse NIH 3T3 fibroblast cells, which showed high biocompatibility up to 15 days compared to the control, Ag-NPs unloaded polymeric scaffold. Thus, these Ag-NPs produced by a cost-effective and greener method could be used for various biomedical in vivo and in vitro applications.
