본 연구는 건조식품을 대상으로 다양한 온도 및 습도 조건에서 저장기간에 따른 미생물학적 변화를 살펴봄으로써 안전성을 보장하기 위한 보관기간 설정의 기초자료를 제공하고자 하였다. 외식업소에서 사용되는 4종의 건조식품(호박, 가지, 홍합, 김)을 선정하여 온도(25℃, 35℃)와 상대습도(50%,70% 및 90%)의 조건에서 2주간 저장된 시료에 대해 수분활성도 측정을 병행하며, 일반세균(aerobic plate counts), 대장균군(coliforms) 및 대장균(Escherichia coli), 효모 및 곰팡이(yeast & mold)의 정량적 수준을 측정하였으며 식중독균 접종실험을 통해 Bacillus cereus와 Listeria monocytogenes를 분석하였다. 시료에 따라 12-17 g, 김은 2.2 g을 채취하여 밀봉한 후 항온항습기에 미리 설정한 온도(25℃, 35℃)와 상대습도(50%, 70%, 90%) 조건으로 맞추어 15일간 보관하였다. 채취한 시료에 대해 하였으며, 식중독 균 접종실험을 통하여 Bacillus cereus와 Listeria monocytogenes의 성장 변화를 알아보았다. 각 조건에 보관된 시료는 1, 3, 5, 7, 9, 12, 15일 기간별로 멸균백에 각 시료 10 g(김은 2 g)에 멸균된 0.1% peptone water로 10단계 희석액을 가하여 로 2분간 고속으로 균질화 시킨 후 단계별 희석액을 만들어 사용하였다. 건조호박은 저장온도 25℃의 경우 상대습도 90%에서는 저장 1일부터 최고값을 나타내었으나 1 log CFU/g 미만의 증가를 보였으며 저장온도 35℃, 90%에서는 저장 1일째 1 log CFU/g 이상의 차이로 최고치를 나타내었다. 35℃ 조건에서는 상대습도가 높을수록 최고값을 나타내는 기간이 짧아졌다. 건가지에서도 건조호박과 유사한 경향을 나타내었으나 저장조건 35℃에서는 최고치를 보인 후 감소하며 다시 증가하는 패턴을 나타내었다. 이는 저장온도 25℃에 비해 35℃가 일반세균수 증식에 더 유리한 것으로 확인되었다. 건조홍합에서는 저장조건 25℃의 경우 상대습도 조건 모두에서 저장 9일째 최고값을 나타내었으며, 저장조건 35℃에서도 상대습도가 높을수록 최고값을 보이는 시점이 짧아졌다. 건조김의 경우 저장 1일부터 급격한 증가를 보였으며, 저장온도 25℃의 90%와 35℃의 70%, 90%에서는 1 log CFU/g 이상의 높은 값을 나타내었다. 또한, 저장온도 35℃에서는 상대습도가 높아질수록 저장 5일째, 3일째, 1일째로 각각 최고치를 보이는 기간이 짧아졌다. 저장기간 동안 수분활성도가 높은 건조식품은 저장 초기에 미생물 증식이 급격하게 증가되는 것을 보여주었다. 대장균군 및 대장균수는 건조호박의 경우 초기값 4.29 log CFU/g이던 것이 저장온도 25℃, 90%에서는 저장 3일부터 유의적으로 증가하여(p<0.001) 저장 7일째 1 log CFU/g의 높은 증가폭을 나타내었다. 저장온도 35℃에서는 세가지 상대습도에서 저장 1일부터 급격한 증가를 보이며 저장 5일째 1 log CFU/g 이상의 차이로 유의적으로 증가하였다(p<0.001). 건조가지의 경우 저장 1일부터 6가지 상대습도 모두에서 1 log CFU/g 이상의 높은 증가를 보이며 유의적으로 높은 값을 나타내었다(p<0.001). 대장균수에서도 일반세균수의 변화에서와 같이 저장온도와 상대습도가 증가할수록 최고치에 이르는 기간이 짧아지는 것으로 나타났다. 건조가지에서는 초기 대장균군수가 4개 실험 시료 중 가장 높게 검출되었으며, 대장균이 검출되었다(detection limit: <1.0 log CFU/g). 건조홍합과 건조김에서는 대장균군 및 대장균이 검출되지 않았다. 건조호박의 초기 곰팡이수는 2.94 log CFU/g으로 저장온도 25℃와 35℃의 모든 상대습도 조건에서는 지속적으로 증가하는 경향을 보였다(p<0.001). 25℃, 50%를 제외한 모든 조건에서 4 log CFU/g 이상의 값을 나타내었는데 그 값에 도달하는 기간이 온도와 상대습도가 높을수록 짧아지는 경향을 보였다. 건조가지의 경우도 초기 4.56 log CFU/g에서 지속적으로 증가하는 경향을 보였으나 저장온도 25℃에서는 1 log CFU/g 미만의 증가를 보였고 저장온도 35℃에서는 최고값이 1 log CFU/g 이상의 높은 증가를 보였다. 건조홍합의 초기 효모 및 곰팡이 수준은 1.94 log CFU/g으로 건조호박과 건조가지에 비해 낮게 검출이 되었으나 저장온도 35℃의 경우 저장 12일째부터 급격한 증가를 보여 1 log CFU/g 이상의 높은 차이를 나타내었다. 건조김의 경우 초기 1.8 log CFU/g에서 저장 1일째부터 높은 증가를 보였으며, 최고값에 이르는 기간이 상대습도가 높을수록 짧아졌다. 효모 및 곰팡이수는 낮은 수분활성도에서도 증식할 수 있는 특징이 나타났으며, 저장온도와 상대습도가 높을수록 효모 및 곰팡이수 증가에 유리한 것으로 나타났다. 건조농산물에 접종한 B. cereus 균은 저장기간 동안 뚜렷한 균수의 변화를 나타내지 않았지만 높은 온도와 상대습도에서는 최고 수준을 유지하는 기간이 더 길게 나타났다. 건조호박에서는 2.23 log CFU/g에서 4 log CFU/g의 수준으로 접종한 후 저장온도 25℃, 35℃의 상대습도 50%와 70%에서는 초기값에서 거의 변화 없이 일정한 수준을 유지하였으나, 90%에서는 1 log CFU/g 미만의 증가를 나타내었다. 건조수산물에 접종한 L. monocytogenes 균수의 변화는 초기 수분활성도가 높았던 건조홍합에서는 상대습도가 높을수록 큰 변화없이 일정한 수준을 유지하는 기간이 길었으며 감소하는 폭도 낮게 나타났다. 초기 수분활성도가 4개 실험 시료 중 가장 낮았던 건조김에서도 건조홍합에 비해 감소하는 폭이 크게 나타났지만, 건조수산물에서도 균수의 변화에서는 건조농산물에 접종한 것과 같이 저장온도와 상대습도가 높을수록 최고수준을 유지하는 기간이 길게 나타났다. 이상의 연구결과로 기온이 증가함에 따라 식중독균도 활성화 되는 것을 확인하였으며, 인위적 접종 실험결과에서도 일정기간 미생물의 증식과 위험을 확인할 수 있었다. 본 연구는 보관조건에 따라 저장기간별로 건조식품의 미생물학적 안전성을 평가한 연구로써, 건조식품에 대한 다양한 연구가 이루어진다면 보관관리 등 안전성을 위한 자료가 충분히 확보될 것이다. 따라서, 본 연구는 건조식품의 미생물학적 품질에 대한 정보를 제공한 연구로 우리나라에서 생산되고 판매되는 미생물의 품질을 평가하기 위한 추가 연구와 건조식품의 리스크 관리 정책 개발을 지원하기 위해 사용될 수 있을 것이다.

Summary
The purpose of this study was to provide basic data concerning the storage periods of dried foods, to ensure safety by examining the microbiological changes during storage various temperature and humidity conditions. Four kinds of dried foods (zucchini, eggplant, mussel, and laver) were selected and stored for 2 weeks, at 25℃ and 35℃ relative humidity (RH) of 50%, 70%, and 90%. The quantitative level of aerobic plate counts (APC) of coliforms, Escherichia coli, yeast, mold, Bacillus cereus and Listeria monocytogenes were analyzed. Twelve to seventeen grams of zucchini, eggplant, and mussel and 2.2 g of laver were collected, sealed, and then stored for 15 days at various temperature and RH conditions. Water activity was measured by a water activity meter. The growth of B. cereus on dried zucchini and dried eggplant and L. monocytogenes on dried mussel and dried laver were investigated by inoculation. Samples stored in each condition were sterilized at 1, 3, 5, 7, 9, 12, and 15 days. by adding a 10-fold dilution of sterile 0.1% peptone water to 10 g of each sample (or 2 g of laver). After homogenization at high speed for 2 minutes, a stepwise dilution was used. The APC of dried zucchini showed the highest value at the first day of storage at RH 90% and 25℃, but showed an increase of less than 1 log CFU/g. The highest above 1 log CFU/g of increase compared to the initial time was shown at the storage condition of RH 90%, 35℃. At 35℃, the higher the RH, the shorter the period to peak value. The microbial growth of dried eggplant was similar to that of dried zucchini but the maximum value at 35℃ and then decreased again till the 15th day of storage. It was confirmed that the storage temperature of 35℃ was more favorable for the growth of APC than 25℃. In dried mussel, the highest value of APC was obtained at the 9th day of storage at all RH values at, 25℃. When stored at 35℃, the higher the RH, the shorter the time until the peak was reached. Regarding the APC of dried laver, a rapid increase of above 1 log CFU/g was observed at RH 90%, 25℃, RH 70%, 25℃, and RH 90%, 35℃ at first day of storage. At the storage temperature of 35℃, the maximum period of APC decreased as the RH increased. For dried foods with higher water activity microbial growth rapidly increased at the early stage of storage. Coliforms counts of dried zucchini significantly increased to above 1 log CFU/g at the 7th day of storage at RH 90%, 25°C. At the storage temperature of 35℃, it increased rapidly on the 1st day at all three RH conditions and had 1 log CFU/g or more increase on the 5th day of storage (p<0.001). Coliforms counts of the dried eggplant were shown to increase by more than 1 log CFU/g in all RH conditions on the 1st day (p<0.001). In E. coli, the time to reach maximum growth was shortened as the storage temperature and RH increased, as in the case of the APC. In the dried eggplant, the initial counts of coliforms were the highest among the four test samples and the counts of E. coli were detected at less than 1.0 log CFU/g, which was the detection limit. However coliforms and E. coli were not detected in dried mussel and dried laver. As for yeast and mold, the initial count from dried zucchini was 2.94 log CFU/g, which increased continuously at 25℃ and 35℃ (p<0.001). At all conditions except RH 50%, 25℃, the counts were found to be more than 4 log CFU/g. The time to reach 4 log CFU/g tended to be shorter with higher temperature and RH. In the case of dried eggplant, the initial counts of 4.56 log CFU/g increased by less than 1 log CFU/g at 25℃, but higher than 1 log CFU/g at 35℃, respectively. The initial yeast and mold levels of dried mussel was 1.94 log CFU/g, which was lower than that of dried zucchini and dried eggplant, but the highest increase was observed at 35℃. The initial count of 1.8 log CFU/g on dried laver showed a high increase at the 1st day of storage and the time to reach the maximum value was shorter as the RH increased. Yeast and mold were able to proliferate even at lower levels of water activity. The higher storage temperature and RH were found to be more favorable in increasing the counts of yeast and mold. Bacillus cereus inoculated on dried produce did not show significant changes in counts during storage. However, the period of maximum growth at higher temperature and RH was longer than for other microbes. In dried zucchini, the B. cereus count was 2.2 log CFU/g to 4 log CFU/g, at RH 50% and RH 70% at 25℃ and 35℃, respectively, and showed an increase of less than 1 log CFU/g at RH 90%. Changes in the population of L. monocytogenes inoculated on the dried seafoods showed that the higher the RH, the longer the period of maximum growth. The maximum population was shown on the 9th day of storage in all RH conditions at 25℃ on dried mussel. In the case of dried seafood, the longer the storage temperature and RH are, the longer the period of maximum growth, as in the case of dried produce. Based on these results, it was confirmed that foodborne bacteria including B. cereus and L. monocytogenes were activated as the temperature increased. Research using a variety of pathogens and various kinds of dried foods will provide, sufficient data to devise approaches for safety in the storage management field. Therefore, this study provides information on the microbiological quality of dried foods and could be used to support further research on the microbiological quality of products and to develop the risk management policy of for dried foods in Korea.

Bacillus cereus inoculated on dried produce did not show significant changes in counts during storage. However, the period of maximum growth at higher temperature and RH was longer than for other microbes. In dried zucchini, the B. cereus count was 2.2 log CFU/g to 4 log CFU/g, at RH 50% and RH 70% at 25℃ and 35℃, respectively, and showed an increase of less than 1 log CFU/g at RH 90%. Changes in the population of L. monocytogenes inoculated on the dried seafoods showed that the higher the RH, the longer the period of maximum growth. The maximum population was shown on the 9th day of storage in all RH conditions at 25℃ on dried mussel. In the case of dried seafood, the longer the storage temperature and RH are, the longer the period of maximum growth, as in the case of dried produce. Based on these results, it was confirmed that foodborne bacteria including B. cereus and L. monocytogenes were activated as the temperature increased. Research using a variety of pathogens and various kinds of dried foods will provide, sufficient data to devise approaches for safety in the storage management field. Therefore, this study provides information on the microbiological quality of dried foods and could be used to support further research on the microbiological quality of products and to develop the risk management policy of for dried foods in Korea.
