Triptolide(TPL)는 식물의 생합성 천연물로 항염증작용과 면역억제작용, 항암작용 등의 다양한 생물학적 작용을 가진 것으로 알려져 있다. 그러나 TPL이 인간 호흡기 상피세포에서 중요 점액유전자들의 발현에 어떠한 영향을 미치는지는 아직 밝혀지지 않은 상태이다. 따라서 호흡기 상피세포에서 TPL이 점액유전자의 발현과 점액단백 생성에 어떠한 영향을 미치는지와 어떠한 경로를 통하여 점액유전자 발현이 되는지 알아보고자 본 연구를 시행하였다. 이 연구는 real-time polymerase chain reaction과 enzyme linked immunosorbent assay, western blot을 이용하여 인간 호흡기 상피세포에서 lipopolysaccharide(LPS)에 의해 유도된 MUC5AC와 MUC5B 유전자의 발현과 점액단백 생성에 대한 TPL의 효과, 그리고 유전자 발현과정에서의 nuclear factor kappa B(NF-kB)의 효과를 분석하였다. 사람 폐의 점액상피양 암 세포인 NCI-H292 세포와 인간 호흡기 상피세포 실험 모두에서 TPL이 NF-kB 경로를 억제 함으로서 LPS에 의해 유도된 MUC5AC와 MUC5B 유전자의 발현과 점액단백 생성을 유의하게 감소시켰다. 이 연구의 결과는 TPL이 인간 호흡기 상포세포에서 NF-kB 경로를 통해 MUC5AC와 MUC5B 유전자의 발현과 점액단백 생성을 억제시키는 효과가 있다는 것을 제시하였고, 이는 점액 과분비 억제를 통한 호흡기 ...

There are many anti-inflammatory substances were known such as triptolide(TPL), udenafil, betulinic acid, changkil saponin, glucosteroid. TPL is a diterpenoid compound from the thunder god vine, used in traditional Chinese medicine for treatment of immune inflammatory diseases including rheumatoid arthritis, systemic lupus erythematosus, nephritis and asthma. However, the effects of TPL on mucin secretion of human airway epithelial cells still have not been reported. Therefore, in this study, the effect of TPL on lipopolysaccharide(LPS)-induced MUC5AC and MUC5B expression were investigated in human airway epithelial cells. In the NCI-H292 cells and the primary cultures of human nasal epithelial cells, the effects of TPL on LPS-induced MUC5AC and MUC5B expression were investigated using real-time polymerase chain reaction, enzyme immunoassay and western blot. TPL significantly decreased the LPS-induced MUC5AC and MUC5B expression. And TPL also significantly decreased nuclear factor kappa B(NF-kB) phosphorylation. These results suggested that TPL down regulates MUC5AC and MUC5B expression via inhibition of NF-kB activation in human airway epithelial cells. Therefore, This study may provide important information about the biological role of triptolide on mucus-secreation in airway inflammatory disease and the development of the novel therapeutic agent for controling airway inflammatory disease.
