First, I elucidated the role of IL-4-induced innate CD8+ T cells in vivo, which are memory-like CD8+ T cells developed through IL-4 signaling in the thymus of CIITA transgenic (CIITATg) mice. When CIITATg mice were infected with chronic lymphocytic choriomeningitis virus (LCMV), the anti-viral CD8+ T-cell response was enhanced and viral control was accelerated compared to that in wild-type mice. IL-4-induced innate CD8+ T cells immediately accumulated after chronic viral infection and produced a robust amount of effector cytokines. Thus, IL-4-induced innate CD8+ T cells prevented persistent viral infections via effective viral control during the early phase of chronic viral infection. Furthermore, I investigated whether peripheral memory-like CD8+ T cells can control chronic diseases such as chronic viral infections and cancer, through the peripheral injection of IL-4 into wild-type mice. I used the IL-4/anti-IL-4 antibody complex (IL-4C) to prolong the in vivo effect of IL-4. Following IL-4C administration, memory-like CD8+ T cells were induced by the differentiation of naïve-origin CD8+ T cells and the expansion of memory phenotype-origin CD8+ T cells. Peripheral memory-like CD8+ T cells displayed a phenotype and function similar to that of IL-4-induced innate CD8+ T cells in the thymus of CIITATg mice. Similarly, IL-4C-treated mice also rapidly controlled chronic viruses and inhibited tumorigenesis. Inhibition of tumorigenesis in IL-4C-treated mice is also known to result from memory-like CD8+ T cells. These results demonstrate that IL-4 can induce memory-like CD8+ T cells in peripheral lymphoid organs, and that CD8+ T cells with this function can contribute to the treatment of chronic diseases by controlling antigen levels.
