Finally, the effects of positive and negative words on in vitro macrophages were tested molecular-biologically through Realtime PCR.

In the absence of the macrophages before the experiment, a researcher repeated positive and negative words to himself ten times in the same manner as in the actual intervention, and Emotiv was used to observe whether there was any tendency in the parts where the researcher's brain was activated. Then, three times a day at 4-hour intervals (9:30/13:30/17:30) for 3 days, the researcher spoke positive words or negative words directly to the two groups of the in vitro macrophages in a noiseless space in the same environment as a cell culture room. Each of the two groups was taken out of the incubator separately, and the positive and negative words were spoken to them 10 times for about 7 minutes, and the researcher tried to maintain the same speed and intensity of her voice each time. Twenty-four hours later, RNA was extracted and concentrated, and cDNA was synthesized, and Realtime PCR was performed using the four previously prepared gene primers (IL-12, TNF-α, IL-1β and iNOS). In the results of the Realtime PCR, positive words increased the expression levels of all tumor-immunity-related cytokines compared to GAPDH. On the other hand, the negative words reduced the expression levels of all tumor-immunity-related cytokines compared to GAPDH.
