However, there is a lack of precise studies and study results of various peptides in respecting of skin activating mechanism. Therefore, we produced 3D artificial skin to secure skin components such as skin cells and extracellular matrix by using tissue engineering technology and this 3D artificial skin was used to study for the skin wrinkle-improving effect of peptides. First, we made artificial skin dermis with controlled mechanical stiffness to have various young's modulus values from 0.7 kPa to 17 kPa by controlling the content of collagen and glutaraldehyde (GTA). After that, the skin cells were collected by using porous at inside the dermal layer introduced through freeze-drying and the growth and behavior of the cells were confirmed. when the GHK-copper peptide was treated, Skin cells were found to stimulate collagen synthesis according to the concentration of the applied GHK-copper peptide. In particular, the adenosine, which is known as a wrinkle-improving functional material, collagen synthesis rate is lowered in artificial skin dermis with a high stiffness of the mechnical dermis, whereas the GHK-copper peptide promotes collagen synthesis regardless of the stiffness of the dermis. Cell activity was influenced by the material properties of the extracellular matrix of dermis layer and the degree of application of the functional material. This result is expected to be utilized as an underlying technology in high functional cosmetic applications in the future.
