Flavourzyme과 Alcalase단백질 분해효소를 각각 단백질 g당 12%, 3% 처리하여 가수분해물을 제조하고, o-Phthaldialdehyde (OPA), SDS-PAGE, 크기배제 크로마토그래피(GPC)로 분해율 증가를 확인하였다. 효소 처리 후 생성된 가수분해물의 용해도와 유화능과 같은 기능적 특성은 대조군에 비해 증가하는 결과를 보였다. 가수분해물을 예비 역상 액체 크로마토크래피(Prep-LC)을 이용하여 4개의 분획물을 분리, 정제 한 후 ACE저해능을 분석 한 결과, IC50 = 18 µg/mL의 저해능을 보였다. 이 결과는 누에나방 번데기의 소수성펩타이드가 ACE 저해능의 근원인 것을 확인하였으며, 이러한 소수성 펩타이드는 향후 잠재적인 기능성식품이 될 수 있을 것이고 생각한다.

Silkworm pupae (Bombyx mori) has been eaten traditionally for centuries and recent reports display valuable properties associated with the insect including efficient angiotensin I-converting enzyme inhibition for blood pressure regulation. The objective of this study was to provide an overall investigation of silkworm pupae protein through determination of the most efficient methods for protein extraction, enzyme hydrolysis, physiochemical and physiological properties with partial purification of active fractions. Hexane defatting for 36 hours coupled with ultrasound extraction for 5 min resulted in a 74% protein yield. Flavourzyme and Alcalase proteases treated at 12% and 3% per gram protein respectively showed increased degrees of hydrolysis detected by o-Phthaldialdehyde (OPA), sodium-dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and gel-permeation chromatography (GPC). Functional properties of the hydrolysates including solubility and emulsifying properties showed increases compared to the control. The samples were separated through preparatory reverse-phase liquid chromatography (Prep RP-LC) into 4 different fractions and assayed for ACE inhibition, yielding an IC50 value of 18 µg/mL. The results suggest that our methods have successfully produced a hydrophobic peptide that may be the source of silkworm pupae’s ACE inhibitory activity for potential use as a future nutraceutical.
