인간의 산업 활동으로 인해 수계에 부영양화 현상이 심각해지고 있다. 부영양화 현상으로 질소와 인의 농도가 증가함에 따라 미세조류의 과도한 생장 및 증식에 의해 녹조 문제가 발생하면서 수생태계의 파괴, 호소에서의 심미적 불쾌감 형성, 인간에게 공급되는 물의 냄새와 맛에 영향을 미치는 등 녹조 문제에 따른 피해가 발생하고 있다. 이러한 문제로 인해 환경적인 측면에서 미세조류는 부정적인 요소로 인식되고 있다. 하지만 생명 공학 및 자원 순환 측면에서 미세조류는 다양한 이점을 가진 잠재력 높은 원료로 평가되고 있다. 예를 들어, 바이오매스, 화장품의 원료, 식품 분야뿐만 아니라 의학적인 사용에 이르기까지 다양한 방면에서 활용에 대한 연구가 활발히 진행 중에 있다. 따라서, 이와 같은 미세조류의 특성 및 부가적인 이점을 활용하여 폐수에서 Chlorella vulgaris를 배양하여 질소와 인을 저감하고 질소와 인을 소비하면서 증식하여 생산된 Chlorella vulgaris를 바이오연료 및 다양한 용도의 원료로써 공급하고자 하였다. 먼저, 미세조류를 통한 폐수처리의 가능성을 확인하기 위하여 인공폐수를 이용한 Chlorella vulgaris 배양에서 영양 염류의 변화를 확인하고 탁도를 이용하여 미세조류의 증식을 파악하고 비교 분석하였다. 총 질소의 경우 28일 동안 약 99%의 매우 높은 제거율을 보였고 총 인은 동일한 조건에서 92%의 우수한 제거율을 보였다. 또한 탁도를 통해 미세조류의 증식이 활발한 기간과 영양 염류의 제거율이 높은 기간이 비슷하게 관찰된 점을 미루어 보아 미세조류의 증식이 영양 염류의 제거율에 영향을 미치는 것을 확인하였다. 결론적으로 실험을 통해 적정 조건을 잘 파악한다면 미세조류의 배양을 통한 폐수의 영양 염류 제거가 가능할 것으로 확인되었다. 다음으로, 실제 폐수를 이용한 배양을 통해 Chlorella vulgaris의 생산이 가능한지 확인하기 위하여 쓰레기 매립지 침출수, 세차 폐수, 축산 폐수를 이용해 Chlorella vulgaris를 배양하고 생장 및 증식을 파악하기 위해 탁도를 측정하였다. 각 폐수에 대한 성상을 분석하고 그 데이터와 폐수에 따른 특성을 고려하여 각각 폐수 원액, 10배 희석, 100배 희석, 1000배 희석으로 다양한 조건에서 실폐수 배양 실험을 수행하였다. 침출수의 경우 10배 희석하여 배양을 진행한 배지에서 가장 높은 탁도를 보이면서 비교적 효율적인 배양이 가능할 것으로 나타났으나 유해물질이나 중금속 성분이 포함될 가능성이 높고 폐수의 성상 변화가 심한 침출수의 특성상 적응력이 우수하고 중금속 제거에 활용할 수 있는 미세조류 등 효율적인 배양을 위해서는 아직 연구가 필요할 것으로 생각된다. 세차 폐수는 희석 배수에 상관없이 모든 배지에서 Chlorella vulgaris의 배양이 이루어지지 않았다. 세차 폐수의 성상 분석에서 영양 염류 농도가 매우 낮은 것으로 보아 Chlorella vulgaris의 생장 및 증식에 필요한 영양 염류나 성분 농도가 극히 낮고 일찍 소진됨으로써 더 이상 생장 및 증식에 필요한 영양분이 존재하지 않아 생장 및 증식이 일어나지 않은 것으로 사료된다. 질산화조에서 채취한 축산 폐수는 10배 희석 > 100배 희석 > 1000배 희석 순으로 희석 배수에 따라 탁도의 증가율이 단계적으로 낮은 결과를 보였다. 과도한 영양 염류의 농도가 미세조류 배양에 영향을 미치는 주된 요인인 축산 폐수의 경우 희석 배수가 높아짐에 따라 영양 염류의 농도가 낮아지고 이러한 이유가 실험 결과의 원인으로 작용한 것으로 보였다. 10배 희석 배수에서 가장 긴 시간 동안 생존 및 증식을 함에 따라 본 실험 조건 중 10배 희석 배지가 Chlorella vulgaris 배양에 가장 적합한 것으로 보였다. 마지막으로 탈인조에서 채취한 축산 폐수 역시 10배 희석한 배지에서 가장 높은 탁도 증가율과 긴 생존 시간을 보였다. 결과적으로 모든 폐수에서 희석을 통해 어느 정도 미세조류 배양이 가능한 것으로 나타났고 10배 희석한 배지가 본 실험 조건 중 Chlorella vulgaris의 배양에 가장 적합한 것으로 확인되었다. 폐수의 더욱 자세한 성상 파악과 그에 따른 적절한 미세조류 종의 선정을 통해 폐수처리 및 미세조류 생산에 대한 효율성 개선이 가능할 것으로 생각된다. 다음으로 효율적인 미세조류 회수를 위한 응집 조건에 대한 실험을 수행하였다. 먼저 자가응집 조건에서 응집 첨가물로써 CaCl2, Ca(OH)2, MgCl2, Mg(OH)2를 이용하여 응집 실험을 진행하였다. 그 결과 Ca(OH)2를 첨가하였을 때 45.1%로 가장 높은 응집률을 보였다. 이때, 물리적 요인인 교반이 응집률에 얼마나 영향을 미치는지 알아보기 위해 Ca(OH)2를 첨가하여 다양한 교반 조건에 따른 응집 실험을 진행하였다. 교반으로 인하여 응집률에 큰 차이가 나타나지는 않았지만 근소한 차이로 교반 세기 60 rpm에서 68.4%로 가장 응집률이 높았고 교반 시간의 경우는 60분 조건에서 63%로 가장 높은 응집률을 보였다. 물리적 조건이 응집률에 있어서 결정적인 요인은 아니지만 응집률 향상에 영향을 미치는 것을 확인할 수 있었다. 다음으로 응집제인 PAC와 FeCl3를 사용하여 응집 실험을 진행한 결과, 각각 평균적으로 94.5%와 92.2%의 응집률로 매우 우수하고 안정적인 결과를 보였다. 추가적으로 효율적인 응집제의 사용을 위하여 최소한의 농도로 90% 이상의 응집률을 보이는 최적 조건을 도출하고자 PAC와 FeCl3에 대하여 농도 100 mg/L를 기준으로 10 mg/L까지 10 mg/L씩 단계별로 농도를 감소시키면서 응집 실험을 반복적으로 수행하였다. 그 결과 PAC의 경우 30 mg/L에서 최소한의 농도로 90%의 응집률을 넘는 결과를 보였고 FeCl3의 경우 40 mg/L에서 90% 이상의 응집률에 대한 최적의 농도로 확인되었다. 본 연구에서는 미세조류를 이용하여 폐수처리의 일환으로 질소, 인을 제거함과 동시에 폐수를 활용한 미세조류 생산을 통해 다양한 용도로 활용·연구되고 있는 미세조류에 대한 원료로서의 공급을 위하여 미세조류 배양을 통한 폐수의 영양 염류 저감에 대한 가능성과 실폐수를 이용한 미세조류 배양 및 생산의 가능성을 확인하였고 미세조류의 효율적인 회수에 필수적으로 고려되어야 하는 요인인 응집에 대한 다양한 실험을 진행하였다. 향후에 다양한 미세조류 종과 그에 따른 배양 조건에 대한 연구 및 더욱 자세한 성상 분석을 통한 적절한 폐수 선정 등 지속적인 연구가 이루어진다면 폐수 처리에 대한 효율성 측면과 자원으로써의 미세조류 활용을 통한 경제적 측면 개선이 될 수 있을 것으로 보인다.

However, microalgae are considered as a potential biomass resource in the biotechnology field and the resource circulation. For example, various researches are being carried out to apply microalgae in many fields from biofuel and cosmetics to medicine and medical supplies. Based on the microalgal characteristics and additional advantages, the microalgae can reduce the concentrations of nitrogen and phosphorus in the wastewater treatment process, and the grown algae can be supplied to the various fields as raw material. This study aims at investigating the applicability of microalgae to remove nutrients such as total nitrogen(T-N) and total phosphorus(T-P) in the various types of wastewater including artificial wastewater, landfill leachate, car-wash wastewater, and livestock wastewater. For this purpose, several experiments in this study were conducted to provide the possibility of wastewater as a culture medium for microalgae and to find the optimal sedimentation efficiency by the coagulation-flocculation (C-F) condition for the separation of microalgae from aquatic solution. For the experiments providing the possibility of wastewater as a culture medium of microalgae, Chlorella vulgaris was selected to be cultivated in the various types of wastewater for a 28 day duration for each of the 5 cultivations. The growth of Chlorella vulgaris was measured by turbidity. Also, this study determined the concentration changes of nutrients (total nitrogen, T-N, and total phosphorus, T-P) when Chlorella vulgaris grew in the various types of wastewater during the cultivating period. First of all, as the results of the artificial wastewater condition, T-N presents about 99% of removal efficiency for 28 days and T-P appears about 92% of removal efficiency under the same cultivation condition. According to the excellent removal values of nutrients, this study proved the applicability of microaglae to treat the nutrients in the artificial wastewater. Also, this study conducted experiments to examine the applicability of Chlorella vulgaris in real wastewater with three different types of wastewater including landfill leachate, car-wash wastewater, and livestock wastewater. The cultivation of Chlorella vulgaris within the 10 times diluted leachate only showed relatively effective results, but the leachate seemed to be an improper culture for Chlorella vulgaris since the leachate included various toxic compounds and heavy metals. Because the car-wash wastewater have very low concentrations of nutrients, all cultures from it resulted to be not proper to cultivate Chlorella vulgaris. The turbidity was increased at the initial stage of cultivation for a while, but the growth of Chlorella vulgaris was ceased due to the deficiency of nutrients. For the livestock wastewater, which was sampled from the nitrification tank, the culture medium were diluted 10 times, 100 times, and 1000 times, respectively. As the medium was less diluted, higher turbidity, longer life time, and more active growth were detected. Under the livestock wastewater collected from the dephosphorus tank, the culture medium was applied with the same dilution conditions of those sampled from the nitrification tank. The 10 times diluted medium presented the highest increasing ratio of turbidity and the longest life time. As the final outcome, Chlorella vulgaris showed possibility of cultivation in all wastewater, and especially, the most proper culture medium was the 10 times diluted medium. Therefore, if the proper type of microalgae may be selected after characteristics and composition of wastewater are defined well, this study provided the possibilities of higher wastewater treatment efficiency and more microalgae production, simultaneously. Secondly, the experiments were conducted to find the optimal sedimentation efficiency by the C-F condition to separate the microalgae from the aquatic solution. Under the auto-flocculation condition, the highest sedimentation efficiency appeared after the C-F process was conducted by adding Ca(OH)2. Additionally, the experiments were conducted to determine the change in flocculation due to physical conditions. As a result, the floccultaion rate was the highest in the 60 rpm of agitation speed and 60minute of agitation time. As experimental results under the condition using coagulants PAC and FeCl3, excellent and stable sedimentation efficiency was obtained. Also, for the effective use of coagulants, experiments were performed to draw the optimal amount of each coagulant. To deduct the optical condition for the sedimentation efficiency over 90% to a minimum concentration, the experiments were conducted with different concentrations from 100 mg L-1 to 10 mg L-1 with a decrement of 10 mg L-1 for the coagulants, PAC and FeCl3, repeatedly. The minimum concentration of PAC was 30 mg L-1 to reach over 90% of the sedimentation efficiency and the optimal concentration of FeCl3 was 40 mg L-1 for the sedimentation efficiency over 90%. In conclusion, this study confirmed the applicability of microalgae to treat the nutrients in the various types of wastewater. Additionally, the produced microalgae might be used as biomass in the resource circulation field. This study also provided the optimal physical and chemical conditions (the coagulant type and its optimal amount, and agitation speed and time, ect.) to reach over 90% of the sedimentation efficiency. As this study provides the possibility of wastewater treatment by the microalgae growth under the optimal condition for microalgae cultivation and separation, future studies are required to investigate various species of microalgae and their cultivation conditions according to the analyzed characteristics of proper wastewater. The continued research is helping to improve the wastewater treatment efficiency and to apply microalgae which can be made to be an additional economical profit.

Additionally, the experiments were conducted to determine the change in flocculation due to physical conditions. As a result, the floccultaion rate was the highest in the 60 rpm of agitation speed and 60minute of agitation time. As experimental results under the condition using coagulants PAC and FeCl3, excellent and stable sedimentation efficiency was obtained. Also, for the effective use of coagulants, experiments were performed to draw the optimal amount of each coagulant. To deduct the optical condition for the sedimentation efficiency over 90% to a minimum concentration, the experiments were conducted with different concentrations from 100 mg L-1 to 10 mg L-1 with a decrement of 10 mg L-1 for the coagulants, PAC and FeCl3, repeatedly. The minimum concentration of PAC was 30 mg L-1 to reach over 90% of the sedimentation efficiency and the optimal concentration of FeCl3 was 40 mg L-1 for the sedimentation efficiency over 90%. In conclusion, this study confirmed the applicability of microalgae to treat the nutrients in the various types of wastewater. Additionally, the produced microalgae might be used as biomass in the resource circulation field. This study also provided the optimal physical and chemical conditions (the coagulant type and its optimal amount, and agitation speed and time, ect.) to reach over 90% of the sedimentation efficiency. As this study provides the possibility of wastewater treatment by the microalgae growth under the optimal condition for microalgae cultivation and separation, future studies are required to investigate various species of microalgae and their cultivation conditions according to the analyzed characteristics of proper wastewater. The continued research is helping to improve the wastewater treatment efficiency and to apply microalgae which can be made to be an additional economical profit.
