본 연구에서는 glycosyltransferase 활성을 지니는 유산균을 이용하여 올리고당을 제조하고, 제조된 올리고당의 프리바이오틱스 및 면역조절 효과에 대하여 연구하였다. 또한 유산균주의 유전체를 분석하여 균주의 특성을 확인하고, 정제된 효소의 특성을 조사하였다. 국내 21개의 전통시장에서 구입한 123개의 발효식품 및 과채류 등에서 유산균을 분리하였고, glycosyltransferase 활성을 나타내는 유산균은 총 326주가 확인되었다. 그 중 효소 활성이 918.2 mU/mL로 가장 높고, sucrose와 maltose를 이용한 올리고당의 형성이 가장 좋은 균주인 CCK940을 본 연구의 올리고당 생합성 균주로 최종 선정하였다. 최종 선정된 CCK940은 배추김치에서 분리되었고, Leuconostoc lactis CCK940으로 동정 및 명명하였다. Leu. lactis CCK940을 이용한 올리고당의 최적 생산조건을 확립하기 위하여 pH, 공여체 분자와 수용체 분자의 비율 및 농도, 최적 수용체의 선별에 대하여 연구하였고, 반응표면분석법을 이용하여 올리고당의 최적 생산조건을 확립하였다. Leu. lactis CCK940을 이용한 올리고당의 최적 생산조건은 배지의 초기 pH 6, 배양 온도가 30℃, sucrose와 maltose의 농도가 각각 9.6, 7.4% (w/v)로 확인되었다. 최적 조건으로 생성된 올리고당은 Bio-gel P2를 이용하여 정제하였고, 정제된 올리고당은 glucose중합체로 이루어져 있었으며, NMR 분석결과 α–(1–6) 결합이 77.6%, α–(1–4) 결합이 22.4%를 차지하고 있었다. Leu. lactis CCK940가 생산하는 glycosyltransferase는 sepharose CL6B로 정제하여 조효소를 얻었다. 정제된 조효소의 분자량은 136 kDa으로 측정되었고, 최적 반응 온도와 pH는 30℃, pH 6.0이었고, Km, Vmax, kcat은 각각 9.1 mM, 1.4 U/min, 11.326 s–1으로 측정되었다. 정제한 올리고당은 L. casei, L. pentosus, L. plantarum, W. cibaria, B. animalis, S. cerevisiae의 성장을 유의적으로 증가시켰다. 올리고당을 대식세포에 처리한 결과 10 mg/mL의 농도에서도 독성을 나타내지 않았고, LPS를 처리한 것보다 높은 NO 생성능을 나타내었다. 면역증진 관련 사이토카인의 mRNA 발현량을 비교한 결과, TNF–α, IL–1β, IL–6가 농도의존적으로 증가하였고, iNOS의 발현량 또한 농도의존적으로 증가하였다. 올리고당은 P38, ERK, JNK 단백질의 발현량을 증가시켜 MAPK 신호전달경로에 관여하였다. 올리고당은 대식세포의 대식작용을 증진시켜 LPS로 유도한 대식세포와 유의적인 차이가 없는 것으로 확인되었다. Leu. lactis CCK940의 유전체는 2개의 contig로 구성되었고, 그 크기는 1,741,511 bp이며, G+C의 비율은 43.33%로 나타났다. 염색체 에는1,698개의 코딩 유전자, 12개의 rRNA 유전자, 68개의 tRNA 유전자가 확인되었다. Leu. lactis CCK940은 올리고당을 생산할 수 있는 glycosyltransferase, sucrose phosphorylase, maltose phosphorylase, β–galactosidase 등의 glycosyltransferase 생합성 유전자들을 지니고 있었다. 본 균주는 GenBank에 등록하여 accession number NZ_NQLF00000000를 부여받았다. 상기 결과를 통하여 선정된 Leu. lactis CCK940가 생산하는 glycosyltransferase는 올리고당을 생성하는 데 효과적임을 확인하였고, 생성된 올리고당은 프리바이오틱스 효과와 면역증진 효과를 지니고 있었다.

In this study, a lactic acid bacteria strain which producing glycosyltransferase was isolated from kimchi and the prebiotic and immunomodulatory effects of the gluco–oligosaccharides produced from this strain were evaluated. Out of the lactic acid bacteria isolated from 123 fermented foods, vegetables, and fruits purchased from 21 traditional markets in Korea, a total of 326 strains showed glycosyltransferase activity. Among these, CCK940 strain, which showed the highest enzyme activity (918.2 mU/mL) as well as the best production of oligosaccharides using sucrose and maltose as substrates, was selected for this study. The CCK940 strain was isolated from kimchi and identified as Leuconostoc lactis. To optimize the production of CCK–oligosaccharides using Leu. lactis CCK940, the initial pH of the culture broth, the suitable acceptor molecule, the ratio and the concentrations of donor and acceptor molecules were investigated. The optimum conditions for the CCK–oligosaccharides production using Leu. lactis CC940 determined by response surface methodology were as follows; initial pH of 6.0, incubation temperature of 30℃, and the concentrations of sucrose and maltose at 9.6% (w/v) and 7.4% (w/v), respectively. Under these conditions, the CCK–oligosaccharides were produced from Leu. lactis CCK940 using a fermenter, and the resulting CCK–oligosaccharides with a degree of polymerization greater than 4 were purified by Bio–gel P2 gel permeation column chromatography. The CCK–oligosaccharides were consisted of glucose only and their glycosidic linkages were composed of 77.6% of α–1,6 and 22.4% of α–1,4, which was determined by nuclear magnetic resonance. Glycosyltransferase produced from Leu. lactis CCK940 was partially purified using CL6B gel permeation column chromatography and the molecular weight of enzyme identified by SDS–polyacrylamide gel electrophoresis was 136 kDa. The optimum temperature and pH of the enzyme were determined to be 30℃ and pH 6.0. The Km, Vmax, and kcat values were 9.1 mM, 1.4 U/min, and 11.3 s–1, respectively. The CCK–oligosaccharides significantly promoted the growth of Lactobacillus casei, L. pentosus, L. plantarum, Weissella cibaria, Bifidobacterium animalis, and Saccharomyces cerevisiae. When macrophages were treated with the CCK–oligosaccharides at concentrations of 0.1–10 mg/mL, no cytotoxicity was observed. Nitric oxide production levels were similar to those observed after treatment with 1 μg/mL lipopolysaccharide. The mRNA expression levels of tumor necrosis factor–α, interleukin (IL)–1β, IL–6, as well as inducible nitric oxide synthase were increased in a dose–dependent manner by treatment with the CCK–oligosaccharides. Consistent with these results, the expression levels of mitogen–activated protein kinases such as P38, ERK, and JNK also increased in a dose–dependent manner after the treatment. When the genome sequence of Leu. lactis CCK940 was determined, the genome assembly revealed 2 contigs. The draft genome had 1,741,511 base pairs with a G+C content of 43.33%, containing 1,698 coding sequences, 12 rRNA genes, and 68 tRNA genes. Phylogenetic tree analysis revealed a high degree of homology between Leu. lactis CCK940 and Leu. lactis DSM 8581. The average nucleotide identity value between Leu. lactis CCK940 and Leu. lactis KFRI01 was 98.0%. Leu. lactis CCK940 contains several genes capable of synthesizing CCK–oligosaccharides such as the genes encoding glycosyltransferase (E.C. 2.4), β–galactosidase (EC 3.2.1.23), sucrose phosphorylase (EC 2.4.1.7), and maltose phosphorylase (EC 2.4.1.8), which belong to the GH1, GH1, GH13, and GH65 families, respectively. The sequence data of this strain was submitted in GenBank and its accession number is NZ_NQLF00000000. Based on this study, CCK–oligosaccharides produced from Leu. lactis CCK940 showed potential suitability as prebiotics and/or immunostimulant agents in food and pharmaceutical products.
