It is well known that side effects of anticancer drugs cause peripheral neuropathy. Here I confirmed that the effect of epothilone B on neurons depended on the types and developmental stages of neurons. I observed beneficial effects in the cortical neurons of the CNS but detected deleterious effects in mature neurons, which were prominent in the neurons of dorsal root ganglion (DRG) belonging to the PNS. By comparing results that microtubule disruption was induced differentially by treating microtubule destabilizing drugs, the distinct effect by neuronal type and age were shown to be based on microtubule stability. Next, I examined the effect of molecules, which were differentially regulated by PNS and CNS injury. I found that LRRK2 mRNA expression decreased after injury of the sciatic nerves, whereas the expression of LRRK2 mRNA and protein was increased when the CNS was damaged. In culture, regenerative ability was further increased when the expression level of LRRK2 was reduced by genetic manipulation. By contrast, axon regrowth was decreased by increasing LRRK2 expression. The results imply that LRRK2 might act as a regulator of nerve regeneration. This paper is expected to contribute to our understanding of the mechanism of nerve injury.
