C57BL / 6 마우스에 8 주 간 0.25 % UP 식이를 섭취시킴으로써 UP가 골격근에 미치는 영향에 대해서 조사했다. 결과적으로, UP 섭취 마우스의 총 주행거리 및 총 주행시간이 향상되었고 extensor digitorum longus (EDL) 및 gastrocnemius (Gastroc)근육의 질량이 증가했다. 근섬유 비대에 대한 UP의 효과를 알아보기 위해 laminin 염색을 시행하였고, UP 섭취 그룹의 Gastroc 근섬유 단면적 증가를 확인하였다. UP 섭취 그룹의 Gastroc 근육을 분석해본 결과 산화성 근섬유인 Myosin heavy chain (MHC) 1과 MHC2A의 단백질 발현이 증가되었고, 산화성 근섬유의 형성을 조절하는 Cytochrome c (Cyt C), Cytochrome c oxidase subunit 5a (Cox5a), Uncoupling Protein 3 (UCP3), Pyruvate Dehydrogenase Kinase 4 (PDK4) 및 Myoglobin의 mRNA 발현 또한 증가한 것을 확인하였다. 또한 UP 섭취 마우스의 근육에서 미토콘드리아의 크기가 증가하고 미토콘드리아 생합성에 관여하는 nuclear respiratory factor (NRF) 1 (NRF1), NRF2 및 mitochondrial transcription factor A (TFAM)의 mRNA 발현이 증가되었으며, oxidative phosphorylation (OXPHOS)의 단백질 및 mRNA 수준 또한 증가되었다. 혈관 신생에 대한 UP의 영향을 조사하기 위해 마우스 Gastroc 근육을 CD31 염색한 결과 UP 섭취 마우스에서 더 많은 혈관이 생성된 것을 확인하였고, 혈관 신생을 조절하는 Vascular Endothelial Growth Factor (VEGF) a, VEGFb, Fibroblast Growth Factor 1 (FGF1), Angiopoietin Like (Angptl) 1 , Angptl2의 mRNA 발현 또한 증가한 것을 확인하였다. 이러한 이유는 UP가 미토콘드리아의 기능과 산화성대사를 조절한다고 알려진 Peroxisome proliferator-activated receptor gamma co-activator 1-alpha (PGC-1α), AMP-activated protein kinase (AMPK), Sirtuin 1 (Sirt1), Estrogen-related receptor (ERR) α, ERRγ과 같은 유전자들을 활성화시켰기 때문으로 사료된다. 또한 C2C12 세포에 UP 와 그 주요 생체활성 성분인 fucoxanthin과 fucoidan을 처리한 결과, p-AMPK, PGC1α, Sirt1, ERRγ, ERRα 및 NRF2의 단백질 발현이 증가한 것을 확인했다. 따라서, UP는 산화 대사의 향상뿐만 아니라 골격근 질량과 지구력 향상에도 도움을 주는 기능성식품으로써의 가능성이 있다고 판단된다.

Skeletal muscle mass and function can deteriorate due to aging and pathological conditions including obesity and type 2 diabetes. In recent years, research has suggested that skeletal muscle performance can be improved by supplementation with plant-derived compounds. Recent studies have shown that Undaria pinnatifida (UP) is effective in regulating pathways associated with obesity and insulin resistance. However, the effect of UP on skeletal muscle metabolism and function has not previously been investigated. We sought to determine the effect of UP on muscle metabolism and exercise performance in C57BL/6 mice following dietary supplementation for 8 weeks. UP-treated mice showed increased running distance and total running time and increased extensor digitorum longus (EDL) and gastrocnemius muscle mass. Laminin staining confirmed that the cross-sectional area of gastrocnemius muscle fiber was increased by UP. The mRNA expression of and myocyte enhancer factor 2 (MEF2) isoforms as well as the protein expression of myosin heavy chain 1 (MHC1) and myosin heavy chain 2A (MHC2A) were also increased together with cytochrome c (Cyt C), cytochrome c oxidase subunit 5a (Cox5a), uncoupling Protein 3 (UCP3), pyruvate dehydrogenase kinase 4 (PDK4) and myoglobin in gastrocnemius muscles. Transmission electron microscopy revealed that UP-treated mice had larger mitochondria than the control mice. UP also increased protein and mRNA expression of OXPHOS as well as mRNA expression of NRF1, NRF2 and mitochondrial transcription factor A (TFAM). UP treatment also increased angiogenesis as well as mRNA expression of angiogenesis markers including VEGFa, VEGFb, FGF1, angiopoietin-like 1 (Angptl1) and angiopoietin-like 2 (Angptl2). In addition, UP-treated mice exhibited higher protein expression of peroxisome proliferator-activated receptor gamma co-activator 1-alpha (PGC-1α), phospho-AMPK), sirtuin 1 (Sirt1), estrogen-related receptor α (ERRα) and estrogen-related receptor γ (ERRγ). In C2C12 myotubes, the expression of p-AMPK, PGC1α, Sirt1, ERRγ, ERRα and NRF2 was increased by treatment with fucoidan and fucoxanthin, two major components of UP. Our findings suggest that UP may have applications in the maintenance and improvement of metabolism, as well as skeletal muscle mass and endurance.
