Tumor cells actively suppress immune responses and escape from the host immunosurveillance via multiple immuno-editing mechanisms, including downregulation of antigen presentation and the production of immune-regulatory molecules. To overcome such obstacles, various anticancer immunotherapies have been developed that eliminate the immunoediting mechanism or activating antitumor immune response. As part of this effort, immune checkpoint inhibitor (ICI), chimeric antigen receptor (CAR) T cell, and various tumor associated antigen (TAA)-based vaccines including peptide, virus or dendritic cell (DC)-based vaccines have been developed. These immunotherapies have attracted attention as promising anticancer therapy, however, these therapies have critical limitations that therapeutic efficacy varies depending on tumor types and more importantly by individual difference, due to the tumor microenvironment and heterogenicity of tumor cells. In situ vaccination (ISV) represents an alternative approach to the limitations of conventional immunotherapies. ISV promotes a powerful immune response by producing cancer vaccines through inducing immunogenic cell death (ICD) in the patient themselves without the need for ex vivo TAA identification and isolate processes. The notable feature of ISV is that it induces powerful immune response by using released whole TAAs and antigenic repertoire of each patient’s tumors. These properties allow to ISV enable personalized immunotherapy by overcoming variable therapeutic effects of conventional immunotherapies by individuals. In this study, various immuno-sensitizers that promote antitumor immune response by inducing tumor specific ICD, including 1) biocompatibility controllable polyethylene imine-based immuno-sensitizer and 2) cancer targetable PDT combined zinc-based immuno-sensitizer were developed, and their immunological activity and antitumor effect was demonstrated.
